Studies on the Hemolytic Mechanism of Antitumor Platinum Complex (항암성 백금화합물의 용혈독성기전에 관한 연구)

  • This study was designed to determine the hemolytic mechanism of antitumor agent tetraphosphine platinum (II) complex (RC-1), which was synthesized recently. Erythrocytes treated with RC-1 showed concentration and time dependent lipid peroxydation, methemoglobin synthesis and hemolysis. And also treatment of radical scavengers showed the inhibitory effect of hemolysis and the decrease of malondialdehyde levels in RC-1 treated erythrocytes. So, the mechanism of hemolysis was considered to be the generation of free radicals, methemoglobin synthesis and the lipid peroxidation of phospholipid which composed of erythrocyte membrane.

Changes in Available Lysine and Lipid Peroxydation During Drying and Storage of Bioled Shrimp (자열(煮熱) 새우의 건조방법(乾燥方法) 및 저장중(貯藏中) 지질(脂質)의 산화(酸化)와 유효성(有效性) Lysine의 변화(變化))

  • Lipid oxidation is one of the major factors affecting on deterioration of nutritional quality in boiled and dired fish products. In this paper, the relationship between oxidized products of lipid, brown pigments, and available lysine during the drying and the storage of boiled and dried shrimp (Metapenaeus joynri) was investigated. Fresh shrimps were bioled in 5% sodium chloride solution. The boiled shrimps were treated in two ways, sun drying and hot air drying at $30{\pm}5^{\circ}C$. And the two dried products were stored at $30{\pm}5^{\circ}C$ for one month. The results obtained are as follows: TBA value increased up to 20 days and hereafter gradually diminished. POV was increased for processing and increased 15 days of storage. TBA value and POV increased rapidly while available lysine diminished during the sun drying and hot air drying. Brown pigment was increased during lipid oxidation but it was not statistically significant. This result implicits that the drying had greatly influenced on the oxidation of lipid and makes amino acids 'unavailable'. But there is no remarkable difference between the sun dried shrimps and the hot air dried shrimps so far as the lipid oxidation and available lysine.

  • In order to evaluate the anticancer activity of 1,4-naphthoquinone derivatives, several 1,4-naphthoquinone derivatives were newly synthesized and subjected to mouse leukemia p-388 cell line by MTT cytotoxicity assay. Among the several 1,4-naphtho-quinone derivatives, YC-001 has showed the most potent anticancer activity. To determine the safety of YC-001, hematotoxicity was tested. YC-001 induced hemolysis increased with both concentration and time dependent manner. The mechanism of hemolysis considered to be the generation of oxygen free radicals and lipid peroxydation of erythrocyte membrane which composed of phospholipids. Also methemoglobin, oxidized form of hemoglobin, was formed by YC-001.

Protective Effects of Angelica tenuissima Nakai on Hepatotoxicity by Carbon Tetrachloride in Rats (사염화탄소에 의한 간손상에 미치는 고본의 보호작용)

  • Hepatoprotective activity of methanol extract of Angelica tenuissima Nakai on the $CCl_4$-induced hepatotoxicity was investigated. To elucidate the hepatoprotective activity and free radical scavenging effect, we examined alanine aminotransferase (ALT), aspartate aminotransferase (AST), albumin, total protein, cholesterol, malondialdehyde (MDA) levels in serum and activities of superoxide dismutase (SOD), catalase (CAT) in hepatic tissue as compared with those of carbon tetrachloride-induced rats. The action mechanism also has been estimated by quantative analysis of cytochrome P450 (CYP), NADPH-CYP reductase for phase I metabolism and glutathion (GSH), glutathion S-transferase (GST) level for phase II metabolsim. Treatment of Angelica tenuissima methanol extract significantly lowered the levels of alanine aminotransferase and aspartate aminotransferase. In addition, the levels of cholesterol, triglyceride, MDA, CAT were decreased, and SOD was activated. This result indicates that the hepatoprotective effect of Angelica tenuissima methanol extract on the CCl4-induced hepatotoxicity would be originated from reduction of the NADPH-CYP reductase, GSH and the enhancement of the activities of GST, CYP.

Scavenging Property of Pyungwi-san Herbal-acupuncture Solution on ROS and RNS (평위산(平胃散) 약침액(藥鍼液)의 활성산소 및 활성질소 소거능)

  • Pyungwi-san(PWS) have been using as a basic prescription of digestive disorder in Korean traditional medicine. This study was performed to examine the in vitro antioxidant activity of the extract using different antioxidant tests including by 1,1-diphenyl-2-picryl-hydrazil (DPPH) radical scavenging, superoxide anion radical scavenging, metal chelating hydrogen peroxide scavenging, lipid peroxydation protective effect and scavenging effect of nitric oxide and peroxynitrite. Herbal-acupuncture solution of PWS(PWS-HS) exhibited a concentration-dependent inhibition of DPPH radical adduct formation and it showed dose-dependent free radical scavenging activity onto superoxide anions. In addition, the result of metal chelating hydrogen peroxide scavenging and ammonium thiocyanate experiments showed that PWS-HS was an active scavenger of hydroxyl radicals. Furthermore, it was also found to be effective in scavenging nitric oxide and peroxynitrite, well-known cytotoxic species that can oxidize several cellular components such as proteins, lipids and DNA.

Effects of the Antioxidants on the Diabetic Complications (당뇨병성 합병증에 대한 항산화제 효과)

  • These studies were performed to investigate the effect of antioxidants, melatonin (MLT) and ascorbic acid (AA), on the diabetic complications. In the experiment, twenty-four Sprague Dawley rats were divided into four groups; Group 1: no treatment (control) (n=6), Group 2: Streptozotocin (STZ; 65 mg/kg b.w. ip) treatment (n=6), Group 3: MLT (10 mg/kg b.w. ip) treatment (n=6), Group 4: ascorbic acid (AA: 40 mg/kg b.w.ip) treatment (n=6). Rats were sacrificed and collected designated samples (blood, kidney, pancreas) at the 21 days after antioxidant treatment (the 8th week after STZ injection). There was no significant change of body weight during the experiment period in the all groups. The concentration of blood glucose was decreased below 200 mg/dl in MLT. The concentration of HbA$_1$c level was decreased significantly in MLT (7.3$\pm$0.66%) compared with STZ (15.1$\pm$1.58%) (p<0.01). No significant change of SOD activity in the RBC and kidney was found in the all groups. In the pancreas, MLT (1.42$\pm$0.17 U/ml) and AA(1.65$\pm$0.16 U/ml) decreased SOD activity compared with STZ (4.10$\pm$0.27 U/ml). However, there was no significant difference among MLT and AA. The level of malondialdehyde(MDA) as an indicator of lipid peroxydation was decreased in MLT compared with STZ in the plasma, kidney and pancreas(p<0.01). These findings suggest that melatonin could prevent the complications of diabetes mellitus.

Attenuation of ROS Generation by KCNE1 Genes in Cisplatin-treated Auditory Cells

  • Potassium is essential for the proper functioning of the ears. The inner ear's endolymph differs from all other extracellular fluids (in its positive potential) and in the ionic compositions in the various parts of the endolymphatic space. Ion concentration of the endolymph is 150 mM of potassium, which is comparable to the concentrations in other organs. Cisplatin (cis-diamminedichloroplatinum II: CDDP) is one of the most effective anticancer drugs, widely used against various tumors. However, its clinical use is limited by the onset of severe side effects, including ototoxicity and nephrotoxicity. For ototoxicity, a number of evidences in cytotoxic mechanism of cisplatin, including perturbation of redox status, increase in lipid peroxydation, and formation of DNA adduct, have been suggested. Therefore, in this study, the author investigated the relationship between the potassium ions on cisplatin-induced cytotoxicity in HEI-OC1 cells associated with reactive oxygen species (ROS). KCNE1 gene expression by the concentration of intracellular potassium appeared in the plasma membrane and increased the concentration of intracellular potassium. Cisplatin decreased the viability of HEI-OC1 cells, but the KCNE1 gene increased. Also, the KCNE1 gene significantly suppressed generation of intracellular ROS by cisplatin. Western blot analysis showed that the KCNE1 gene increased phase II detoxification enzymes markers such as superoxide dismutase 1 (SOD1), superoxide dismutase (SOD2), NAD(P)H:quinine oxidoreductases (NQO1), which were associated with the scavenger of ROS. These results suggest that the KCNE1 gene for intracellular potassium concentration ultimately prevents ROS generation from cisplatin and further contributes to protect auditory sensory hair cells from ROS produced by cisplatin.

The Protective Effect of Epigallocatechin-3 Gallate on Ischemia/Reperfusion Injury in Isolated Rat Hearts: An ex vivo Approach

  • The aim of this study was to evaluate the preventive role of epigallocatechin-3 gallate (EGCG, a derivative of green tea) in ischemia/reperfusion (I/R) injury of isolated rat hearts. It has been suggested that EGCG has beneficial health effects, including prevention of cancer and heart disease, and it is also a potent antioxidant. Rat hearts were subjected to 20 min of normoxia, 20 min of zero-flow ischemia and then 50 min of reperfusion. EGCG was perfused 10 min before ischemia and during the whole reperfusion period. EGCG significantly increased left ventricular developed pressure (LVDP) and increased maximum positive and negative dP/dt (+/-dP/dtmax). EGCG also significantly increased the coronary flow (CF) at baseline before ischemia and at the onset of the reperfusion period. Moreover, EGCG decreased left ventricular end diastolic pressure (LVEDP). This study showed that lipid peroxydation was inhibited and Mn-SOD and catalase expressions were increased in the presence of EGCG. In addition, EGCG increased levels of Bcl-2, Mn-superoxide dismutase (SOD), and catalase expression and decreased levels of Bax and increased the ratio of Bcl-2/Bax in isolated rat hearts. Cleaved caspase-3 was decreased after EGCG treatment. EGCG markedly decreased the infarct size while attenuating the increase in lactate dehydrogenase (LDH) levels in the effluent. In summary, we suggest that EGCG has a protective effect on I/R-associated hemodynamic alteration and injury by acting as an antioxidant and anti-apoptotic agent in one.
