Optical imaging of epileptic activity and epilepsy treatments in neocortex

  • Optical imaging offers excellent spatio-temporal sensitivity that is unparalleled by any other perfusion based imaging techniques. We used in vivo optical recording of intrinsic signals (ORIS) to map neurovascular hemodynamics of perfusion, oximetry and membrane potential during epileptic events in rat and mouse neocortex. Studies of hemodynamic changes with ORIS alone were also performed in human. Laboratory studies in rodent epilepsy models have demonstrated a persistent increase in deoxygenated hemoglobin (Hbr) and a decrease in tissue oxygenation during interictal spikes and ictal events. This "epileptic dip", like the "initial dip" recorded during normal sensory processing, implies that the enormous rise in cerebral blood flow (CBF) is inadequate to meet the increased metabolic demands associated with synchronized epileptic activity. These findings are critically important to the interpretation of the perfusion-based imaging studies, such as fMRI. In addition, we visualized the effect of direct cortical electrical stimulation, an alterative epilepsy treatment. The optical data following direct cortical electrical stimulation showed that hemodynamic signals are sensitive to different electrical stimulation parameters. Furthermore, our recent data demonstrated that the application of unilateral electrical stimulation is able to elicit bilateral hemodynamic responses in rat neocortex.

  • 소량으로 다품종을 생산하는 소규모 공장에서 적은 비용으로 쉽게 부품의 양 불량을 판별하는 시스템의 개발이 절실히 요구된다. 이러한 시스템의 개발을 위하여 다양한 환경에서도 물체를 인식할 수 있는 사람의 인식 능력과 같은 기능이 필요하다. 인간의 고등 지능은 대부분 두뇌의 신피질(neocortex)에서 비롯된다. 최근 이 신피질의 동작 원리를 컴퓨터에 접목시키는 연구 중의 하나가 Jeff Hopkins가 제안한 HTM 이론이다. 본 논문은 이 HTM 이론을 적용하여 소량 다품종 부품 생산 현장에서 부품의 이미지를 인식하여 양 불량을 판별하는 시스템인 PRESM(Parts image REcognition System for small scale Manufacturing industry)을 개발하였다. 개발 결과를 현장에 적용해 본 결과 실제 환경에서 부품의 양 불량 판별이 우수한 것으로 확인되었다.

A Recognition System for Multi-Form Korean Characters Based on Hierarchical Temporal Memory

  • Traditional character recognition systems usually aim at characters with simple variation. With the development of multimedia technology, printed characters may appear more diversely. Existing recognition technologies can't deal with Hangul recognition effectively in diverse environments. This paper presents a recognition system for multi-form Korean characters called RSMFK, which is based on the model of Hierarchical Temporal Memory (HTM). Our system can effectively recognize the printed Korean characters of different fonts, scales, rotation, noise and background. HTM is a model which simulates the neocortex of human brain to recognize and memorize intelligently. Experimental results show that RSMFK performs a good recognition rate of 97.8% on average, which is proved to be obviously improved over the conventional methods.

지능형 홈을 위한 HTM 기반의 얼굴 이미지 인식 시스템의 개발 (Development of An HTM Based Face Image Recognition System for Intelligent Home)

  • 본 논문에서는 Hopkins가 제안한 인간두뇌의 신피질(neocortex)의 동작 원리를 컴퓨터에 접목시킨 HTM 이론을 적용하여 가족 구성원들의 얼굴을 인식하는 지능형 홈을 위한 얼굴 이미지 인식 시스템인 FRESH(Face image REcognition System for intelligent Home)를 개발하였다. 개발 결과를 확인하기 위해 실제 사진을 촬영하여 깨끗한 이미지와 왜곡된 이미지에 대하여 실험해 본 결과 깨끗한 이미지뿐만 아니라 실제 환경에서 흔히 입력되는 다양하게 왜곡된 얼굴 이미지에 대한 인식의 정확도가 높았다. 또한, 기존의 연구들은 얼굴영역 인식에 초점이 맞추어져 있어서 지능형 홈을 위한 시스템에 적용하기가 어려우나 본 연구는 얼굴 이미지와 그 사람의 이름을 일치시킨 것으로 지능형 홈에 적합한 시스템으로 개발하였다.

  • 하나의 프레스로 여러 종류의 부품을 소량으로 생산하는 중 소형 공장에서는 부품 생산 과정에 발생하는 소리가 다양하게 나타난다. 이에 우리는 제품의 생산 순간의 소리를 인식하여 제품의 양 불량을 판별하는 시스템을 계층형 시간적 메모리(HTM Hierarchical Temporal Memory) 기술을 이용하여 개발하였다. HTM 이론은 인간 두뇌의 신피질(neocortex)의 동작 원리를 컴퓨터에 접목시킨 이론이다. 이는 실세계에 대한 시공간적인 패턴을 계층적으로 기억하는 것으로 기존의 인식 기술보다 여러 경우에 인식률이 뛰어난 것으로 알려져 있다. 우리는 이 HTM 기술을 소리 인식에 적용하여 부품에 대한 양 불량 판별 시스템을 개발하였다. 개발 결과를 검증하기 위해 실제 공장에서 부품 생산 순간의 다양한 소리들을 녹음하고, 소리 HTM 네트워크를 구성한 후, 학습과 훈련을 반복하여 해당 부품의 불량여부를 판정하도록 하였다. 그 결과 잡음이 많은 생산 현장에서도 판정의 정확도가 높은 것으로 확인하였다.

  • The present study was designed to investigate the appearance of the congenital aggregates of the ectodermal glial cells in the brain of the normal rodents. The brain samples were taken from mice fetus, juvenile mice, rats and rabbits. The appearance regions of the glial cell aggregates (GCA) were investigated and the cells in the GCA were identified with electron microscope. 1. GCA in the mouse fetus tended to be higher in cell density, larger in size and lower frequency in appearance than juvenile mouse. The regions of higher appearance frequency of GCA in the juveniles of mice, rats and rabbits were ordered as subependymal layer in the collateral trigone of lateral ventricles, molecular layer of the neocortex, inner layer except the molecular layer in the neocortex, cerebral medulla, corpus callosum and hippocampus. Appearance frequency of GCA in the neonatal mice tended to be higher until 5 day after birth, and were markedly decreased on 10 and 15 day after birth. 2. GCA tended to be closed on one side of the blood vessels or neurons but not perivascular or perineuronal appearance. 3. In electron microscophy, GCA were composed of immature oligodendrocytes and astrocytes in the subependymal, and tended to be more mature and loose in the neocortex and to be appended some microglia cells with age. The cells in the GCA of older mice tended to be more mature than in young mice.

RNA Binding Protein Rbms1 Enables Neuronal Differentiation and Radial Migration during Neocortical Development by Binding and Stabilizing the RNA Message for Efr3a

  • Various RNA-binding proteins (RBPs) are key components in RNA metabolism and contribute to several neurodevelopmental disorders. To date, only a few of such RBPs have been characterized for their roles in neocortex development. Here, we show that the RBP, Rbms1, is required for radial migration, polarization and differentiation of neuronal progenitors to neurons in the neocortex development. Rbms1 expression is highest in the early development in the developing cortex, with its expression gradually diminishing from embryonic day 13.5 (E13.5) to postnatal day 0 (P0). From in utero electroporation (IUE) experiments when Rbms1 levels are knocked down in neuronal progenitors, their transition from multipolar to bipolar state is delayed and this is accompanied by a delay in radial migration of these cells. Reduced Rbms1 levels in vivo also reduces differentiation as evidenced by a decrease in levels of several differentiation markers, meanwhile having no significant effects on proliferation and cell cycle rates of these cells. As an RNA binding protein, we profiled the RNA binders of Rbms1 by a cross-linked-RIP sequencing assay, followed by quantitative real-time polymerase chain reaction verification and showed that Rbms1 binds and stabilizes the mRNA for Efr3a, a signaling adapter protein. We also demonstrate that ectopic Efr3a can recover the cells from the migration defects due to loss of Rbms1, both in vivo and in vitro migration assays with cultured cells. These imply that one of the functions of Rbms1 involves the stabilization of Efr3a RNA message, required for migration and maturation of neuronal progenitors in radial migration in the developing neocortex.

부품 이미지 인식을 위한 HTM 네트워크 훈련 시스템 개발 (Development of an HTM Network Training System for Recognition of Molding Parts)

  • 여러 품종을 소량으로 생산하는 소형 공장에서 불량품으로 인한 손실을 줄이기 위하여 부품의 양불량을 판단하는 시스템의 개발이 필요하다. 그러한 시스템은 계층형 시간적 메모리(HTM : Hierarchical Temporal Memory) 기술을 이용하여 개발할 수 있다. HTM은 인간 두뇌의 신피질(neocortex)의 동작 원리를 기계학습에 접목시킨 모델이다. HTM 기반의 기계학습 시스템을 사용하기 위해서는 훈련된 HTM 네트워크를 개발해야 하는데, 이를 위해서는 HTM 이론에 대한 지식이 필요하다. 본 연구는 이 HTM 기술을 부품의 이미지 인식에 적용하여 부품에 대한 양 불량을 판별하는 시스템에서, HTM 네트워크 개발을 지원하는 훈련시스템의 설계와 구현을 제시한다. 이 시스템은 HTM 이론에 대한 지식이 없어도 작업현장의 기술자가 HTM 네트워크를 정확히 훈련시킬 수 있으며, 부품에 대한 모든 종류의 HTM 기반의 판정시스템에 그대로 적용될 수 있다.

Analysis of allele-specific expression using RNA-seq of the Korean native pig and Landrace reciprocal cross

  • Objective: We tried to analyze allele-specific expression in the pig neocortex using bioinformatic analysis of high-throughput sequencing results from the parental genomes and offspring transcriptomes from reciprocal crosses between Korean Native and Landrace pigs. Methods: We carried out sequencing of parental genomes and offspring transcriptomes using next generation sequencing. We subsequently carried out genome scale identification of single nucleotide polymorphisms (SNPs) in two different ways using either individual genome mapping or joint genome mapping of the same breed parents that were used for the reciprocal crosses. Using parent-specific SNPs, allele-specifically expressed genes were analyzed. Results: Because of the low genome coverage (${\sim}4{\times}$) of the sequencing results, most SNPs were non-informative for parental lineage determination of the expressed alleles in the offspring and were thus excluded from our analysis. Consequently, 436 SNPs covering 336 genes were applicable to measure the imbalanced expression of paternal alleles in the offspring. By calculating the read ratios of parental alleles in the offspring, we identified seven genes showing allele-biased expression (p<0.05) including three previously reported and four newly identified genes in this study. Conclusion: The newly identified allele-specifically expressing genes in the neocortex of pigs should contribute to improving our knowledge on genomic imprinting in pigs. To our knowledge, this is the first study of allelic imbalance using high throughput analysis of both parental genomes and offspring transcriptomes of the reciprocal cross in outbred animals. Our study also showed the effect of the number of informative animals on the genome level investigation of allele-specific expression using RNA-seq analysis in livestock species.

치매에 관한 최근의 연구 동향 (The latest development in Dementia)

  • Dementia is the neurodegenerative process that affects cognition, behavior and function and one of the most prominent diseases of dementia is Alzheimer's disease(AD). AD is a dementing illness characterized clinically by the progressive and irreversible deafferentation of the limbic system, association neocortex and basal forebrain. A number of conditions are known to be predisposing risk factors for AD. In several of these, initiation of glial-mediated inflammatory pathways as a mechanism of AD is getting a lot of attention. On the other hand, a biochemical marker for monitoring the onset and progression of the disease would be a valuable tool for disease management. Also such a marker might be used as an end point in clinical intervention protocols. This biochemical marker will have the potential for identifying subjects afflicted with the disease and possibly for monitoring the onset and longitudinal progression of the disease. Here we have reviewed the latest papers of different approaches to AD. Of course, there is a section of PET which is very useful clinically nowadays.
