삼투압 스트레스를 받은 Yersinia enterocolitica의 성장에 미치는 glycine betaine을 비롯한 osmolyte의 영향 (Effects of Glycine Betaine and Related Osmolytes on Growth of Osmotically Stressed Yersinia enterocolitica)

  • 고염분으로 삼투압 스트레스를 받은 Yersinia enterocolitica ATCC 9610 세포내에 축적되는 osmolyte를 $^{13}C$ NMR을 통해 조사하였다. 자연계에 흔히 존재하는 가장 강력한 osmolyte로 알려진 glycine betaine의 삼투압 스트레스를 받은 Y. enterocolitica ATCC 9610 세포내 농도 (801.9 nmol/mg protein)는 삼투압 스트레스를 받지 않은 세포 (19.2 nmol/mg protein) 보다 41.8배 많이 검출되어 Y. enterocolitica ATCC 9610의 가장 주요한 osmolyte로 작용하였다. Proline도 284.8 nmol/mg protein의 농도로 검출되어 Y. enterocolitica ATCC 9610의 osmolyte로 작용하였다. Glycine betaine을 비롯한 각종 osmolyte가 삼투압 스트레스를 받은 Y. enterocolitica 세포성장에 미치는 역할을 검증하기 위해 Y. enterocolitica의 성장속도에 미치는 이들의 영향을 조사하였다. 2.5%의 NaCl이 첨가된 MMA 배지에 glycine betaine과 proline을 각각 첨가했을때, 1mM glycine betaine의 경우 osmolyte를 첨가하지 않은 대조구에 비해 성장속도가 3.6배 증가하였으며 5mM proline의 경우는 1.3배 증가하였다. Dimethylglycine도 2.5%의 NaCl을 첨가한 MMA 배지에 5mM의 농도로 첨가하였을때 대조구에 비해 성장속도가 3.1배 증가하였는데, monomethylglycine은 삼투압 스트레스를 받은 Y. enterocolitica의 성장에 어떤 영향도 미치지 못하였다. Carnitine은 2.5%의 NaCl이 첨가된 MMA 배지에 5mM의 농도로 첨가되었을때 대조구에 비해 2.4배 성장속도가 증가하였으며 choline은 성장에 어떤 영향도 미치지 못하였다. 이상의 결과로 glycine betaine은 Y. enterocolitica ATCC 9610의 가장 주요한 osmolyte로 작용하며, proline, dimethyglyine 그리고 carnitine도 Y. enterocolitica ATCC 9610의 osmolyte로 작용하여 삼투압 스트레스를 받은 Y. enterocolitica ATCC 9610의 성장속도를 증가시켰다.

Features and Functions of Purple Pigment Compound in Halophytic Plant Suaeda japonica : Antioxidant/Anticancer Activities and Osmolyte Function in Halotolerance

  • Suaeda japonica is a halophytic plant that lives in mudflat at intertidal zone of western and southern coastal areas of Korea. The seawater-living plants showed a purple color during their whole life. In contrast, freshwater-living plants displayed a green color in leaves. When seawater-living plants were transferred to potting soil, the purple color was gradually changed to green in the leaves. The extracted purple pigment compound exhibited typical characteristics of betacyanin that were represented by water solubility, pH- and temperature-dependent color changes, sensitivity to light, UV-Vis spectra, and gel electrophoretic migration pattern. The LC-MS analysis of the extracted pigment compound showed the presence of two major protonated molecular ions ($[M+H]^+$) at m/z 651.1 and m/z 827.1. Antioxidant activity of the pigment compound was determined using stable free radical DPPH assay. It was found to have an antioxidant activity that is linearly increased in proportion to the reaction time for up to 30 min, and the activity was comparable to that of control BHA at 9.0 mg/ml. The anticancer activity against several tumor cell lines was also examined following the MTT assay. The significant growth inhibitory effect was observed on two tumor cell lines, SW-156 (human kidney carcinoma) and HEC-1B (human endometrial adenocarcinoma). Probably, the pigment compound may function as an osmolyte to uphold halotolerant physiological processes in saline environment.

Physiological responses to salt stress by native and introduced red algae in New Zealand

  • Intertidal macroalgae are regularly exposed to hypo- or hypersaline conditions which are stressful. However, red algae in New Zealand are generally poorly studied in terms of salinity tolerance. Consequently, two native (Bostrychia arbuscula W. H. Harvey [Ceramiales], Champia novae-zelandiae [J. D. Hooker & Harvey] Harvey [Rhodymeniales]) and one introduced red algal taxon (Schizymenia spp. J. Agardh [Nemastomatales]) were exposed for 5 days in a controlled salt stress experiment to investigate photosynthetic activity and osmotic acclimation. The photosynthetic activity of B. arbuscula was not affected by salinity, as reflected in an almost unchanged maximum quantum yield (Fv/Fm). In contrast, the Fv/Fm of C. novae-zelandiae and Schizymenia spp. strongly decreased under hypo- and hypersaline conditions. Treatment with different salinities led to an increase of the total organic osmolyte concentrations with rising salt stress in B. arbuscula and Schizymenia spp. In C. novae-zelandiae the highest organic osmolyte concentrations were recorded at SA 38, followed by declining amounts with further hypersaline exposure. In B. arbuscula, sorbitol was the main organic osmolyte, while the other taxa contained floridoside. The data presented indicate that all three red algal species conspicuously differ in their salt tolerance. The upper intertidal B. arbuscula exhibited a wide salinity tolerance as reflected by unaffected photosynthetic parameters and strong sorbitol accumulation under increasing salinities, and hence can be characterized as euryhaline. In contrast, the introduced Schizymenia spp. and native C. novae-zelandiae, which preferentially occur in the mid-intertidal, showed a narrower salinity tolerance. The species-specific responses reflect their respective vertical positions in the intertidal zone.

Glycine betaine 엽면 처리가 토마토 유묘의 생육과 삼투조절물질 함량에 미치는 영향 (Effects of Foliar Application of Glycine Betaine on the Growth and Contents of Osmolyte in Tomato Seedling)

  • 토마토 유묘의 생육에 미치는 glycine betaine의 엽면처리 효과를 분석한 결과, 초장이나 건물중 등의 생육이 대조구인 증류수 처리에 비해 촉진되는 경향을 보였으며 25mM glycine betaine 처리가 가장 좋았다. 저온스트레스 하에서 삼투조절 역할을 하는 가용성 당과 proline의 축적량이 증류수 처리에 비해 glycine betaine처리에서 낮은 경향을 보여 외부에서 엽면 처리한 glycine betaine이 삼투조절 역할을 한 것으로 판단되었으며 이러한 결과는 수용성 단백질과 유리 아미노산의 축적 양상에서도 잘 반영되었다. 이상의 결과로 보아 glycine betaine의 엽면 처리는 저온기 시설 토마토 재배 시 야간 저온장해를 극복할 수 있는 방법으로서 이용 가능성이 있을 것으로 사료된다.

산란계 사료에 비태인의 수준별 급여가 하절기 생산성 및 계란의 품질에 미치는 영향

  • Betaine functions as an osmoregulators in the cells and its inclusion can spare the choline, carcass fat reduction. Thus, two hundred eighty eight of seventy eight weeks old laying hens were fed with 0, 500, 1,000, 2,000 ppm betaine addition during the environmentally high temperature stress. Basal diets contained 16% CP and 2,800 ME. Egg production, feed intake, feed conversion were examined for eight weeks. Egg qualities, liver betaine, blood osmolarity, AntiDiuretic Hormone(ADH) were measured at the end experiment. Egg production of hens fed 500, 2,000 ppm dietary betaine and showed significance between control and 2,000 ppm betaine treatment(P<0.05). ADH of blood sera tended to increase as dietary supplemental betaine increased. The results of this experiments indicated that dietary supplemental betaine was able to improve the performance, eggshell breaking strength, liver betaine in this experiment.

A chemosystematic investigation of selected Stichococcus-like organisms (Trebouxiophyta)

  • The taxonomy of green microalgae relies traditionally on morphological traits but has been rapidly changing since the advent of molecular methods. Stichococcus Nägeli is a cosmopolitan terrestrial algal genus of the class Trebouxiophyceae that has recently been split into seven lineages, which, along with Pseudostichococcus, comprise the Stichococcuslike group; there is a need to further characterize these genera, since they are morphologically enigmatic. Here we used organic osmolytes as chemotaxonomic marker to verify the phylogenetic position of Stichococcus-like strains and were also able to exclude a strain hitherto identified as Gloeotila contorta from this group. Stichococcus-like organisms, including those recently revised, were characterized by the production of the polyol sorbitol and the disaccharide sucrose in high amounts, as is typical of Prasiola-clade algae. The results demonstrate that organic osmolyte chemotaxonomy can support green algal taxonomic designations as fundamental research.

비병원성 Erwinia carotovora subsp. carotovora 9-3의 건조 및 저장의 과정에서 생존에 미치는 염 효과 (High Concentration of Sodium Chloride Increases on Survival of Non-pathogenic Erwinia carotovora subsp. carotovora 9-3 during Drying and Storage)

  • 길항력이 있는 그람음성균들의 생물적 방제제로 성공적인 개발은 균의 건조와 저장시의 낮은 생존을 때문에 건조제형으로 만드는 것은 제한되어있다. 그람음성균의 배양시 높은 염(NaCl)의 첨가는 삼투압 조절제인 osmolyte의 생성을 유도하여 건조 시 생존율을 높인다고 알려져 있다. 그람음성의 길항력을 내는 비병원성 Erwinia carotovora subsp. carotovora(Ecc) 9-3의 건조와 저장시의 염의 영향에 관해 연구하였다. Ecc 9-3의 생장률은 0.5 M의 염이 첨가되어 있는 배지에서 큰 차이가 없었지만 0.7 M의 염이 첨가된 배지에서는 급격하게 낮아진 것을 확인하였다. 건조와 저장시의 생존율은 0.5 M의 염을 넣은 배지가 염을 넣지 않은 배지보다 상온건조와 동결건조에서 각각 2배 그리고 3배의 높은 생존율을 얻었다. 건조 후 저장에서도 역시 염이 높을수록 Ecc 9-3의 생존율이 더 높았다. 그리고 상온건조와 동결건조 모두 $4^{\circ}C$에 저장하였을 때 생존율에 큰 차이가 없었다. 그러나 생존율은 $27^{\circ}C$, $37^{\circ}C$에 저장하였을 때 염의 농도가 높을수록 생존율이 증가하였다. 첨가제 실험에서는 lactose가 동결건조와 상온건조 모두에 영향을 주었고 dextrin은 상온건조에 큰 영향을 주었다.

해안식물의 유리아미노산과 질소함량 (Free Amino Acid and Nitrogen Contents of the Coastal Plants in Korea)

  • 32종의 해안식물(주로 염생식물)에 대한 유리아미노산 및 총질소함량을 조사하였다. 몇몇 종(참골무꽃과 해란초)을 제외한 모든 식물체는 Pro을 함유하였으나, 번행초, 등골나물아재비 및 갯무에서만, 높은 함량을 나타내어, 이들 식물에서 Pro은 세포질성 삼투물질로 작용하는 것으로 생각된다. 그러나 등대풀, 갯무, 갯방풍, 갯기름나물, 통보리사초, 왕잔디, 갯잔디 등의 식물은 Pro 보다는 hydroxylproline (OH-Pro)을 상당량 축적하였다. 총 아미노산은 갯개미취, 갯질경, 해란초, 순비기나무 및 특히 명아주과와 돌나물과 식물에서 낮은 값을 보였다. 또한 번행초, 등대풀, 갯무, 명아주과 및 콩과 식물은 보편적으로 총 질소 및 수용성 질소함량이 높았으나, 참골무꽃, 해란초, 순비기나무, 통보리사초 및 벼과 식물은 낮은 수준의 질소함량을 보였다. 대부분의 해안식물에서 유리아미노산은 총질소함량에 차지하는 비율이 낮았지만, 등대풀, 모래지치 및 둥근바위솔의 경우 아미노산 질소가 총질소함량의 25∼30%를 차지하였다. 결론적으로, 몇몇 예외는 있지만 세포질성 삼투물질로 널리 알려진 Pro은 조사된 해안식물의 유리아미노산 풀(pool)에서 유의한 몫을 차지하지 않는 것으로 여겨진다.

Metabolic, Osmoregulatory and Nutritional Functions of Betaine in Monogastric Animals

  • This review focuses on the metabolic and osmoregulatory functions of betaine and its impact on nutrient digestibility and performance in pigs and poultry. Betaine is the trimethyl derivative of the amino acid glycine, and is present in plant and animal tissue. It has been shown to play an important role in osmoregulation of plants, bacteria and marine organisms. Due to its chemical structure, betaine exerts a number of functions both at the gastrointestinal and metabolic level. As a methyl group donor, betaine is involved in transmethylation reactions and donates its labile methyl group for the synthesis of several metabolically active substances such as creatine and carnitine. Therefore, supplementation of betaine may reduce the requirement for other methyl group donors such as methionine and choline. Beneficial effects on intestinal cells and intestinal microbes have been reported following betaine supplementation to diets for pigs and poultry, which have been attributed to the osmotic properties of betaine. Furthermore, betaine potentially enhances the digestibility of specific nutrients, in particular fiber and minerals. Moreover, at the metabolic level, betaine is involved in protein and energy metabolism. Growth trials revealed positive effects of supplemental betaine on growth performance in pigs and poultry, and there is evidence that betaine acts as a carcass modifier by reducing the carcass fat content. In conclusion, due to its various metabolic and osmoregulatory functions, betaine plays an important role in the nutrition of monogastric animals.

Can Exogenous Betaine Be an Effective Osmolyte in Broiler Chicks under Water Salinity Stress?

  • A CRD experiment was conducted to evaluate the effects of different exogenous betaine levels (0.000, 0.075, 0.150 and 0.225 percent) on 576 one-day-old male broiler chicks (Ross) under water salinity stress. Different levels of water salinity were made by adding 3 levels of NaCl (0, 1,000 and 2,000 mg/L) to drinking water. Feed and water were available ad libitum. Betaine increased body weight, improved feed conversion ratio, and decreased packed cell volume (p<0.05). Water salinity promoted body weight over the whole period, increased feed intake (11 to 21 and 29 to 42-d) and also improved feed conversion ratio in grower and finisher periods (p<0.01). Breast weight, water consumption (28-d and 42-d) and excreta moisture (28-d) were increased by elevating the level of water salinity (p<0.01). Interaction between dietary betaine and water salinity was significant on plasma osmolarity as well as epithelial osmolarity of the duodenum at 28-d. Epithelial osmolarity was decreased from duodenum to ileum. The data imply that betaine is involved in the protection of intestinal epithelia against osmotic disturbance which can be caused by saline water, but further research is needed to investigate the effects of betaine with higher levels of water salinity.
