초임계 이산화탄소를 이용한 마늘 추출물의 항균효과에 관한 연구 (A Study on the Antimicrobial Effect of Garlic Extract using Super-Critical Carbon Dioxide)

  • Garlic extract using super-critical carbon dioxide is influenced by temperature and pressure, and the optimum condition can make under super-critical state. We can know the defects in process of super-critical extration, It can indicate the drop of product rate, energy loss and equipment expense etcs. The minimum inhibition concentration of microbe which garlic extract contains has apperared the concentration more than 800ppm in this experiment. According to the result of this experiment, we can know that the antibiosis effect in the microbe of staphylococcus and fungus has disappeared in the incubation time more than 12 hours.

산초 정유성분의 식중독균에 대한 항균 활성 (Antibacterial Activities of Essential Oil from Zanthoxylum schinifolium Against Food-Borne Pathogens)

  • In this study, the antibacterial activities of essential oil from Zanthoxylum schinifolium against four Gram-positive bacteria and six Gram-negative bacteria were investigated. The antibacterial activity of the oils was determined using the agar-well diffusion assay, MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration). In particular, essential oil from Z. schinifolium showed higher antibacterial activity against Gram-positive bacteria than against Gram-negative bacteria. Essential oil from Z. schinifolium displayed large inhibition zones especially against Bacillus cereus (31 mm). At concentrations between 0 and $20\;{\mu}g/mL$ the oils showed an antibacterial effect against both Gram-negative and Gram-positive bacteria. The minimum inhibitory concentration (MIC) values against nine bacteria ranged from 1.25 to $5\;{\mu}g/mL$. The minimum bactericidal concentration (MBC) values against eight bacterial ranged from 2.5 to $20\;{\mu}g/mL$, except Shigella sonnei. Furthermore, our finding on the antibacterial activities of essential oils from Zanthoxylum schinifolium validated the use of this plant for medical purposes.

Inhibition of Quorum Sensing Regulated Virulence Factors and Biofilm Formation by Eucalyptus globulus against Multidrug-Resistant Pseudomonas aeruginosa

  • Objectives: The quorum-sensing-inhibitory and anti-biofilm activities of the methanol extract of E. globulus leaves were determined against clinically isolated multidrug-resistant Pseudomonas aeruginosa. Methods: The preliminary anti-quorum-sensing (AQS) activity of eucalyptus was investigated against a biosensor strain Chromobacterium violaceum ATCC 12472 (CV12472) by using the agar well diffusion method. The effect of sub-minimum inhibitory concentrations (sub-MICs) of the methanol extract of eucalyptus on different quorum-sensing-regulated virulence factors, such as swarming motility, pyocyanin pigment, exopolysaccharide (EPS), and biofilm formation, against clinical isolates (CIs 2, 3, and 4) and reference PA01 of Pseudomonas aeruginosa were determined using the swarm diameter (mm)-measurement method, chloroform extraction method, phenol (5%)-sulphuric acid (concentrated) method, and the microtiter plate assay respectively, and the inhibition (%) in formation were calculated. Results: The preliminary AQS activity (violacein pigment inhibition) of eucalyptus was confirmed against Chromobacterium violaceum ATCC 12472 (CV12472). The eucalyptus extract also showed concentration-dependent inhibition (%) of swarming motility, pyocyanin pigment, EPS, and biofilm formation in different CIs and PA01 of P. aeruginosa. Conclusion: Our results revealed the effectiveness of the E. globulus extract for the regulation of quorum-sensing-dependent virulence factors and biofilm formation at a reduced dose (sub-MICs) and suggest that E. globulus may be a therapeutic agent for curing and controlling bacterial infection and thereby reducing the possibility of resistance development in pathogenic strains.

국산 프로폴리스의 항균활성 (Antimicrobial Activities of Korean Propolis)

  • 예천산 프로폴리스는 ethanol과 butanol 분획물이 S.typhimurium과 B. subtilis에 강한 항미생물 활성을 보였으며, 용매별로는 butanol, ethyl acetate, chloroform 분획물의 순이었다. C. utilis와 A. niger에는 모든 분획물이 항균활성을 나타내지 않았다. 농도별 균생육 억제 시험 결과, E. coli의 경우 예천산은 0.4mg/ml에서 초기에 균증식이 억제되었으며 영월은 0.25mg/ml에서 12시간 이후 균증식이 완전히 억제되었다. S. aureus의 경우, 영월과 예천산 프로폴리스가 0.25와 0.30mg/ml에서 완전히 균증식을 억제시켰다. P. aeruginosa의 경우, 예천산 프로폴리스는 0.20mg/ml에서 12시간 이후 균생장 억제를 보였으며, 영월산 프로폴리스는 0.20mg/ml에서 균생장이 완전히 억제되었다. S. typhimurium의 경우, 영월산은 0.20, 0.25mg/ml에서 균증식이 거의 억제되었으며, 예천산은 0.40mg/ml에서 균증식이 거의 억제되었다. MIC는 예천산 프로폴리스가 P. aeruginosa가 >0.2mg/ml로 가장 낮았고 S. aureus 0.25mg/ml, S. typhimurium > 0.35mg/ml 순이었다. 영월산은 P. aeruginosa가 0.15mg/ml로 가장 낮았으며 E. coli, S. aureus 및 S. typhimurium이 0.25mg/ml, B. subtilis 와 C. utilis는 각각 0.3, 0.4mg/ml 이었다.

수용성 프로폴리스의 항균성 (Antimicrobial Activity of Water Soluble Propolis)

  • In this study, the minimum inhibition concentration(MIC), growth inhibition activity, and colony forming inhibitory activity of water soluble propolis against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae and Salmonella enteritidis were tested. The MICs of the water soluble propolis against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae, and Salmonella enteritidis were 312.5 ppm, below 156.3 ppm, 625 ppm, 10,000 ppm, above 10,000 ppm, 10,000 ppm, above 10,000 ppm, above 10,000 ppm, 10,000 ppm, and above 10,000 ppm, respectively. The growth inhibition concentrations against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, and Klebsiella pneumoniae were 156.3 ppm, below 156.3 ppm, 625 ppm, 5,000 ppm, 10,000 ppm, 10,000 ppm, 10,000 ppm, 10,000 ppm, and 5,000 ppm, respectively. However, 10,000 ppm did not inhibit the growth of Salmonella enteritidis. Finally, the colony forming inhibitory activities against Bacillus cereus, Bacillus subtilis, Listeria monocytogenes, Staphylococcus aureus, Streptococcus mutans, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae, and Salmonella enteritidis were 98.0%, 99.8%, 69.8%, 98.1%, 62.0%, 63.1%, 79.5%, 61.9%, 79.6%, and 0.0%, respectively.

표고버섯 용매별 추출물의 구강세균에 대한 항균효과 및 Actinomyces viscosus에 대한 생육저해 효과 (Antimicrobial activities against oral bacteria and growth inhibition against Actinomyces viscosus using Lentinus edodes various extracts)

  • Objectives: Lentinus edodes is an edible mushroom with a variety of beneficial effects such as antitumor, anti-inflammatory, antioxidant, and immune-modulatory activity. This study was carried out to evaluate the antimicrobial activities of Lentinus edodes extracts against oral-related bacteria. Methods: The antimicrobial activities of this extracts were investigated against S. anginosus, S. sobrinus, S. aureus, S. mutans, S. ratti, S. sanguinis, A. viscosus, A. naeslundii, and A. actinomycetemcomitans by the disc diffusion method, minimum inhibitory concentration (MIC), and growth inhibition. Results: Ethanol extracts had no antimicrobial activities, but acetone extracts showed antimicrobial activities against A. viscosus and A. actinomycetemcomitans and ethyl acetate extracts had effects against S. aureus, S. sanguinis, A. viscosus, and A. actinomycetemcomitans. Conclusions: The inhibitory effect of Lentinus edodes extracts was investigated on the growth of A. viscosus. Ethyl acetate and acetone extracts showed 90% and 77% inhibitory effect, respectively, against A. viscosus for 24 hrs. Ethyl acetate extracts had MIC of 25.0 mg/ml and acetone extracts showed MIC of >25.0 mg/ml.

Antimicrobial Activities of White, Red, and Extruded Ginsengs with Different Extraction Conditions

  • White, red, and extruded ginsengs were studied against 8 strains of food-borne pathogens and/or food spoilage microorganisms. The ginseng powders were extracted with different extractants and screened for antimicrobial activity using the disc diffusion and broth dilution techniques. The results showed that the yield of extraction was higher with increase of aqueous solution content and temperature. Preliminary screening revealed that the red ginseng extracts were most active, that has been found to be highly effective against all tested microbe except Listeria monocytogenes. Moreover, Bacillus subtilis has shown highly susceptible, which the diameters of inhibition zone values of 28 extracts were between 7 and 14 mm. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) recorded for the different crude ginseng extracts against microorganism using ranged from 6.25 to 100 mg/mL, indicated that the methanol extract of ginseng were more effective than ethanol and water extracts. The 60% methanol extract of red ginseng had the greatest effects against B. subtilis with MIC and MBC values at 6.25 mg/mL.

시판되는 구강양치액과 천연한방 추출물의 Streptococcus mutans에 대한 항균효과 비교 (Antimicrobial Effect of Commercially Available Mouth Rinsing Solutions and Natural Herbal Extracts on Streptococcus mutans)

  • 본 연구는 천연한방 식물 추출물의 치아우식 예방제제로서 가능성을 평가하고자 구강 양치용액과 함께 S. mutans에 대한 항균활성을 측정하고, 정상인 치은섬유모세포에 대한 독성 유무를 평가하여 다음과 같은 결과를 얻었다. 용매별 천연한방 추출물의 S. mutans에 대한 최소성장억제농도는 증류수보다 에탄올 추출물에서 우수한 성장억제 효과를 보였다. S. mutans에 대한 최소성장억제농도를 비교한 결과, 천연한방 에탄올 추출물은 보골지가, 구강양치액은 헥사메딘이 가장 낮은 농도에서 성장억제 효과를 보였다. S. mutans에 대한 최소살균농도를 비교한 결과, 천연한방 에탄올 추출물은 보골지가, 구강양치액은 헥사메딘과 가그린이 낮은 농도에서 사멸효과를 나타냈다. 정상인의 치은섬유모세포에 천연한방 에탄올 추출물을 최소성장억제농인 10, 39, $78{\mu}g/ml$로 처리하여 독성 유무를 측정한 결과 세포독성이 없었다. 그러나 최소살균농도 값을 토대로 각 추출물별 농도를 달리하여 처리한 결과, 보골지를 제외하고는 다른 추출물은 농도가 높아짐에 따라 세포독성이 있는 것으로 나타났다. 이상의 결과를 종합하면, 4종의 천연한방 추출물은 합성 화학물질인 구강양치액과 비교 시 유사한 항균효과가 있었다. 따라서 천연한방 추출물은 적정 농도를 첨가함으로써 치아우식 대체 예방제제로 실용화할 수 있을 것으로 생각된다.

Corosolic acid에 의한 Staphylococcus aureus의 생장 및 생물막 형성 저해 (Inhibition of growth and biofilm formation of Staphylococcus aureus by corosolic acid)

  • 본 연구에서는 지금까지 거의 보고된 바 없는 corosolic acid의 항균활성에 대해 대표적 식중독균 중 하나인 S. aureus를 대상으로 연구하였다. 대수기의 S. aureus에 corosolic acid를 처리하여 생장 저해 여부를 확인하였고, broth micro-dilution 방법과 agar disc diffusion 방법을 통해 corosolic acid의 S. aureus에 대한 항균활성을 측정해본 결과, corosolic acid의 농도에 비례하여 S. aureus 생장이 저해되는 것으로 확인되었다. 또한 corosolic acid의 존재 조건에서 S. aureus의 바이오필름 형성능이 MIC 보다 높은 농도뿐 아니라 낮은 농도에서 역시 저해되는 것으로 관찰되었다. 따라서 천연물질인 corosolic acid는 S. aureus에 의한 식품 오염을 억제하고 식중독 예방 및 잠재적 치료제로서 개발 될 가능성이 있는 것으로 판단되며, 나아가 다른 종류의 식중독균들에 대한 적용 가능 연구가 필요할 것으로 사료된다.

Heavy Metal Tolerance of Fungi Isolated from Contaminated Soil

  • This study was conducted to investigate the tolerance of some resistant fungal strains from soils contaminated with heavy metals. Various fungal strains were isolated from soil samples collected from studied sites which heavy metals and other pollutants have been emitted in effluents for several years. Fungi isolated belong to different genera; however, Penicillium spp. showed the most frequent species. The microbial number was remarkably higher in the control soil than contaminated soil samples collected from mining areas. $Pb^{2+}$ and $Zn^{2+}$ had the highest concentration in the polluted soils ranging from 89 - 3,521 ppm and 98 - 4,383 ppm, respectively. The minimum inhibition concentrations (MICs) of $Pb^{+2}$ and $Zn^{+2}$ showed the highest values against the fungal strains. $Ni^{+2}$ and $Co^{+2}$ were the lowest contaminants in the polluted soils with the concentration of 5 to 12.1 ppm and 1.8 to 4.8 ppm, respectively. The tested resistant strains showed the strongest inhibition for $Ni^{+2}$ and $Co^{+2}$ up to 200-400 ppm. Cadmium was the most highly toxic heavy metal for most of strains, however, 1 mM of $Cr^{3+}$, $Cu^{2+}$ and $Pb^{2+}$ accelerated the growth of Penicillium verrucosum KNU3. $Cu^{+2}$ and $Zn^{+2}$ at concentration of 1 mM did not affect the growth rate P. funiculosum KNU4. Tolerance of fungal species to heavy metals appears to be strain and origin dependent.
