Purification and Characteristic Properties of DNA Polymerase $\alpha$ from Sea-Urchin, Hemicentrotus pulcherrismus (말똥 성게의 DNA Polymerase $\alpha$의 정제와 특성)

  • From the sea-urchin, Hemicentrotus pulcherrismus, we have purified by four column chromatographic steps for DNA polymerase $\alpha$ activity. The molecular weight of DNA polymerase u was determined to be around 137,000-138,000 by Sephadex G-200 gel filtration and SDS-polyacrylamide gel electrophoresis. The purified enzyme had the optimal activity at pH 7.4. This enzyme showed to be a function of the metal ion $K^+,\;Na^+$\;and\;Mg^{2+}$ employed as activators, the optimum $K^+$\;or\;Na^+ concentration were 20 mM or 25mM and the optimum $Mg^{2+}$ concentration was 10 mM. The enzyme activity was inhibited by N-ethyl-maleimide, aphidicolin, cytosine $\beta-D-arabinofuranoside$ 5'-triphoshate (ara CTP) and phosphonoacetic acid.

  • 3,4-Dimethyl-N-[4-chloro-2-fluoro-5-{(pyrazolylmethyl)oxy}phenyl]maleimides or 3,4,5,6-tetrahydro-N-[4-chloro-2-fluoro-5-{(pylazolylmethyl)oxy} phenyl]phthalimides were prepared and evaluated their herbicidal activities under paddy conditions. Those compounds which have N-methyl-5-pyrazolylmethyloxy moiety showed good tolerance in transplanted rice and strong herbicidal activities on barnyardgrass below 60 g/ha of dose.

  • Copolymers of N-vinylpyrrolidone (NVP) comonomer with styrene (St), hydroxypropyl methacrylate (HPMA) and carboxyphenyl maleimide (CPMI) were synthesized by free radical polymerization using 2,2'-azobisisobutyronitrile (AIBN) initiator in 1,4-dioxane solvent. The copolymers formed were characterized by FTIR, $^1H$ NMR and $^{13}C$ NMR techniques and their thermal properties were studied by DSC and TGA. Copolymer composition was determined by $^1H$ NMR and/or by elemental analysis and monomer reactivity ratios (MRR) were estimated by the linear methods of Kelen-Tudos (K-T) and extended Kelen-Tudos (EK-T) and the non-linear approach. Copolymers of St and HPMA with NVP formed blocks of one of the monomer units, whereas alternating copolymers were obtained in CPMI-NVP, depending upon the side chain substitution. The MRR values are discussed in terms of monomer structural properties such as electronegativity and electron delocalization. The sequence distribution of monomers in the copolymers was studied by statistical method based on the average reactivity ratios obtained by EK-T method.

A Study on the SH Structure Change of Myosin Head by Temperature Effect (온도 영향에 따른 Myosin Head의 SH 구조 변화 연구)

  • The effect of temperature on the structure change of the SH of myosin head have been investigated with improved resolution by x-ray diffraction using synchrotron radiation. The movement of myosin head and conformational change of contractile molecules were occurred in the muscle contraction. IASL (iodo acetamide) and MSL (maleimide) disordered the orderly helix arrangement of myosin in the rest state of spin level. The temperature effect on the structure change was great at the UL in the equatorial reflection. But those of IASL and MSL were minor. Equatorial reflection (10, 11) change inferred that myosin head was moved to the vicinity of actin filament by temperature change (from $25^{\circ}C$ to $0^{\circ}C$) at UL, but spin level was not changed. The intensity change of 143 $\AA$ and 72 $\AA$ could offer information of the mass profection of population of myosin heads along the filament axis. The slope of intensity profile of the mass profection of 143$\AA$ and reflection of MSL is appeared sharply and those of UL and IASL were not changed. The decrease of MSL actin reflection at 51 $\AA$ and 59 $\AA$ in the actin reflection change refers that the shifted myosin head binds a certain actin or changes an actin structure. From these results, we could conclude that IASL and MSL were spin labeled on SH of myosin head and disordered the helix arrangement of actin.

  • Metal ion complex of poly(N-(2,4-imidazoly)ethyl)maleimide-alt-l-octadecene (IMO-O) polymer used to confirm the possibility of molecular device made by Langmuir-Blodgett(LB) method. Electrical properties of the metal ion complex LB film were investigated using Metal/Insulator/Metal(MIM) structure. In the surface pressure-area($\pi$-A) isotherm of IMI-O polymer, the surface pressure at collapse point has a difference due to the interaction between polymer and metal ions. And the complex between polymer and metal ions could be verified through the investigation by Raman spectroscopy. In the current-voltage(I-V) property, the conductivity change of IMO-O polymer complexes due to the kinds of metal of metal ions couldn't be observed. However, the limiting area of molecules was changed by the concentration of the metal ions and the conductivity was increased with the occupied molecular area.

  • A polygonal antibody against recombinant hirudin was raised for the development of a ELISA in biological fluids. Recombinant hirudin was conjugated to maleimide activated carrie r protein, KLH and injected to a rabbit. The third booster collection of antiserum was used as primary antibody for the ELISA. The titer for the detection antibody was determined. The direct ELISA could determine the concentration of hirudin in the range of ~10ng/ml. Affinity pulified IgG was obtained and conjugated to horseradish peroxidase. Purified IgG and IgG-HRP could be used as capture and detection antibody, respectively. Although sandwich ELISA would not give the satisfactory results. it could apply for the detection of hirudin level in the range of ~20 ${\mu}$g/ml.

  • N-phenylmaleimide (PMI), a compound for strengthening the heat resistance of ABS resin, is a yellow crystal. Therefore, copolymers modified with PMI exhibit color, which limits their use. In order to overcome such disadvantage, the demand for N-cyclohexylmaleimide (CHMI), which has similar properties to PMI and also is a white crystal, is increasing. However, CHMI is difficult to industrialize due to the formation of various by-products during synthesis, which requires an additional purification process resulting in a low yield. In this study, composite catalysts were developed to improve these problems and industrially produce CHMI. Finally, CHMI was synthesized with a 85% yield and at least 99.5% purity.
