혐기처리한 뽕잎의 혈당강하 효과 (Hypoglycemic Effect of Mulberry Leaves with Anaerobic Treatment in Alloxan-induced Diabetic Mice)

  • The hypoglycemic effect of mulberry leaves using alloxan-induced diabetic mice was investigated. The hypoglycemic effect of mulberry leaves was significantly affected with anaerobic incubation up to for 6 hrs. The body weight of mice treated with 20% anaerobic conditioned mulberry leaves was higher than that in the control group. Mulberry leaves which were treated with anaerobic condition significantly decreased blood glucose level in alloxan-induced type I diabetes mice. Furthermore, glucose tolerance was significantly recovered by feeding mulberry leaves anaerobically treated. Pathological analysis revealed that the disappearance of insulin-secreting beta-cells in the islets of Langerhans from the alloxan induced diabetic mice was strongly inhibited in the mulberry leaves feeding group. The content of 1-deoxynojirimycin increased up to 5% in detached mulberry leaves incubated under nitrogen gas for 6 hrs. Nevertheless, it is not clear whether the synthesis of 1-deoxynojirimycin is increased or the degradation of it is decreased. Overall, results obtained from this study indicate that the hypoglycemic effect was increased by the mulberry leaves with anaerobic treatment.

뽕잎차의 제조와 그 품질 특성 (Preparation of Mulberry Leaves Tea and Its Quality Characteristics)

  • An optimal manufacturing process was established for the mass-production of mulberry leaves tea. The produced tea contained abundant dietary fibers, $\beta$-carotenes, and calcium. Also the polyphenol content and antioxidant activity of mulberry leaves tea were higher than those of simply dried mulberry leaves. These results imply that this tea manufacturing process is effective to utilize the useful components of mulberry leaves.

  • Background : Diabetes is a chronic disease characterized by deficiency of insulin. It has been reported that Fructus aurantii and mulberry leaves have the effect of anti-diabetes. Methods : To investigate the improvement of mixed extracts of Fructus aurantii and mulberry leaves on lipid metabolism, we performed double-blinded, randomized and placebo-controlled multi-center clinical trial. Results : Mixed extracts of Fructus aurantii and mulberry leaves significantly reduced total cholesterol. Conclusion : These results suggest that mixed extracts of Fructus aurantii and mulberry leaves improve lipid metabolism.

뽕잎 항산화능 및 뽕잎가루 머핀의 품질특성 (Studies on Functional Properties of Mulberry Leaf Extracts and Quality Characteristics of Mulberry Leaf Muffins)

  • The function of mulberry leaves and the quality of muffins including mulberry leaves were examined. The electron donating ability of both a water and ethanol extract was 40% for both at 100 ppm, 62 and 72% at 500 ppm, and 77 and 83% at 1,000 ppm respectively. These experiments showed good oxidized substance activity. The SOD-like ability of the water and ethanol extracts at 1,000, 500, and 300 ppm was 49% and 55%, 33% and 39%, and 28% and 33% respectively. These results show that the ethanol extract had higher SOD-like ability than that of the water extract. The nitrite scavenging abilities of 1,000 ppm of the mulberry leaf water extract and ethanol extract at pHs of 1.2 and 3.0 were 40.5 and 47%, and 20 and 22% respectively. The Hunter lightness value tended to be lower as the amount of added mulberry leaves increased compared to the contrast set, so the lightness level with a 7% addition was the lowest. The contrast set was the highest for redness and added mulberry leaves tended to decrease redness at 3% but tended to increase it again with additions >3%. The b-value (yellowness) showed a similar tendency as redness. Adding Mulberry leaves tended to decrease the contrast set to the lowest level, then values increased slowly to a peak at 5% added mulberry leaf and then decreased thereafter. Strength and hardness were less after increasing added mulberry leaf. Cohesiveness, springiness, gumminess and brittleness, decreased at up to 5% added mulberry leaves but the 7% addition showed the same level as the contrast set. The mulberry leaves muffins were less preferred to the contrast set in appearance and color. However, the mulberry leaf muffins were preferred to the contrast set in taste and flavor but the result was not significant. Overall the quality of the muffins with 3% added mulberry leaves was preferred over the 1% added mulberry leaf muffins.

월동 및 해동기 뽕나무의 생화학적 물질의 변동과 내동성과의 관계 (Studies on the Change of Biochemical Components during Wintering and Thawing Periods and Cold Hardiness of Mulberry(Morus))

  • In relations to cold acclimation, experiment was carried out to understand the seasonal changes in reserve substances of the mulberry. The shoot barks and leaves of three mulberry varieties(Kaeryangppong, Shinilppong and Yongcheonppong) were sampled, after that their reserve substances were analyzed. The cold hardiness of mulberry was investigated by DTA(Differential Thermal Analysis) method. To increase cold hardiness, gibberellin(100 ppm), kinetin(100 ppm) and Jambi 8 were sprayed on the mulberry leaves. After spraying, falling of the leaves of Yongcheonppong occured earier than the other varities. After the first frost, all of treatments except gibberellin were entirely fallen. Growth regulator extended the leaves fallen. After spraying, water of the shoot barks was not showed difference in the content among the treatments, but amino acid, carbohydrate and soluble protein increased from September to October. Starch content of the shoot barks and leaves was maximum in October, but thereafter decreased during wintering stage. In Shinilppong, Jambi 8 spray increased cold hardiness by 1-2$^{\circ}C$ more than no spray. It was concluded that the cold hardiness of the mulberry in midwinter is closely related to the reserve substances with spraying Jambi 8 on the mulberry leaves.

Antioxidant Effects of the Mixture of Mulberry Leaves and Silkworm Powder on the Plasma and Liver in Streptozotocin-Induced Diabetic Rats

  • This study was carried out to examine the antioxidant effects of a mixture of mulberry leaves and silkworm powder in plasma and liver of streptozotocin-induced diabetic rats. Sprague-Dawley male rats weighing 100$\pm$10 g were used and their diets were supplemented with $0.4\%$ (4 g/kg) of the mixtures. Experimental groups were diabetic rats without supplements (DM group) or with a combination of the supplements: $100\%$ mulberry leaves (M group), $25\%$ silkworm powder mixed with mulberry leaves (25SM group), $50\%$ silkworm powder mixed with mulberry leaves (50SM group), $75\%$ silkworm powder mixed with mulberry leaves (75SM group) or $100\%$ silkworm powder (100S group). The rats were fed experimental diets and water ad libitum. All animals were injected with streptozotocin at the $3^{rd}$ week for inducing diabetes and were sacrificed on $9^{th}$ day thereafter. Hepatic xanthine oxidase (XOD) activity significantly decreased in the mixture supplemented groups compared to the DM group. Hepatic superoxide dismutase (SOD) activity was not significantly different among any of the experimental groups, but glutathione peroxidase (GSH-px) activity increased in the mixture supplemented groups compared to the DM group. In particular, it was the highest in the 50SM group. The hepatic TBARS values were lower in all the mixture supplemented groups than in the DM group, and it was as lowest when ratio of mulberry leaves to silkworm powder was highest. Hepatic lipofuscin contents were similar with the TBARS value. In conclusion, the mixtures containing silkworm powder reduced oxidative damage by strengtbening the antioxidative system and suppressing oxidative stress in the STZ-induced diabetic rat. The 1:1 blend of silkworm powder and mulberry leaves was the most effective combination for antioxidant activity.

뽕잎분말을 첨가한 당면의 생리활성 평가 (A Study on the Physicochemical Activities of Dangmyon (starch vermicelli) Added with Mulberry Leaves Powder)

  • This study was performed to investigate the physicochemical activities such as mineral contents, antioxidantant activities, ${\partial}$-glucosidase activity, and angiotensin I-converting enzyme(ACE) inhibitory effect of Dangmyon (starch vermicelli) prepared with mulberry leaves powder. Mulberry leaves powder was added in ratio (w/w) of 0% (CON), 0.5% (MD1), 1.0% (MD2), 1.5% (MD3), and 2.0% (MD4), and then mineral contents, total polyphenol contents, antioxidant, antidiabetic, and antihypertensive activities of the Dangmyon were measured. The mineral contents were significantly increased by the addition of mulberry leaves powder (p<0.001). The total polyphenol contents and antioxidant activities of Dangmyon were significantly increased by the addition of mulberry leaves powder (p<0.05). In terms of antidiabetic and antihypertensive activities showed MD1~MD4 samples higher contents of ACE and ${\alpha}$-glucosidase compared to control (p<0.05). Our findings suggest that addition of mulberry leaves powder to Dangmyon may improve the effects of anti-oxidation, anti-DM, and anti-hypertension and provide health benefits of consumers.

YK-209뽕잎이 Streptozotocin 유발 당뇨쥐 소장의 이당류분해 효소 활성과 혈당강하에 미치는 영향 (Effects of YK-209 Mulberry Leaves on Disaccharidase Activites of Small Intestine and Blood Glucose-Lowering in Streptozotocin-Induced Diabetic Rats)

  • YK-209뽕잎이 STZ유발 당뇨쥐의 혈당강하작용에 미치는 영향을 관찰코져 정상군(normal group)과 STZ 유발 당뇨군으로 나누고 당뇨군은 다시 식이내 YK-209 뽕잎 함량에 따라 각각 YK-209 뽕잎을 공급하지 않은 당뇨 대조군(DM group),0.1% 뽕잎 공급군(DM-0.1Y group),0.2%뽕잎 공급군(DM-0.2Y group) 및 0.4% 뽕잎 공급군(DM-0.1Y group)으로 나누어 자유섭식시켰다. 3주후 STZ으로 당뇨를 유발한 후 9일째에 희생하였다. 본 실험에 사용된 YK-209 뽕잎 시료의 기능성 성분 함량은 DNJ(1-deoxynoijirimycin), rutin GABA(${\gamma}$-aminobutyric) 및 vitamin C는 1.93 mg/g,2,97 mg/g, 2.36 mg/g, 1.3 mg/g이었으며 청일 뽕잎에 비해 각 1.3배, 1.4배, 1.3배, 1.6배 높았다. 체중증가는 당뇨유발 후 모든 당뇨군에서 현저하게 감소되었으며 당뇨군간에 유의적인 차이는 없었다. 소장의 길이는 실험군간에 유의적인 차이가 없었다. 혈당수준은 YK-209 뽕잎 공급 군에서 비공급군에 비해 유의적으로 감소되었으며 농도에 따른 유의적인 차이는 보이지 않았다. 소장 점막중의 maltase, sucrase 및 lactase 활성은 DM군의 소장 proximal 부분에서 증가되었으나 뽕잎 첨가군의 proximal 부분에서는 모두 저해되었다. 결론적으로 뽕잎은 STZ 유발 당뇨쥐 소장의 proximal 부분의 이당류 분해효소 활성을 억제시켜 혈당 상승을 완하시킴을 알 수 있다.

오디, 뽕잎 및 누에의 혼합비율에 따른 Streptozotocin 유발 당뇨쥐에서의 항산화 효과 및 지질대사개선 효과 (Antioxidant Effects and Improvement of Lipid Metabolism of Mulberry fruit, Mulberry Leaves and Silkworm Powder with Different Mixing Ratios in Streptozotocin-Induced Diabetic Rats)

  • This study was conducted to investigate the effects of mulberry fruit, mulberry leaves and silkworm powder with different mixing ratios on hepatic antioxidative system and lipid metabolism in streptozotocin-induced diabetic rats. Sprague-Dawley male rats weighing $100{\pm}10g$ were induced diabetic by 50 mg/kg bw streptozotocin and randomly assigned to following experimental groups; normal diet group (DM), 0.3% and 0.6% mulberry fruit diet groups (F and 2F), 0.3% mulberry leaves diet group (M), 0.3% silkworm powder diet group (S), 0.15% mulberry fruit+0.15% mulberry leaves diet group (FM), 0.15% mulberry fruit+0.15% silkworm powder diet group (FS), 0.1 % mulberry fruit+0.1 % mulberry leaves+0.1% silkworm powder diet group (FMS). The experimental diets were fed for 4 weeks. Hepatic SOD activity was not changed significantly by any of single or combined supplementations of mulberry fruit, leaves and silkworm powder but GSH-px and catalase activities were increased by the groups supplemented with two or three of the test ingredients (FM, FS, FMS) as compared with the DM group. Hepatic TBARS value was not reduced significantly by any of the supplementations but lipofuscin contents were significantly reduced in the FM, FS and FMS groups as compared with the DM group. Hepatic mitochondria and microsomal carbonyl values were reduced by the single and combined supplementations of the test ingredients. Hepatic HMG-CoA reductase activities were increased in the all supplementation groups as compared with the DM group. Hepatic total lipid and triglyceride contents were increased but cholesterol contents reduced in the supplemented groups. The effects on the enzyme activities, peroxide or its products and lipid contents were most remarkable in the FMS group. In conclusion, mulberry fruit, mulberry leaves and silkworm powder have the favorable effects on antioxidative system and lipid metabolism in the diabetic liver and the mulberry fruit, leaves and silkworm powder with equal ratio exert the synergistic effect expectedly to prevent diabetic complications.

고탄수화물 식이 섭취 마우스에서 상엽 및 누에 추출물의 혈당강하 효과 (Blood Glucose Lowering Effects of Mulberry Leaves and Silkworm Extracts on Mice Fed with High-Carbohydrate Diet)

  • Mulberry leaves(Mori folium) and silkworm(Bombyx mori) are potnet inhibiters of intestinal $\alpha$-glycosidase, and inhibit the digestion of starch and sucrose in the small intestine. They are able to prevent postprandial hyperglycemia and decrease blood insulin levels. In this study , a high-carbohydrate diet(CHO ; 67.5%, protein ; 20.8%, fat : 11.7%) was received by the control group. In contrast, the experimental groups received a high-carbohydrate diet with extracts of mulberry leaves and silkwork(50mg.100g diet), and acarbose(6.7mg/100g diet). after a 10 week study period , the experimental groups had lower blood glucose and triglyceride levels. The experimental groups tended to have lwer Hb Alc levels. Also, blood insulin levels were lower than the control groups in accordance with blood glucose levels. The activities of intestinal $\alpha$-glucosidase in the middle and distal parts of small intestine were induced by the extracts of mulberry leaves and silkworm in the experimental groups. However, the activities of liver lysosomal glucosidase and the contents of glycogen in the liver were not affected by the mulberry leave and silkworm extracts nor by acarbose. Mulberry leaves and silkworm were able to prevent sudden postprandial peaks in blood glucose as a result of $\alpha$-glycosidase, inhibition, there by decreasing unnecessary insulin secretion.
