Effects of Lobetyolin, Lobetyol and Methyl linoleate on Secretion, Production and Gene Expression of MUC5AC Mucin from Airway Epithelial Cells

  • Background: In this study, we investigated whether lobetyolin, lobetyol, and methyl linoleate derived from Codonopsis pilosula affect MUC5AC mucin secretion, production, and gene expression from airway epithelial cells. Methods: Confluent NCI-H292 cells were pretreated with lobetyolin, lobetyol, or methyl linoleate for 30 minutes and then stimulated with phorbol 12-myristate 13-acetate (PMA) for 24 hours. The MUC5AC mucin gene expression, and mucin protein production and secretion were measured by reverse transcription polymerase chain reaction and enzyme-linked immunosorbent assay, respectively. Results: Lobetyolin, lobetyol, and methyl linoleate inhibited the gene expression of MUC5AC mucin induced by PMA; lobetyolin did not affect PMA-induced MUC5AC mucin production. However, lobetyol and methyl linoleate inhibited the production of MUC5AC mucin; lobetyolin and lobetyol did not significantly affect PMA-induced MUC5AC mucin secretion from NCI-H292 cells. However, methyl linoleate decreased the MUC5AC mucin secretion. Conclusion: These results suggest that among the three compounds, methyl linoleate can regulate gene expression, production, and secretion of MUC5AC mucin by directly acting on the airway epithelial cells.

Induction and Culture of High Polyacetylene-Yielding Hairy Roots in Ballon Flower (Pathycodon grandiflorum) (도라지(Platycodon grandirorum) 뿌리조직에서 고농도 함유 모상근의 유도 및 배양)

  • Hairy roots of Korean ballon flower (Platycodon grandiflorum A. DC) were induced from the root tissues infected with Agrobacterium rhizogenes ATCC 15834. Growth and polyacetylene [lobetyol (1), lobetyolin (2) and lobetyolinin (3)] production fo ten hairy root clones cultured in 1/4 Gamborg B5 (B5) liquid medium were determined. One selected hairy root clone (D6) grew well in hormone free-B5 liquid medium and showed maximum content of polyacetylenes at week 6 for 1 (0.375% dry wt) and at week 7 for 2 and 3 (3.030% and 0.206% dry wt, respectively) whose levels were much higher than those of the intact plant root (1:0.019%, 2:0.077% dry wt, 3 was not detected).

NF-${\kappa}B$ Activation by Compounds Found in Platycodon grandiflorum Extract

  • Compounds extracted from Platycodon grandiflorum were evaluated for an activation effect on nuclear factor-kappa B (NF-${\kappa}B$). In its active state, NF-${\kappa}B$ turns on the expression of genes related to cell proliferation or death. NF-${\kappa}B$ activators promote growth of neuron cells and can be used to control neurodegenerative diseases. The biological activity of P. grandiflorum extracts toward NF-${\kappa}B$ had not yet been studied. Although the biological activity of several compounds extracted from P. grandiflorum was evaluated, only three exhibited any significant activation effect on NF-${\kappa}B$.
