Macrophage Stimulating Activity of Exo-Biopolymer from Submerged Culture of Lentinus edodes with Rice Bran

  • To find a new utilization of rice bran, nine higher fungi were examined for the production of exo-biopolymer with macrophage stimulating activity from rice bran. Among the exo-biopolymers produced from submerged cultures, Lentinus edodes showed the highest activity, followed by Grifola frondosa, Schizophyllum commune, and Coriolus versicolor. L. edodes also had the most potent macrophage stimulating activity in a liquid culture rather than in a solid culture. In order to improve rice bran utilization and the yield of exo-biopolymer with macrophage stimulating activity, the treatment of Rapidase effectively increased the macrophage stimulating activity (about 30% increase), whereas the other enzymes (Econase, Viscozyme, Ultraflo, Celluclast, and Thermylase) treatments did not increase the macrophage stimulating activity. Exo-biopolymer with macrophage stimulating activity from L. edodes contained mainly neutral sugars (58.7%) with considerable amounts of uronic acid (32.2%) and a small amount of proteins (9.1%). Component sugars of exo-biopolymer consisted of mainly arabinose, galactose, glucose, mannose, and xylose (0.95:0.81:0.96:1.00:0.39, respectively). When the exo-biopolymer was treated with $NaIO_4, NaClO_2$, and pronase, the $NaClO_2$ treatment and pronase digestion had little effect, whereas $NaIO_4$ oxidation significantly decreased the macrophage stimulating activity (47.6% reduction at $100\mug/ml$). Therefore, the carbohydrate moiety in exo-biopolymer from L. edodes plays an important role in the expression of the macrophage stimulating activity.

마카 품종별 조다당 획분의 대식세포 활성 (Macrophage Stimulating Activity of Crude Polysaccharide on Maca (Lepidium meyenii) Varieties)

  • Maca roots (Lepidium meyenii) are an important medicinal herb that have long been used by the Andes-indigenous peoples and South Americans. In Korea, recently, it has attracted attention as a health food material because of nutritional values and physiological activities. The purpose of this study was to investigate the industrial applicability of maca (red and golden varieties; R&G) as immunostimulating materials. In the macrophage stimulating assay using RAW 264.7 cells at 125~500 ㎍/mL of non-cytotoxicity doses, G-HW showed the most potent production of TNF-α, IL-6 and nitric oxide compared to red maca or the other extracts. The general component analysis results showed that all extracts comprised more than 90% neutral sugars with small amounts of uronic acid and protein. Meanwhile, component sugar analysis showed the difference in the content of uronic acids of cold- and hot-water extract. Additionally, the further fractionation of G-HW into crude polysaccharide (G-CP) greatly enhanced the macrophage stimulating activity, and G-CP contained macromolecules over 144 kDa, comprised mainly of glucose and galacturonic acid (51.0 and 34.9%). In conclusion, crude polysaccharide from maca showed industrial applicability as immunostimulating material, and especially golden maca showed higher macrophage stimulating activity than red maca.

A Mouse Thymic Stromal Cell Line Producing Macrophage-Colony Stimulating Factor and Interleukin-6

  • A thymic stromal cell line, TFGD, was established from a thymic tumor mass developed spontaneously in p53 knock out mouse, and was found to produce cytokines that could induce bone marrow hematopoietic stem cells (HSCs) to differentiate into macrophages. The cytokines produced by the TFGD line were assessed by immunoassays. High level of macrophage-colony stimulating factor (M-CSF) and interleukin (IL)-6 was detected in the TFGD-culture supernatant, whereas granulocyte/macrophage-colony stimulating factor (GM-CSF), IL-3, IL-4, IL-5, IL-13, or interferon (IFN)-$\gamma$ was undetectable. Blocking experiments showed that anti-M-CSF monoclonal antibody could neutralize the differentiation-inducing activity shown by the TFGD-culture supernatant. Dot blot analysis of the total RNA isolated from the cultured fetal thymic stromal cells showed that M-CSF transcripts were expressed in the normal thymus. These observations, together with the earlier finding that M-CSF plus IL-6 is the optimal combination of cytokines for the induction of macrophage differentiation from HSCs in vitro, may indicate that thymic macrophages could be generated within the thymus by cytokines involving M-CSF.

  • Forty seven amylolytic Bifidobacterium strains were isolated on starch-containing agar medium from the faecal samples of the various age groups of Korean. From these amyloytic Bifidobacterium spp., two strains of KFRI 1535, identified temporarily as Bifidobacterium longum, and KFRI 1550, identified as Bifidobacterium breve, showed great macrophage-stimulating activity for the production of tumor necrosis factor-$\alpha$ and inteleukin-6. As the cell concentration increased the cytokine production increased, although in some strains the cytokine levels started to decline over cell concentration increased the cytokine production increased, although in some strains the cytokine levels started to decline over cell concentration of $250\mu\textrm{g}$/ml. the strains which showed high cytokine-stimulating activity generally showed greater production of nitric oxide even though differences were less between strains. Selected Bifidobacterium strains were compared for their fermentation capability in saccharified rice solution and in apple pomace mixture.

유산균에 의해 변화된 황기-포스트바이오틱스 다당류가 대식세포에 미치는 영향 (Effect of Astragalus membranaceus-postbiotics Polysaccharide Changed by Lactic Acid Bacteria on Macrophage)

  • To increase industrial applicability of Astragalus membranaceus (AM) as immunostimulating materials, hot-water extract (AME) was prepared from AM and fermented with Kimchi-lactic acid bacteria (Lactobacillus sakei & Leuconostoc mesenteroides) to prepare fermented AM-postbiotics (FAME). Although FAME prepared from AM-postbiotics did not show a significant enhancement in macrophage stimulating activity compared to non-fermented AME, crude polysaccharide (FAME-CP) fractionated by EtOH precipitation from FAME showed significantly higher macrophage stimulating activity than AME-CP. Compared to AME-CP, FAME-CP showed dramatic changes in component sugar and molecular weight distribution. FAME-CP was a polysaccharide with a major molecular weight distribution of 113.4 kDa containing Man (44.2%), Glc (19.3%), Gal (10.2%), GalA (10.2%), and Ara (7.4%) as sugar components. FAME-CP with enhanced macrophage stimulatory activity not only increased expression levels of mRNA genes encoding macrophage-activated factors (iNOS, TNF-α, MCP-1, IL-6, and COX-2), but also led the nuclear translocation of activated p65 and c-Jun. In conclusion, crude polysaccharide from AM-postbiotics fermented with lactic acid bacteria could increase industrial applicability as a functional material with enhanced immunostimulating activity than AME-CP.

A Recombinant Mous GM-CSF Protein Expressed as an Inclusion Form Shows Colony Stimulating Activity

  • Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a pleiotropic hematopoietic growth factor and an activator of lmature myeloid cells and recombinant GM-CSF is increasingly under clinical studies for the treatment of various diseases including cancer, infectious diseases and hematopoietic diseases. We constructed a reconbinant mouse GM-CSF expression plasmid with pelB leader sequence and His. Tag under T7 promoter control, and showed that the construct produced a 20 kDa recombinant protein in 8M urea. We also showed that the 20 kDa recombinant protein prepared in 8M urea sitmulated colony formation in vitro, indicating that the recombinant mGM-CSF can be renatured to its native form to show the colony stimulating activity.

홍화(Carthamus tinctorius L.)잎으로부터 분리한 다당류의 Macrophage 활성화에 대한 연구 (Study of Macrophage Stimulating Activity of the Polysaccharide Isolated from Leaves of Carthamus tinctorius L.)

  • 전통차 제조용 식물 63종을 대상으로 macrophage lysosomal enzyme activity를 검색한 결과, 홍화 냉수 추출물에서 높은 macrophage lysosomal enzyme 활성 (219%)을 발견하여 냉수 추출물 CT-0 획분에 대하여 metahnol 환류, ethanol 침전, 투석, 동결건조를 실시하여 macrophage lysosomal enzyme 활성이 더욱 증가된(227%) 고분자 획분 CT-1을 얻었다. 이 획분의 macrophage활성화 성분의 본체를 파악하기 위하여 pronate처리에 의한 단백질 분해와 periodate를 이용한 당 부위의 선택적 실활 후 활성을 검토한 결과, pronase를 처리한 CT-1에서는 약 8% 정도의 활성 증가를 보인반면 periodate 산화물에서는 활성이 약 8% 정도 감소되는 것으로 보아 홍화로부터 macrophage 활성을 나타내는 냉수 추출물의 활성 본체는 다당임을 알 수 있었다. 조다당 활성획분에 대하여 anion exchange column chromatography를 실시하여 9개의 획분(CT-1-I~CT-1-VIII)을 얻었으며 수율과 활성이 가장 높은 CT-1-IIa 획분을 Sepharose CL-6B 및 Sephacrl S-200의 gel permeation chromatography를 수행하여 주요 활성 다당인 CT-1-IIa-2-1을 최종적으로 정제하였다. HPLC상에서 순수한 단일 peak로 확인된 CT-1-IIa-2-1은 분자량이 68 kDa정도의 다당인 것으로 나타났고 macrophage의 lysosomal enzyme 활성은 대조군을 100%로 비교했을 때 243%를 나타내었다. 또한 구성당의 조성은 xylose(27.4%), arabinose(16.1%), mannose(15.9%), glucose(14.5%)의 순이었다. 본 연구에서 CT-1-IIa-2-1은 macrophage의 면역활성을 증가시키는 물질임이 확인되었으나 mouse를 대상으로 급성독성 검사를 실시한 결과 LD$_{50}$값이 397mg/kg으로 일정 농도 이상의 고농도에서는 독성을 나타냈다. 따라서 본 시료에 대하여 아만성.만성 독성과 유전 및 면역 독성과 같은 구체적인 독성 검사를 실시하여 안전농도를 산출한다면, 면역증강물질로의 개발이 가능할 것으로 사료된다.다.

김치 유산균 발효 혼합생약재 추출물의 강화된 대식세포 활성 (Enhanced Macrophage Stimulatory Activities of Extracts from Kimchi Lactic Acid Bacteria-Fermented Mixed Herbal Medicines)

  • To enhance the bioavailability and bioactivities of mixed herbal medicines (RW), they were fermented with lactic-acid bacteria isolated from kimchi into postbiotics (FRW). Then, from the results of the 16s rRNA sequencing analysis, lactic acid bacteria isolated from kimchi were identified to be of two species, namely Lactobacillus sakei and Leuconostoc mesenteroides. The FRW prepared from the RW were extracted using hot water (HW) and 70% EtOH (EtOH) for comparison of their macrophage-stimulating activities. Based on a comparison of the activities of the FRW extracts, nitric oxide (NO) production of HW was significantly higher than that in EtOH. An analysis of the chemical properties of the extracts showed that HW had higher contents of neutral sugar and uronic acid than EtOH as well as contained a large amount of glucose. In addition, crude polysaccharide (CP) was prepared to enhance the macrophage-stimulating activity. The FRW-CP not only secreted immunostimulatory mediators but also increased the expression of immunostimulatory genes (iNOS, TNF-α, MCP-1, and IL-6). The fractionated FRW-CP contained about 90% neutral sugars, and these sugars were mainly composed of glucose, galacturonic acid, and arabinose. Thus, FRW prepared by fermentation of RW with kimchi lactic acid bacteria were found to be immunostimulatory modulators.

Changes in Physiochemical Properties during the Fermentation of Doenjang Prepared with Black Soybeans

  • A physicochemical assessment of Doenjang (traditional fermented soyfood) prepared with Korean black soybeans (Glycine max) was carried out. The T-N rate increased slowly during storage up to 120 days and the A-N rate increased up to 80 days of ripening and then decreased slightly. The caseinolytic activity increased slowly during storage up to 80 days and then decreased after 80 days. In addition, the fibrinolytic and ${\beta}-glucosidase$ activities increased up to 80 and 30 days and then decreased. Genistin and daidzin concentrations gradually decreased with increased fermentation time. However, genistein and daidzein slowly increased with fermentation time. Genistein and daidzein reached maximum concentrations (316.8 and $305.2{\mu}g/g$, respectively) and plateaued thereafter. The anthocyanins increased greatly during fermentation up to 50 days and then remained constant between 50 and 90 days. Polyphenol contents showed a slight increase up to 80 days and then slowly decreased. The DPPH and ABTS radical scavenging activities increased linearly during storage up to 50 days, reached about 28.9% and 2.17 mg/g, respectively, and then slowly decreased. At 20 days of fermentation, macrophage-stimulating activity of the extract showed a maximum activity.

Granulocyte Macrophage Colony Stimulating Factor에 의한 생쥐 초기 배아 발생의 신호전달 (Granulocyte Macrophage-Colony Stimulating Factor Signaling in Development of Mouse Embryos)

  • Objective: Present study was aimed to verify the effect of granulocyte macrophage-colony stimulating factor (GM-CSF) in the preimplantation development of mouse embryos and the involvement of the mitogen activated protein kiase (MAPK) in the GM-CSF signaling. Methods: Two-cell embryos were cultured for 96 h in the presence or absence of GM-CSF (0, 0.4, 2, 10 ng/ml) and PD98059, a MEK inhibitor (10 ${\mu}M$). Morphological development, cell number per blastocyst, and apoptotic nuclei, were eamined. MAPK activity of embryonic immunoprecipitate by MAPK (ERK1/2) antibody was measured by in vitro phosphorylation of myelin basic protein. Results: At post hCG 122 h the embryonic development among the experimental groups was significantly different (p=0.018). The rate of blastocyst development and cell number per embryo were the highest in 2 ng/ml GM-CSF treatment group. The percent of apoptotic cells of the GM-CSF-treated embryos was the lowest among the group. In blastocysts, GM-CSF treatment transiently increased MAPK activity. PD098059 attenuated the effect of GM-CSF on the morphological development, increase in cell number per blastocyst, down regulation of apoptosis, and upregulation of MAPK activity, suggesting that activation of MAPK cascade possibly mediated the embryotropic effect of GM-CSF. Conclusion: This result suggested that GM-CSF potentiated the development of preimplantation mouse embryos by activation of MAPK.
