상백피 추출물의 향균력 및 최적추출조건 검토 (Investigation of Optimum Extracting Condition and Antimicrobial Activity of the Extract from the Root Bark of Morus alba)

  • In order to develop a natural food preservative, the root bark of Morus alba was extracted with several solvents, and then antimicrobial activity was investigated. The optimum extracting condition for the antimicrobial substance from the sample, minimum inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. The antimicrobial activity of the ethanol extract form the sample was stronger than those of the extracts by the other solvents such as water, methanol. ethyl acetate and acetone. The optimum extracting condition for antimicrobial substance from the sample was shaking extraction twice for 5 hours at room temperature in case of 7 times of absolute ethanol added to the crushed root bark of Morus alba. The ethanol extract from the root bark of Morus alba had strong B. cereus, L. monocytogenes and S. aureus. Especially, Bacillus species was the most susceptible to the extracted substance. The ethanol extract showed antimicrobial activity against Gram negative bacteria(MIC, 160~1600 ug/ml) and yeasts(MIC, 1600 ug/ml) such as C. albicans and S. acidifaeciens. The extract also showed growth inhibition against molds such as A. niger, A. parasiticus, A versicolar and T. viride.

  • Antimicrobial studies on reproductive organs have been scarcely reported. Candida albicans and Neisseria gonorrhoea test to find out the natural substances of various concentrations in the antimicrobial experiments have been made. Antimicrobial effect of biopole as the natural compound biosynthesis matter that contain anti-inflammatory material, catechin polyphenols and lactic acid as a single natural substance on C. albicans were of great significance. Also the results of antimicrobial effects of biopole, catechin and lactic acid as a single natural substance on N. gonorrhoea, respectively, showed lower concentration than those of the antimicrobial effects on C. albicans. Through the survival of Lactobacillus acidofillus that acts for the protection of the genital tissue the importance of lactic acid was confirmed. Lactobacillus acidofillus protection and C. albicans firmly into disinfected to low concentrations of the natural mixture from biopole and catechin with lactic acid were found and the antimicrobial effects of this natural mixture on N. gonorrhoea were perfect. C. albicans and N. gonorrhoea to disinfect the optimal natural mixture from 2% concentration biopole, 0.2% concentration of catechin and 2% lactic acid were obtained. Through the survival of Lactobacillus acidofillus in the reproductive organs protectable effects were acquired to prevent the infections of reproductive tissue and the recurrence.

  • To investigate the effect of several bactericides and fungicides against fruit & vegetable decay grapefruit seed extract(GFSE) was tested in a dilution solution. GFSE was shown to be effective against decay and rind breakdown and to extend the shelf-life of fruits and vegetables during storage and tiansport. Antimicrobial efficiency of GFSE on a wide spectra of gram + & - bacteria, moulds and yeasts was demonstrated by the measurement of minimal inhibitory concentrations performed (Bacteria 10-500ppm, Fungi:250-1,000ppm, Yeasts:100-250ppm). GFSE was separated and extracted into water-soluble fraction, water-insoluble and non-dialyzed fraction to isolate the antimicrobial substances. The water-soluble fraction showed the most active antimicrobial effect. The antimicrobial substances were isolated by gas chromatography. As the result of the isolation using GC, Peak-D was found to be the antimicrobial compound in GFSE. The identification of the most antimicrobial substance was carried out by using GC-MS.

  • The extract of Ulmus davidiana var. japonica cortex has known as natural anti-inflammatory substance in East Asia. For the identification of antimicrobial substance, it was extracted by using methanol and fractionated by using different organic solvents. The fraction of butanol was represented the highest antimicrobial activities. Therefore, the butanol fraction was purified and identified the chemical structure by $^1$H and $^{13}$ C-NMR spectra, FT-IR and EI/MS spectroscopies. The isolated antimicrobial substance was identified as cis-2-[3,4-dihydroxy phenyl]-3,4-dihydro-2H-1-benzopyran-3,5,7-triol, which has commonly known as (-)epicatechin. Its minimum inhibitory concentrations (MICs) against Staphylococcus aureus and Listeria monocytogenes were shown as 100 $\mu\textrm{g}$/mL, respectively.

The optimum extracting condition for the antimicrobial sustance from the sample, minimal inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of the initial ethanol extract from the sample was the strongest compared to those of other solvent extracts such as n-hexane, acetone, butanol, methanol and water. the optimum extractingcondition for antimicrobial substance from the sample was shaking extraction for 2 hours at room temperature incase that 10 volumes of absolute ethanol was added to crushed Saliva Miltiorrhiza. The ethanol extract had strong growth inhibition activity against Gram-positive Bacteria (MIC, 3.13-50$\mu\textrm{g}$/ml) such as B. cereus, B, subtilis, L. minocytogenes, S. aureus, Sc. Mutans. Among Grampositive bacteria tested, Bacillus species was the most susceptibile to the extracted substance. The antimicrobial activity of the ethanol extract from the sample was weak to Gram -negative bacteria yeasts, for example MIC for Gram-negative bacteria and yeasts was 0.8mg/ml and 0.4-0.8mg/ml , respectively.

  • 218종의 목본식물 자원으포부터 높은 항율력올 보인 Schima wallichii subsp. liukiuellsis로부터 항균활성물질을 추출, 정제하였다. Schim$\alpha$의 향균물질 추출에 가장 적합한 용매는 70% 에탄올이었으며, 계젤에 따른 항균활성은 차이를 보이지 않았고, 부위별로는 가을에 채취한 수피가 가장 좋았다. 조추출물을 유가용매로 정제하여 최종적으혹 buthanol 분획올 얻었고, 이를 silica gel과 sephadex LH-20 및 HPLC분석을 통 하여 흰색결정의 항균물질 Compound I을 얻었다. 항균물질은 UV, IR, MS분석 결과 aglycone으로 ${\alpha}$-sitosterol에 rhamnose, galactose, glucose가 1:1:1로 결합된 물질로 추정되었다. Compound I의 미생물에 대한 MIC는 3종의 bacteria에 L 1.25 g/L, 2종의 fungi에 5.0 g/L로 나타났으며, 효모에 대한 MIC는 0.04 g/L로 매우 높게 나타났다. Compound I은 천연 보존제, 의약품 등으로의 향후 개발이 기대된다.

  • In order to develop a antimicrobial agent, dried Sophora flavescens Ait. was extracted with several solvents, and than antimicrobial activity was investigated. The minimal inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of the initial methanol extract from the sample was the strongest compared to those of other solvent extracts such as $H_2O$, n-hexane, chloroform, ethyl acetate and methanol. The methanol extract had strong growth inhibition activity against gram-postive bacteria (MIC, $25~50{\;}{\mu\textrm{g}}/ml$) such as B. subtilis. Among gram-positive bacteria tested, B. subtilis was the most susceptible to the extracted substance. The antimicrobial activity of the methanol extract from the sample had strong growth inhibition activity against gram-negative bacteria such as P. aeruginosa (MIC, $25{\;}{\mu\textrm{g}}$/ml).

  • Salt is a common seasoning agent used in various processed foods, especially in kimchi and salted seafood (jeotgal). This study was conducted to investigate the efficacy of salt with antimicrobial substances (acetic acid, garlic extract, carvacrol, nisin, thymol, and their combination (acetic acid+nisin+thymol)) on improvement of antibacterial effects of salt against foodborne pathogens. Salt (10%) was prepared using six different types of 0.2% natural antimicrobial substances. The antibacterial effect of salt combined with natural antimicrobial substances was evaluated against foodborne pathogens using the broth micro-dilution method and growth curve plotted using absorbance measurements. For the five foodborne pathogens, the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of salt without antimicrobial substances as control were in the range of 24~>50,000 ㎍/mL and >50,000 ㎍/mL, respectively. Salt with nisin, thymol, or garlic extract showed strong inhibitory effects and their MIC against L. monocytogenes were 49, 12,500, and 24 ㎍/mL, respectively. In particular, salt with nisin showed inhibitory activities against Gram-positive bacteria. However, all the antimicrobial substances were less effective against Gram-negative bacteria such as E. coli O157:H7 and S. Typhimurium than Gram-positive bacteria. These results could be used for the development of salt with natural antimicrobial substances especially targeted against L. monocytogenes. This would enable the lowering of saline concentration while improving the storability of food.

  • 땅콩껍질에 함유된 항미생물 활성물질의 탐색을 위해 땅콩껍질을 MeOH로 추출하고 이 MeOH 추출물을 용매분획하여 EtOAc 중성획분을 얻었다. 이 획분에 함유된 항미생물 활성물질을 silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, ODS column chromatography로 정제한 다음 HPLC를 이용하여 1종의 화합물을 단리하였다. $^1H-NMR$, MS 및 NOESY 분석에 의해 땅콩껍질로부터 분리된 항미생물 활성물질은 pratensein으로 동정되었다.

  • This study was carried out to evaluate cytotoxic effects of Houttuynia cordata Thunberg extracts on murine leukemia tumor cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazol iumbromide (MMT) assay. The comparison of $IC_{50}$ values of Houttuynia cordata Thunberg extracts on $L1210,\;P388D_1$ and Vero cell lines showed that the methanol extract of Houttuynia cordata Thunberg indicated the most antitumor activity in the MTT assay. In order to develop a antimicrobial agent, dried Houttuynia cordata Thunberg was extracted with several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of amocla and ketoconazole as references was compared to those of other solvent extracts such as $H_2O$, n-hexane, chloroform, ethyl acetate ethanol and methanol. The antimicrobial activity of all extracts from the sample had growth inhibition activity against gram-negative bacteria, yam-positive bacteria and fungi $(MIC,\;>\;200\;{\mu} g/ml)$. These results suggest that the methanol soluble extract of Houttuynia cordata Thunberg may be a valuable choice for the studies on the treaeent of murine leukemia tumor cell lines and antimicrobial agents.
