Metabolite analysis in the type 1 diabetic mouse model

  • Type 1 diabetes mellitus (T1DM) is caused by insufficient production of insulin, which is involved in carbohydrate metabolism. Type 2 diabetes mellitus (T2DM) has insulin resistance in which cells do not respond adequately to insulin. The purpose of this study was to estimate the characteristics of type 1 diabetes using streptozotocin-treated mice (STZ-mouse). The sera samples were collected from the models of hyperglycemic mouse and healthy mouse. Based on the pair-wise comparison, five metabolites were found to be noticeable: glucose, malonic acid, 3-hyroxybutyrate, methanol, and tryptophan. It was very natural glucose was upregulated in STZ-mouse. 3-hyroxybutyrate was also increased in the model. However, malonic acid, tryptophan, and methanol was downregulated in STZ-mouse. Several metabolites acetoacetate, acetone, alanine, arginine, asparagine, histidine, lysine, malate, methionine, ornithine, proline, propylene glycol, threonine, tyrosine, and urea tended to be varied in STZ-mouse while the statistical significance was not stratified for the variation. The multivariate model of PCA clearly showed the group separation between healthy control and STZ-mouse. The most significant metabolites that contributed the group separation included glucose, citrate, ascorbate, and lactate. Lactate did not show the statistical significance of change in t-test while it tends to down-regulated both in DNP and Diabetes.

  • An important factor in eye-tracking PC interface for general paralyzed patients is the implementation of the mouse interface, for manipulating the GUI. With a successfully implemented mouse interface, users can generate mouse events exactly at the point of their choosing. However, it is difficult to define this interaction in the eye-tracking interface. This problem has been defined as the Midas touch problem and has been a major focus of eye-tracking research. There have been many attempts to solve this problem using blink, voice input, etc. However, it was not suitable for general paralyzed patients because some of them cannot wink or speak. In this paper, we propose a mouth-pop-up, eye-tracking mouse interface that solves the Midas touch problem as well as becoming a suitable interface for general paralyzed patients using a common RGB camera. The interface presented in this paper implements a mouse interface that detects the opening and closing of the mouth to activate a pop-up menu that the user can select the mouse event. After implementation, a performance experiment was conducted. As a result, we found that the number of malfunctions and the time to perform tasks were reduced compared to the existing method.

Exercise-induced beige adipogenesis of iWAT in Cidea reporter mice

  • Obesity is caused by an imbalance between energy intake and energy expenditure. Exercise is attracting attention as one of the ways to treat obesity. Exercise induces 'beige adipogenesis' in white adipose tissue, increasing total energy expenditure via energy dissipation in the form of heat. Also, beige adipogenesis can be induced by treatment with a beta-adrenergic receptor agonist. We developed a Cidea-dual reporter mouse (Cidea-P2A-Luc2-T2A-tdTomato, Luciferase/tdTomato) model to trace and measure beige adipogenesis in vivo. As a result, both exercise and injection of beta-adrenergic receptor agonist induced beige adipogenesis and was detected through fluorescence and luminescence. We confirmed that exercise and beta-adrenergic receptor agonist induce beige adipogenesis in Cidea-dual reporter mouse, which will be widely used for detecting beige adipogenesis in vivo.

Mouse Strain-Dependent Osteoclastogenesis in Response to Lipopolysaccharide

  • Bacterial lipopolysaccharide (LPS) is a potent stimulator of bone resorption in periodontitis. Co-culture systems of mouse calvaria-derived osteoblasts and bone marrow-derived preosteoclasts were used as an in vitro osteoclast differentiation. This study revealed that co-cultures using ddY or ICR mouse strain responded differently to LPS while responded equally to $1{\alpha},25(OH)_2D_3$. Thus, the different response to LPS indicates dissimilarity of two mouse stains in their capacity for generating osteoclasts while the two mouse strains share the similarity in response to $1{\alpha},25(OH)_2D_3$. To identify which cells between osteoblasts and preosteoclasts in the co-culture are responsible for the dissimilarity, the reciprocal co-cultures were performed between ddY and ICR mouse strains. The treatment of $1,25(OH)_2D_3$ to ddY/ICR (osteoblasts from ddY/preosteoclasts from ICR) and ICR/ddY reciprocal co-cultures also showed the similarity. In case of LPS treatment, the results of ddY/ICR were similar to ddY/ddY and the results of the other reciprocal co-culture, ICR/ddY combination, were consistent with those of ICR/ICR. It suggests that the dissimilarity between the two mouse strains may resident in osteoblasts but not in preosteoclasts. Therefore, the osteoblast is responsible for mouse strain-dependent osteoclastogenesis in response to LPS. Although mouse models will continue to provide insights into molecular mechanisms of osteoclastogenesis, caution should be exercised when using different mouse strains, especially ddY and ICR strains as models for osteoclast differentiation.

성숙난포액을 이용한 생쥐배아의 발달에 관한 연구 (Effect of Mature Human Follicular Fluid on the Development of Mouse Embryos in vitro)

  • The possible effect of human follicular fluid(hFF) on the growth and development of fertilized oocytes and embryos is important because the fallopian tubes are exposed to FF after follicular rupture and the processes of fertilization and embryo cleavage occur inside the fallopian tubes. Previously, it was suggested that human FF might adversely affect on the development of early mouse embryos. In order to investigate the effect of hFF on the development of embryos, early mouse embryos were cultured in media containing various protein sources as bovine serum albumin(BSA), fetal cord serum(FCS) and FF. And we evaluated the development of early mouse embryos in terms of the morphology, cleavage rate, and cell count of blastcysts. There were no significant differences in the morula and blstocyst formation rates of 2-cell mouse embryos cultured in the media containg three different protein sources and three different concentrations of FF. The blastocyst formation rate of 1-cell mouse embryo cultured in FF group was significantly higher than that cultured in BSA group(P<0.05). The morula and blastocyst formation rates of 2-cell mouse embryos of the group cultured in the media containing FF were comparable with those of other two groups, in addition, the cell count of blastocysts of FF group in the 2-cell embryo culture was higher than those of BSA group and HCS group(P<0.01), and this finding was also noted in 1-cell embryo culture. There was no difference in the morula and blastocyst formation rates of the 2-cell mouse embryos cultured in the media containing different concentrations of FF. These results suggest that mature human follicular fluid has no inhibitory activity on the development of early mouse embryos even in high concentration and may be a good protein source which is positively associated with the development of mouse embryos in vitro especially in 1 cell embryo culture.

Characterization of Brain Tumor Cell using Vasopressin-SV40 T Ag Transgenic Mouse

  • In previous reports, pVPSV.IGR2.1 transgenic mouse were described that brain tumor and lymphoma by reason of Vasopressin-SV40 T antigen. In this study, we produced pVPSV.IGR3.6 transgenic mouse that used pVPSV.IGR3.6 vector. Expression of transgene was vary different in transgenic mouse. We obtained 6 transgenic mouse line, moreover they had died at the age of 2~6 weeks without transmitting the transgene to their offspring, and had tumorigenesis on same location with pVPSV.IGR2.1 transgenic mouse. Only a founder mouse was investigated for expression of fusion gene. Here we extended this transgenic approach to the study of tumor progression. From the mouse, we confirmed brain tumor cell, after then cultured for investigate characterization. In this report, we demonstrate that reduction of survival rate in transgenic mouse fused vasopressin gene length, acquisition of brain tumor cell, composition with astrocyte cells and neuronal cells. Finally, cells had no change with increase of passage.

진동촉각 햅틱 마우스 기반 버블포핑 증강현실 시스템 (Bubble Popping Augmented Reality System Using a Vibro-Tactile Haptic Mouse)

  • 본 논문에서는 증강현실 응용으로 진동촉각 햅틱 마우스를 이용하여 버블 포핑을 구현하는 버블포핑 증강현실 시스템을 제안한다. 이 시스템에서는 가상 그래픽 버블이 3차원 공간에서 랜덤하게 떠다닌다. 사용자의 진동촉각마우스가 버블과 접촉하면 버블이 터지는 포핑이 발생한다. 또한 포핑 효과와 더불어, 마우스가 진동을 함으로써 마우스를 잡고 있는 사용자의 손에 보다 실감나는 버블포핑을 전달한다. 제안 시스템은 진동촉각마우스, 비디오 입력용 웹캠, 마커 패턴 영상, 그래픽 구 버블 객체, 그래픽 마우스 등으로 구성된다. 버블과의 충돌이 발생하게 되면, 마우스 진동과 버블 포핑의 두가지 효과를 동시에 제공한다. 버블포핑 이벤트로 페이드 아웃 및 발산 이드 아웃 기법을 제안한다. 따라서 시각 및 촉각 버블 포핑은 단일 효과보다 실감있는 증강현실을 전달한다.

Epidermal Growth Factor(EGF)가 생쥐 초기배아의 발생에 미치는 영향 (Effect of Epidermal Growth Factor(EGF) on Early Embryonic Development in Mouse)

  • Growth factors (GFs) produced by the embryo or by the maternal reproductive tract have been reported to regulate the embryonic development and differentiation. Among GFs, EGF as a mitogen plays a role in mitosis and functional differentiation of trophectoderm cells in mouse. The present study was carried out to investigate the effect of EGF on development of mouse embryos and to localize EGF in the mouse oocytes and embryos, which has been reported to be detected in the reproductive tract in mammals. To investigate the effect of EGF on the development of the embryo, mouse 2-cell embryos were cultured to blastocysts stage in Ham's F10 medium, treated with EGF(10-50 ng/ml) for 72 hrs. Immunocytochemistry was performed from oocyte to blastocyst stage with anti-EGF and anti-Mouse IgG, in order to determine the stage which EGF would be expressed in mouse. Exogenous EGF (more than 10 ng/ml) in the culture medium improved the developmental and hatching rates in the mouse embryos. As a result of immunocytochemistry, the embryonic EGF was expressed after the late 4-cell stage. EGF is thought to enhance preimplantation embryonic development and hatching. Exogenous EGF in the culture medium is thought to activate EGF receptor in the late 4-cell embryos and to enhance blastulation and hatching in mouse embryos. It is concluded that EGF enhances the developmental and hatching rates in the mouse embryos.

Mouse Models of Gastric Carcinogenesis

  • Gastric cancer is one of the most common cancers in the world. Animal models have been used to elucidate the details of the molecular mechanisms of various cancers. However, most inbred strains of mice have resistance to gastric carcinogenesis. Helicobacter infection and carcinogen treatment have been used to establish mouse models that exhibit phenotypes similar to those of human gastric cancer. A large number of transgenic and knockout mouse models of gastric cancer have been developed using genetic engineering. A combination of carcinogens and gene manipulation has been applied to facilitate development of advanced gastric cancer; however, it is rare for mouse models of gastric cancer to show aggressive, metastatic phenotypes required for preclinical studies. Here, we review current mouse models of gastric carcinogenesis and provide our perspectives on future developments in this field.

햅틱마우스를 이용한 인터랙티브 게임 (An Interactive Game with a Haptic Mouse)

  • 본 연구에서는 몰입형 인간 컴퓨터 상호작용을 위한 햅틱 마우스 시스템을 제작한다. 제작한 햅틱 마우스는 진동감각 뿐 아니라 온열감도 제공하기 때문에 사용자는 더욱 몰입감 있게 가상의 공간을 경험할 수 있다. 진동 감각과 온열감을 제공하기 위하여 편심모터들과 솔레노이드, 그리고 펠티어 엑츄에이터를 이용하였으며 제작한 시스템의 평가를 위하여 레이싱 게임 프로토타입을 구현하여 테스트를 수행한다. 실험결과로부터 본 햅틱 마우스는 가상 공간에서 사용자에게 사실감을 전달함을 알 수 있다.
