The Production of Methyl Trans-cinnamate in the Submerged Cultures of Tricholoma matsutake Mycelia

  • Methyl trans-cinnamate is a significant flavor compound of Tricholoma matsutake. Attempts were made to produce this compound by culturing the mycelium using submerged culture. No methyl trans-cinnamate could be detected when the mycelium was cultured in the basal liquid medium. However, the addition of Pinus desiflora extracts to the medium, methyl trans-cinnamate was largely produced. To find out compounds or fractions inducing methyl trans-cinnamate, dichloromethane fraction obtained from the wood extracts of P. densiflora was subjected to column chromatography. Three sub-fractions were obtained from the $CH_2CI_2$ fraction. Submerged cultured mycelium treated with sub-fraction 1 has the highest content of methyl trans-cinnamate. Maximum methyl trans-cinnamate $(470.2{\mu}g/g)$ was obtained when the first sub-fraction of dichloromethane fraction of the P. densiflora wood extracts was added to the medium. This indicates that wood extracts of P. densiflora contains inducer of the methyl trans-cinnamate production in the T. matsutake submerged culture.

Changes of Methyl trans-cinnamate Levels During Fruit-body Development in Tricholoma matsutake

  • The relationship between the contents of methyl trans-cinnamate and the ratio of DP/DS (diameter of pileus / diameter of stalk) in the fruit-body of Tricholoma matsutake during its development was investigated. The stages of development were divided as follows: stage A is less than 1, stage B is from 1 to less than 2, stage C is from 2 to less than 3, and stage D is more than 3 of the values of DP/DS. The contents of methyl trans-cinnamate in pileus and stalk of pine mushroom ranged from $77{\mu}g/g$ to $824{\mu}g/g$ and from $7.6{\mu}g/g$ to $22.4{\mu}g/g$, respectively during its development. In the part of pileus, there is no relevance of the methyl trans-cinnamate content of pine mushroom between the stage A and B, but there was significantly different among the stage of B, C and D. In the case of stalk, the relevance of the methyl trans-cinnamate content of pine mushroom between stage D and other stages showed a low difference. In addition, as pileus of pine mushroom developed the level of the aroma compound increased as well and showed higher correlation relationship ($r^2=0.877$) between the contents of methyl trans-cinnamate in the pileus and the ratio of DP/DS. From the results of this study, we can conclude that the aromatic component of pine mushroom can be deduced from the value of DP/DS, which indicates the stage of the development appearance.

  • One alkaloid derivatives, one amide and two steroids were isolated from the fruit bodies of Tricholoma matsutake. The structures were determinded as adenosine, methyl trans-cinnamate, ergosterol and ergosta-4, 6, 8 (14), 22-tetraen-3-one, respectively, on the basis of spectroscopic data.

  • This research aims at developing a new plant growth inhibitors related to abscisic acid by means of esterification of (S)-(+)-ABA with p-hydroxy methyl cinnamate and umbelliferone, and testing its biological activity on growth of rice seedlings. The over-all yield of ABA-methyl cinnamate(AC) and ABA-umbelliferone(AC) ester compounds were 83% and 78%, respectively. The growth inhibition activity of these synthetic compounds were shown about 3 to 10 times(AC) and 10 to 30 times(AU) higher than (S)-(+)-ABA.

Antioxidative Effectiveness and Oxidized Products in Mixture of Methyl Linoleate and Phenolic Compounds (Methyl Linoleate에 대한 Phenol성 물질의 항산화성과 산화 생성물)

  • Antioxidative effectiveness and oxidized products in mixture of methyl linoleate(ML) and phenolic compounds were investigated under oxygen blowing at $37^{\circ}C$ for 9 days. Caffeic acid (3,4-dihydroxy cinnamate ; CML) and phloroglucinol(1,3,5-trihydroxy benzene ; PML) showed higher antioxidative effectiveness for methyl linoleate than 0.05% ${\alpha}-tocopherol$ (TML). Oxidized products in ML group were methyl 8-(2-furyl)-octanoate, 9,13-trans, cis hydroperoxide isomer, 9,13-trans, trans hydroperoxide isomer, and 9-TMSO-12,13-epoxy-10-octadecenoate. In CML group the oxidized products were methyl-8-(2-furyl)-octanoate, 9-trans, cis hydroperoxide isomer and 9-trans, trans hydroperoxide isomer, but 13-hydroxy isomer was not identified. It was shown that CML were oxidized more slowly than ML group and at 6th day of oxidation, caffeic quinone was found to be major oxidized product of caffeic acid. Oxidixed Products in PML group were methyl-8-(2-furyl)-octanoate, 9-trans, cis hydroperoxide isomer, and 9-trans, trans hydroperoxide isomer but phloroglucinol was not oxidized even at the 9th day of reaction.

$TiO_2$-Mediated Photoreactions of Cinnamic Acid and Related Compounds in Methanol

  • Photochemical reactions of some organic molecules on titanium dioxide were investigated in methanol. A methanolic solution of trans-cinnamic acid and titanium dioxide was irradiated with 300 nm UV lamps for 24 h to afford methyl cinnamate. In the case of trans-cinnamamide, the major product was found to be 3-phenylpropionamide, i.e., a saturation product of ethylenic double bond. However, irradiation of urocanic acid, caffeic acid, ethyl cinnamate, trans-chalcone, trans-cinnamonitrile, trans-stilbene or trans, trans-1,4-diphenyl-1,3-butadiene on titanium dioxide under the same conditions did not give any noticeable products. Meanwhile, when irradiated some aromatic aldehydes, such as trans-cinnamaldehyde, l-naphthaldehyde, and 2-naphthaldehyde in methanol, vicinal diols and alcohols derived from the diols were produced. On the other hand, irradiation of 9-anthraldehyde and titanium dioxide in methanol afforded only alcohols, in which diol was not observed.

Comparison of Volatile Components in Organs of Ocimum basilicum L. cultivated in Korea (국내 재배 바질의 품종간 부위별 휘발성 성분 비교)

  • This study was conducted to compare volatile components from the ten kinds of basils cultivated in Korea. The ten kinds of basils were separated flower, leaf, and stem part from whole plants, respectively. All sample separated were extracted by simultaneous steam distillation-extraction method(SDE) and were analyzed by gas chromatography(GC) and mass selective detector(MSD). Total 42 components were identified in essential oils including 11 alcohols, 6 carbonyls, 20 hydrocarbons and 5 esters components. The major components were linalool, methyl chavicol, eugenol, trans-methyl cinnamate, ${\beta}-cubebene$ and 1,8-cineole. The content of linalool was high significantly in the flower$(31.8{\sim}53.0%)$, the leaf and stem showed $21.8{\sim}35.8%$ and $3.5{\sim}22.4%$, respectively. Especially, the content of methyl chavicol was high relatively in the leaf$(0.4{\sim}32.9%)$, the flower and stem showed $0.2{\sim}24.1%$ and $0{\sim}2.2%)$, respectively. Articock, figz, glove, and greek basils were rich in eugenol$(18.8{\sim}48.7%)$ and poor in methyl chavicol$(0{\sim}5.4%)$ when compared with others kinds of basils. The composition of the components identified showed quite difference between kinds of basils, and the number of components identified in stem was much less than that in flower and leaf

Electroluminescent Property of Tetrahydrochrysene as a Potential Emitting Layer in Organic Electroluminescent Device

  • As a potential electroluminescent material, tetrahydrochrysene (THC) is prepared using the dehydrocyclization following the acyloin condensation of methyl-3-phenyl propionate as key step from trans-mehyl cinnamate in 3 steps. THC showed emission at 428 and 456 nm after the photo- and electro-excitation, respectively. The luminance of THC doped on PVK was about /$25 cdm^2$ at 30 voltage with 70 nm of thickness. The results suggests that a new fluorescent organic dye, THC can be used organic electroluminescent device.
