Low-ester Pectin Gel의 단단함과 강도(强度)의 예측 (Prediction of Firmness and Strength of Low-ester Pectin Gel from Chemical Composition)

  • High-ester pectin을 HCI 단독 또는 HCI 과 $NH_4OH$를 병용하여 demethylation 시킨뒤 얻어진 low-ester pectin(LM pectin)으로 gel을 제조하였다. LM pectin gel의 단단함과 강도(强度)를 측정한 결과 HCI로 처리된 LM pectin gel은 일반적으로 강도(强度)가 약하고 거칠며 탄력이 없었다. 반면 $HCl-NH_4OH$로 처리한 LM pectin gel은 텍스쳐가 부드럽고 탄력이 좋았다. 물리적 성질에 대한 LM pectin의 화학적 조성의 상관 관계에서 단단함(Sag값)에 대한 ester함량 및 분자량, 그리고 강도(强度)(puncture stress)에 대한 amide 및 ester함량 EH는 분자량 간에는 유의성 있는 관계가 밝혀졌다. 상관성이 높은 화학적 인자에서 LM pectin gel의 물리적 성질을 예측할 수 있는 관계식을 산출하였다.

Aspergillus japonicus에서 추출한 Pectinesterase를 이용한 Low Methoxyl Pectin Gel 및 LMP 사과 Jelly 제조에 관한 연구 (The Preparation of Low Methoxyl Pectin Gel and LMP Apple Jelly with Pectinesterase Isolated from Aspergillus Japonicus)

  • Pectinesterase was isolated from the culture medium of Aspergillus japonicus and partially purified by DEAE-Sephadex batchwise, Sephadex G-75 gel filtration and ion exchange chromatography. The purified enzyme solution was completely free from polygalacturonase which depolymerizes pectin molecule. The ability of the pectinesterase to demethylate high methoxyl pectin was investigated. On 20 minute of incubation methoxyl content of low methoxyl pectin decreased from 88% to 6.93%. In general gel prepared with the pectin containing lower methoxyl content showed the lower value of percent sag, and showed the hieher Bel strength. Textural characteristics of pectin gel determined by Rheometer showed that as the methoxyl content was lowered, hardness and resilience of the gel were increased and cohesiveness was decreased. Apple juice containing HMP and organic acids can be converted into low methoxyl pectin apple jelly by the action of pectinesterase and addition of calcium ion. The strength of low methoxyl pectin apple jelly increased when it stored at room temperature.

Alginate와 pectin 첨가에 의한 콩묵의 텍스쳐 특성 (Textural Properties of Soygel with Added Alginate and Pectin)

  • Alginate, pectin과 Ca염이 콩묵의 텍스쳐 특성에 미치는 영향을 조사하였다. 콩묵은 전지대두분(300 mesh) 분산액에 sodium alginate나 pectin 및 Ca염을 첨가한 다음 혼합하여 제조하였다. Sodium alginate나 pectin을 전지대두분 분산액에 첨가했을 때 견고성, 부착성, 응집성은 유의적으로 증가하였다. 가장 균일한 콩묵은 전지 대두분의 $12{\sim}15%$의 sodium alginate 또는 pectin을 첨가했을 때 얻어진 반면 sodium alginate와 pectin의 혼합 첨가는 텍스쳐 특성을 감소시켰다. 순위법에 의한 콩묵의 관능적 특성은 12% sodium alginate를 첨가하고 전지대두분 1g당 0.125g의 $CaSO_4$로 제조한 콩묵이 탄력성, 균일성, 부드러움성, 기호도면에서 가장 좋은 것으로 나타났다. 따라서 콩묵의 최적 제조조건은 전지대두분의 10배의 수분을 첨가한 분산액에 대두분의 12% sodium alginate를 혼합한 후 대두분의 12.5% $CaSO_{4}$ 또는 Ca $gluconate-CaSO_4$(50 : 50)를 첨가하여 $4^{\circ}C$에서 냉각시킨 것이다.

  • 본 연구에서는 gel화를 유도할 수 있는 첨가제를 첨가하여 만든 5가지 옥수수 전분 gels와 아무것도 첨가하지 않은 control옥수수 전분 gel의 관능적 물성적 특성을 조사 비교하였다. 1. 관능검사를 실시한 결과 \circled1 Agar의 hardness는 control보다 유의하게 높았고, pectin은 control 보다 유리하게 낮았다. \circled2 CMC와 pectin의 adhesiveness는 control 보다 유의하게 높았고 agar는 control보다 유의하게 낮았다. \circled3 전체적인 acceptability는 CMC와 Pectin은 control보다 유의하게 선호도가 낮았고 나머지 시료들은 control과 유의차가 없었다. 2. Instron을 이용한 물성검사 결과 \circled1 Agar의 hardness는 control보다 유의하게 높았고, 나머지 시료들의 hardness는 control보다 유의적으로 낮았다. \circled2 Agar와 casein의 cohesiveness는 control보다 유의하게 높았으며 gelatin은 control보다 유의하게 낮았다. 3. 관능검사나 Instron에 의한 기계적 검사에 있어서 시료의 차이를 가장 잘 구분해 주는 특성치는 hardness라고 생각되어진다

Pectin from Passion Fruit Fiber and Its Modification by Pectinmethylesterase

  • Passion fruit fiber pectin gels represent a new alternative pectin source with potential for food and non-food applications on a commercial scale. Pectic polysaccharides were extracted from passion fruit (Passiflora edulis) fiber using citric acid as a clean catalyst and autoclaved for 20 to 60 min at $121^{\circ}C$. The best condition of pectin yield with the highest molecular weight was obtained with 1.0% of citric acid (250 mg/g dry passion fruit fiber pectin) for 20 min of autoclaving. Spectroscopic analyses by Fourier transform infrared, enzymatic degradation reactions, and ion-exchange chromatography assays showed that passion fruit pectin extracted for 20 min was homogeneous high methoxylated pectin (70%). Gel permeation analysis confirmed that the pectin extract obtained by autoclaving by 20 min showed higher molecular weights than those autoclaved for 40 and 60 min. Passion fruit pectin extracted for 20 min was enzymatically modified with fungal pectinmethylesterase to create restructured gels. Short autoclave treatment (20 min) with citric acid as extractant resulted in a significant increase of gel strength, improving pectin extraction in terms of functionality. The treatment of solubilized material (pectic polysaccharides) in the presence of insoluble material (cellulose and hemicellulose) with pectinmethylesterase and calcium led to the creation of a stiffer passion fruit fiber pectin gel, while syneresis was not observed.

과일 내의 pectin 함량 및 성숙도에 따른 pectin 특성 변화에 관한 연구 (Chemical and Gelation characteristics of Pectins Obtained from Ripened and Unripened Crabapples)

  • This study was attempted to determine the pectin contents of various fruits(Cherry, strawberry, plum, peach, apricot, apple, crabapple) and to investigate the changes in the properties of pectin and pectin gel during crabapple ripening. Methoxyl content, molecular weight and intrinsic viscosity were measured in ripened and unripened crabapples. In order to determine the characteristics of pectin gel, percent sag was measured by exchange Ridgelimeter, hardness and textural characteristics of jellies were measured by a penetrometer and a texturometer. The experimental results are summarized as follows: 1. The pectin contents of various fruits were 1.16% for cherry, 3.17% fir strawberry, 4.29% for plum, 5.51% for apricot, and pectin contents of peaches and apples varies depends on varieties from 4.54% to 5.61% to 6.52, respectively. The pectin contents of unripened and ripened crabapple wee 9.21% and 9.84% respectively, indicating that pectin content increased with fruit maturity. 2. The methoxyl contents, which was 6.61%~6.95%, was very low and it increased significantly during fruit ripening. Further, a decrease in acetyl content, intrinsic viscosity, molecular weight occurred during the ripening of crabapples. 3. The percent sag of crabapple jellies increased with fruit maturity from 5.26% to 5.79%, and the corresponding jelly grade decreased in ripened crabapple. 4. The hardness of jellies determined by a penetrometer was 79.15 pp.U. for ripened crabapple jelly, 90.62 pp.U. for unripe ned crabapple jelly. 5. In the textural characteristics of gels measured by a texturometer, the gels made from unripened crabapple were shown to have the highest values in hardness, cohesiveness, adhesiveness and gumminess.

Ganoderma lucidum이 생산하는 Polygalacturonase의 정제 및 특성 (Purification and Properties of Polygalacturonase from Ganoderma lucidum)

  • Ganoderma lucidum이 생산하는 polygalacturonase의 유용활용 방안을 위한 효소의 특성을 연구하기 위하여 버섯배양물로부터 효소를 생산하고 정제하여 그 특성을 검토한 결과는 다음과 같다. Endo-polygalacturonase는 배양여액으로부터 ammonium sulfate 침전, Biogel P-100, DEAE-cellulose, Sehpadex G-150 column chromatography에 의하여 순차적으로 정제되었고, Sehpadex G-150 column chromatography에서 re-gel filtration까지 실시한 결과 specific activity가 892unit/mg protein로 배양여액보다도 약 56배의 정제도를 나타냈으며, exo-polygalacturonase는 ammonium sulfate침전, Biogel P-100, DEAE-cellulose column chromatography까지 부분정제한 결과 9.2배의 정제도를 나타냈다. SDS polyacrylamide gel에서 전기영동을 실시한 결과 단일 band를 나타냈으며, 분자량은 54,000 dalton이었다. Endo-polygalacturonase와 exo-polygalacturonase는 citrus pectin이나 pectic acid보다 apple pectin에 친화도가 높았으며, Lineweaver-Burk plot로부터 계산된 apple pectin에 대한 Km 치는 각각 1.44와 10.6 mg/ml이었다. 이 두 효소의 작용 최적 pH는 4.0이었으나 pH 안정성에서는 endo-polygalacturonase는 $pH\;4.0{\sim}6.0$에서, exo-polygalacturonase는 $pH\;3.5{\sim}5.5$ 범위에서 안정하였다. 효소반응 최적 온도는 endo-polygalacturonase는 $40^{\circ}C$ 이었으나 exo-polygalacturonase는 $60^{\circ}C$에서 최대의 활성을 나타냈고 열안정성도 exo-polygalacturonase가 endo-polygalacturonase보다 더 높은 온도에서도 안정하였다. 또한 endo-polygalacturonase는 $Ca^{++}$$Mn^{++}\;ion$에, exo-polygalacturonase에는 $Ca^{++}\;ion$에 의해 효소반응이 촉진되었다.

Structural and gelling properties of very low methoxyl pectin produced by an alkali-treatment

  • Very low methoxyl pectin (VLMP) has different physical and rheological properties compared to high and low methoxyl pectins (HMP and LMP). In this study, we produced LMP and VLMP by alkaline de-esterification, and investigated the structural and textural properties. Apple peel pectin was kept at pH 12 using 5.0 M NaOH solution for 3 and 24 h to produce LMP and VLMP, respectively. The molecular weight was decreased due to the removal of an esterified group in the pectin backbones by the alkali treatment, and the VLMP showed a higher calcium ion sensitivity which leads to the production of the gel with increased hardness. The result clearly showed that VLMP has the potential to improve the texture and stability in food products depending on their degree of esterification, and this result can be applied as a functional ingredient in food industrial area application to enhance the current commercial pectins.

미류나무 분화조직중 세포벽다당류의 화학적 성상 (Chemical Characteristics of Cell-Wall Polysaccharides in Differentiating Xylem of Populus deltoides M.)

  • The chemical composition of differentiating xylem of Populus deltoides M. were investigated and compared with those from sapwood. The cell wall polysaccharides were extracted sequentially from a differentiating xylem and sugar composition was analyzed with G.L.C, H.P.L.C and gel chromatograpy. The pectin substance and hemicellulose are rich in the cell wall of differentiating xylem. The $H_2O$ extract polysaccharides from differentiating xylem were composed with xylose-glucose residues which seem to be xyloglucan and a pectin. The arabinogalactan and the mannan were extracted with $Na_2CO_3$ solution and also the xylan was extracted with KOH solution. Sugar composition of each fractions in gel filteration of purified $H_2O$ polysaccharide suggests that the xyloglucan can be extracted with $H_2O$ from differentiating xylem.

친수성 정제의 겔층두께와 겔팽창 영역의 조직 특성화 (Textural Characterization of Gel Layer Thickness and Swelling Boundary in a Hydrophilic Compact)

  • This study was to investigate the relationship between the gel layer thickness and swelling boundary via strength measurements using texture analysis. The novel texture analysis approach was used to examine the dynamics of swelling behavior in a ternary polymeric matrix tablet. The method permitted the characterization of the changes occurring at the peripheral as well as within interior boundary of the swelling during water ingress. The increase in gel strength for pectin, HPMC, and a ternary mixture with gelatin was found to depend on polymer concentration. Therefore, this method is further applicable to characterize the swelling behavior and provide opportunity to differentiate the gel-layer from that of swelling boundary.
