A $3{\times}2$ factorial experiment was conducted to investigate the proper dietary protein and lipid levels for the growth of sub-adult flounder Paralichthys olivaceus reared during the summer season. Six experimental diets were formulated to contain three levels of protein (45%, 50% and 55%) and two levels of lipid (9% and 14%). Duplicate groups of fish (initial body weight of 298 g) were hand-fed to apparent satiation during the summer season ($21.8{\pm}1.7^{\circ}C$) for 60 days. Survival of each group was over 83% and there was not significant difference among all groups. Weight gain of fish fed the 45% protein diet with 14% lipid was not significantly different from that of fish fed the 50% and 55% protein diets with 9% and 14% lipids, but weight gain of fish fed the 45% protein diet with 9% lipid was significantly lower than that of fish fed the 55% protein diets with 14% lipid. Feed efficiency tended to increase with increasing dietary lipid level at each protein levels, although no significant differences were observed at 50% and 55% protein levels. Protein efficiency ratio, daily feed intake, condition factor and hepatosomatic index were not significantly affected by dietary protein and lipid levels. Crude lipid content of the liver tended to increase with increasing dietary lipid level at the same protein levels, but the opposite appearance was found for moisture content. The contents of moisture, crude protein and crude lipid of the dorsal muscle were not significantly affected by dietary protein and lipid levels. Based on data obtained form this study, inclusion of dietary protein at level of 45% appears sufficient to support optimal growth, and an increase of dietary lipid level from 9% to 14% has beneficial effects on feed utilization of sub-adult flounder during the summer season.
Preparative isolation of lipid granules from Fhodotorula glutinis, which has been studied for long time to produce edible lipids, was carried out by flotation method in Ficoll-Linear density gradient. When the isolated lipid granules were suspended in a series of solutions containing varying concentration of osmotic stabilizer (sorbitoal and mannitol) ranging from 0.8M to 0M, the lipid granules appeared to be disrupted at a concentration between 0.8M and 0.7, and again at a concentration below 0.1M, suggesting that lipid granules have a membraneous structure and that at least two types of lipid granules are present. Compositional analysis of lipids from lipid granules revealed that lipids are composed mainly of neutral lipids (87.8% of total lipids), predominantly as triacylglycerols (71.89%). Marked differences were observed inphospholipids between lipids of lipid granules and those of whole cells . The major components of phospholipids in lipid granules and inwhole cells are phosphatidylcholine(38.6%) and phosphatidylserine(42.8%), respectively. In addition, significant differences were also observed in the fatty acid composition of phospholipids. As phospholipids are important structural components of membranes, these differences lead to the suggesting that the membrane of lipid granules may be distinct functionally and structurally from other membranes of yeast cells. The major fatty acid components of neutral lipidss of whole cells and lipid granules are palmitic , oleic and linoleic acid. However , degreeof fatty acid unsaturation of neutal lipids of lipid granules was much lower than that of neutral lipids of whole cells.

A simple, reproducible , and accurate enzymatic method using a cholesterol assay kit was developed to quantify total cholesterol content in egg yolk. Total egg yolk lipid was extracted with hexane : isopropanol(3 : 2, v/v) mixture. Samples containing various amount of the total lipid(0-3mg) in optically identifical culture tubes were reacted for 10 min in a water bath (37$^{\circ}C$) with the enzyme solution (5ml) from the cholesterol assay kit. Cholesterol content of the reaction mixturesin culture tubes was spectrophotometrically determined by two different ways : (1) using the culture tube as a curvette(designate culture tube method ; CTM) and (2) the quartz cvette containing the reaction mixture transferred from the culture tube (designate standard cvette method, SCM). CTM revealed lower cholesterol content in 0.1-1.0mg lipid sample range that SCM did, but not significant. For more than 2.0mg lipid sample, CTM gave significantly (p<0.01) lower cholesterol content relative to that by SCM, suggesting that SCM give a false positive result from the sample containing more than 2 mg lipid due to the interference of absorbance by lipid dispersed in the reaction solution . Cholesterol content of less than 1.0mg lipid sample by CTM was proportional to the amount of lipid used, but its linear relationship was not seen in more than 2mg lipid sample. Thus, to determine the appropriate lipid amounts (mg) analyzed . A constant level (41$\mu\textrm{g}$/mg) of cholesterol concentration was observed from the sample containing 0.1-1mg lipid. after which the cholesterol level was dropped to less than 41$\mu\textrm{g}$ /mg. Cholesterol concentration in egg yolk samples quantified by CTM was in accordance with that by GC method. These results suggest that CTM is an useful method for the quantification of cholesterol in egg yolk lipid and other lipids as well.

To develop an easy and rapid method of quantifying lipid contents of marine dinoflagellates, we quantified lipid contents of common dinoflagellate species using a colorimetric method based on the sulpho-phospho-vanillin reaction. In this method, the optical density measured using a spectrophotometer was significantly positively correlated with the known lipid content of a standard oil (Canola oil). When using this method, the lipid content of each of the dinoflagellates Alexandrium minutum, Prorocentrum micans, P. minimum, and Lingulodinium polyedrum was also significantly positively correlated with the optical density and equivalent intensity of color. Thus, when comparing the color intensity or the optical density of a sample of a microalgal species with known color intensities or optical density, the lipid content of the target species could be rapidly quantified. Furthermore, the results of the sensitivity tests showed that only $1-3{\times}10^5cells$ of P. minimum and A. minutum, $10^4cells$ of P. micans, and $10^3cells$ of L. polyedrum (approximately 1-5 mL of dense cultures) were needed to determine the lipid content per cell. When the lipid content per cell of 9 dinoflagellates, a diatom, and a chlorophyte was analyzed using this method, the lipid content per cell of these microalgae, with the exception of the diatom, were significantly positively correlated with cell size, however, volume specific lipid content per cell was negatively correlated with cell size. Thus, this sulpho-phospho-vanillin method is an easy and rapid method of quantifying the lipid content of autotrophic, mixotrophic, and heterotrophic dinoflagellate species.
Infection with virulent or attenuated Salmonella typhimuriumhas known to induce reduction in proliferative responses of spleen cells. We investigated a role of lipid A from S. typhimurium, a B cell mitogen, on proliferation of spleen cells by T cell mitogens such as concanavaline A and phytohemagglutinin under in vitro and ex vivo conditions. Lipid A alone induced proliferation of spleen cells in vitroin a dose-dependent manner. However, subsequent treatment of concanavaline A or phytohemagglutin in after lipid A treatment induced proliferation suppression of murine spleen cells in vitro and ex vivo. Removal of macrophages from spleen cells, which were obtained from a lipid A-injected mouse, restored proliferation by concanavaline A and phytohemagglutinin, indicating that macrophages appeared to play a role in lipid A-induced suppression. Secreted molecules from macrophages did not accounted for the suppression because suppressive effect was not achieved when the supernatant from macrophage-containing spleen cell culture was conditoned to macrophage-depleted spleen cell culture. Co-culture of spleen cells from lipid A-treated and - untreated mice showed proliferation suppression as increasing cell numbers of lipid A-treated mouse. These data suggested that the cell-to-cell contact of macrophage with splenic lymphocyte cells is responsible for immune responses against lipid A, which is applicable to the case of human S. typhi infection.

The lipid compositions including fatty acid sterol lipid class and the lipid component of the total lipid extracted from the flesh of some fresh water fishes I. e. mandarin cornet cat fish and Korean perch were compared. The levels of total lipid of flesh tissues from the fresh water fishes were high(7.4%) in cat fish but low(1.4-2.2%) in mandarin fish Korean perch and cornet fish. On the contrary the content of unsaponifiable matters found in total lipid was low(2.6%) in cat fish but high(6.0%-6.5%) in mandarin fish Korean perch and cornet fish. Total lipids were mainly composed of triglyceride(74.6-86.5%) as major component in each sample and the other lipid components of total lipid e. g. polar lipid free fatty acids and free sterol were the minor components, The major fatty acids in total lipid of each sample were{{{{ {C }_{16 { }:_{ }0 } }}}}(19.6-29.2%) {{{{ {C }_{16 { }:_{ }1 } }}}}(17.3-30.7%) and {{{{ {C }_{18 { }:_{ }1 } }}}}(16.8-29.2%) and additionally it chiefly consisted of {{{{ {C }_{14 { }:_{ }0 } }}}} and {{{{ {C }_{18 { }:_{ }2 } }}}} Particularly the contents of polyenoic acids in total lipid of cat fish were higher than those of the other fish samples. The level of cholesterol in total lipid was low (8.3mg/g) in cat fish but were high(36.9-59.9mg/g) in mandarin fish Korean perch and cornet fish. The contents of fractionated neutral lipid(NL) were higher than those of polar lipid(PL) in each sample. Particularly phospholipid content in PL was low(6.0%) in cat fish but were high(23.1-36.3%) in mandarin fish Korean perch and cornet fish. Neutral lipids were mainly consisted of triglyceride(84.5-93.4%) as amjor component in each sample and the other lipid components of neutral lipid e, g, free fatty acids and free wterol were exhibited as a minor components, The fatty acid compositon of neutral lipids was very resembled to total lipids. The phospholipid in mandarin cornet and cat-fish were mainly composed of phosphatidyl serine(23.1-49.8%) and phosphatidyl choline(20.8-45.3%) The relatively higher amounts of phosphatidyl serine were observed in mandarine cornet and cat-fist than in Korean perch, But phosphatidyl ethanolamine(42.3%) and phosphatidyl choline (49.9%) were the main phospholipid in Korean perch. The extraordinary high content of phos-phatidyl ethanolamine compared to other fishes was characteristics in phospholipid composition of Korean perch. The major fatty acids in phospholipid of each sample were {{{{ {C }_{16 { }:_{ }0 } }}}} (38.3-46.5%) {{{{ {C}_{18 { }:_{ }0 } }}}}(14.2-21.7) and C16:1(11.6-13.8%) and additionally it chiefly consisted of C18:2, C18:0 and C17:0 The major fatty acids in glycolipid of each sample were C16:0(28.8-40.1%) C18:1(5.4-29.9%) C18:0(5.1-28.9%) and C16:1(8.2-20.1%) and additio-nally it chiefly consisted of C14:0 and C20:1.
This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. The experimentally cultivated potatoes of 4 varieties, Irish Cobbler, Warba, Shimabara, and Saco were dried in a frozen state, powdered subsequently and subjected to the extraction of free and bound lipids. Constituents of the prepared lipids were fractionated, quantified, and compared by the methods of column and gas-liquid chromatographies. The results were summarized as follows : 1. The total crude lipid content in potato on a dry weight basis was 0.57 % of which 0.2 % was free lipid and 0.37 % was bound lipid. 2. The neutral lipid content in the free lipid was 14.9 %, approximately 3 times as much as the 4.5 % contained in the bound lipid, whereas the glycolipid content in the free lipid was 15.1 %, slightly less than 22.2 % contained in the bound lipid. However, the phospholipid content was 33.9 % in the bound lipid, approximately 4.5 times as much as the 7.2 % contained in the free lipid. This fact revealed that the bound lipid consisted mainly of polar lipid, while the free lipid consisted of neutral lipids, glycolipids and phospholipids in about the same proportion. 3. The main fatty acids constituting more than 90 % in the free and bound lipids were linoleic, palmitic and linolenic acids. The content of the saturated fatty acid was slightly less in the free lipid than in the bound lipid, whereas the unsaturated fatty acids were more abundant in the free lipid.

In Med Dept. Daehan Education Insurance, Total 690 cases(male 376 cases, female 314 cases) were inspected to study the change of serum lipid value. The objects of investigation were total cholesterol, HDL cholesterol, triglyceride, phospholipid and total lipid value. The results are as follows: 1) In the normal subjects, with the relation of serum lipid value and the age. triglyceride, phospholipid and total lipid value showed the increasing tendency by 70 age, but total cholesterol value and HDL cholesterol value showed the increasing tendency by 59 age and the decreasing tendency after 59 age. 2) In the normal subject the difference of serum lipid value between sexes did not make the significant difference of level statistically in all items which can be inspected. 3) The weight change and serum lipid value are very significantly related. Total cholesterol, HDL cholesterol and total lipid value are high in female if the weight is light. But they are high in male the weight is heavy phospholipid value is opposite to the above result. Triglyceride value is high in male regardless the weight, but the significant difference happened in the high weight group. 4) As serum lipid value of hypertension patients is compared with that of the normal subjects, HDL cholesterol, triglyceride, phospholipid and total lipid value are significantly high. 5) The doubtful group of the fatty infittration of liver among the hypertension pafients have very significant difference in serum lipid value compared with not only the value of the normal subject but also that of the obesity group. 6) The doubtless group of the fatty infitration of liver among the hypertension patients have the significant difference in all serum lipid value compared with that of the normal subject, but compared with the obesity group males had the significant difference of HDL cholesterol value and both sexes had the significant difference in total lipid value, but had no significant difference in total cholesterol, triglyceride and phosprolipid value. 7) The ratio of total cholesterol per HDL cholesterol had the significant difference in the doubtful hypertension patients of the fatty infittration of liver and the relative weight, but it had no significant difference compared with the normal subject in the doubtless hypertension patients.

We ran a feeding trial to determine optimal dietary protein and lipid levels for growth of juvenile long snout bullhead Leiocassis longirostris Gunther. Eight experimental diets (P20L7, P20L14, P30L7, P30L14, P40L7, P40L14, P50L7 and P50L14) were formulated to contain 20%, 30%, 40% or 50% protein combined with either 7% or 14% lipid. Three replicate groups of fish (mean mass: 3.9 g/fish) were fed one of the experimental diets ad libitum for 8 weeks. Survival of fish fed the P20L14 diet was lower than that of fish fed the P40L14, P50L7 and P50L14 diets. Growth of fish fed diets containing 7% lipid increased with increasing protein level (up to 50% protein); growth of fish fed diets containing 14% lipid increased with increasing protein level (up to 30% protein). The feed efficiency of fish fed a diet with 50% protein and 7% lipid was higher than that of other groups. Whole body moisture and lipid contents were affected by dietary lipid level but not by dietary protein level. The crude lipid contents of fish fed 14% lipid diets were higher than those fed 7% lipid diets across all protein levels (other than the 50% level). Thus, under our experimental conditions, an increase in dietary protein level improved growth and feed efficiency of fish; a diet containing 50% protein with 7% lipid was optimal for growth and effective feed utilization in juvenile long snout bullhead.
