Intraspecific Diversity of Korean Takydromus wolteri(Reptilia: Squamata) Based on Randomly Amplified Polymorphic DNA (RAPD) Analysis (RAPD를 이용한 한국산 줄장지뱀(Reptilia: Squamata)의 종내 다양성에 관한 연구)

  • The lacertid lizard Takydromus wolteri widely distributed in South Korea, but intraspecific diversity of this species was not almost studied. T. wolteri represented by 8 specimens collected from 5 localities were analyzed by the Randomly Amplified Polymorphic DNA (RAPD) method with 28 random decamer primers and the phenogram constructed by using the Unweighted Pair Group Method with Arithmetic Mean (UPGMA) method based on RAPD data. The populations of T. wolteri using in this paper were collected from Gyeonggi-do, Chungcheongbuk-do, Jeju-do, Jeollanam-do and Gyeongsangnam -do in South Korea. Among 68 different bands detected in RAPD analysis, 59 bands (87%) showed polymorphism. The phenogram showed that the populations of T. wolteri were grouped into two. These results suggest that T. wolteri were supported intraspecific diversity.

  • A total of 10 main characters of 16 species of family Rhodospirllaceae were phenetically and cladistically analyzed by Farris' method. The obtained phenogram and cladistic tree were compared with Bergey's manual and other papers. The results supported that the system of 5 subgroups (genera) is available in family Rhodospirllaceae and indicated that close affinities between Rhodospirllum tenue and Rhodopseudomonas gelatinosa and between Rhodomicrobium vannielii and other species of genus Rhodopseudomonas were proved.

Analysis of Relationship of the Korean Styracaceae by RAPD Markers (RAPD markers에 의한 한국산 때죽나무과(Styracaceae)식물의 유연관계)

  • In order to estimate the relationships among four taxa (Styrax japonicus, Styrax obassia, Styrax shiraianus and Pterostyrax hispidus) of two genus in the Styracaceae and their regional populations, RAPDs analysis was performed. Fifteen random primers generating 238 RAPD bands were enough to reveal differences in genotype bands. They were clustered into three groups by the UPGMA phenogram. The first group was Styrax japonicus populations, the second group was consisted of Styrax obassia populations and S. shiraianus populations, and third group was Pterostyrax hispidus population. The UPGMA phenogram based on RAPD analyses showed that Styrax obassia and S. shiraianus had the closest relationship among four taxa. Species as well as genera of the Korean Styracaceae were well discriminated by the RAPD analyses.

RAPD Analyses on the Regional Populations of Two Species of Saururaceae in Korea (삼백초와 식물 2종의 지역개체군별 RAPD 분석)

  • In order to presume the genetic relationship about two species and their 30 regional populations of the Saururus and Houttuynia of Saururaceae, RAPD analyses were performed. The length of amplified DNA fragments ranged from 300 to 2,000 bp. 156 scorable RAPD makers were found from PCR reactions with sixteen random oligoprimers. Also, some regional populations were clustered separately from the UPGMA phenogram. The OTUs between cultivated and natural populations were distinguished distinctly on the UPGMA phenogram. And the regionals populations of the treated taxa were clustered. Among the regional populations of two species, GN populations had the close relationship JJ populations rather than JN populations. The RAPD data was very useful to define the genetic relationship and distinguish the cultivated populations from natural populations by regional distributions in this study.

  • Morphological characteristics of Kalopanox pictus Nakai were studied to examine population differentiation of this species. Based on a phenogram of using 23 morphological characteristics, differentiation of regions were distinct. Collections of 138 specimens from nine populations served as operational taxonomic unit (OTU's) were examined for phenotypic similarity and morphological variation using clustering (Ward's minimum variance method) and principal component analysis (PCA). The first three principal components were responsible for 77.0% of the total variance. Principal component 1 explained 52% of the total variance and was contributed to by the number of palmately parted, the number of pinnately lobed, and width between two lateral lobe apex.

Phenetic Variability in Leaf Morphological Characteristics of the Korean Rubus crataegifolius Populations (한국 산딸기 집단에서 잎 형태 특성에 나타난 표현형 변이)

  • Rubus crataegifolius is a shrub species and is primarily distributed throughout Asia and Europe. We collected 550 representative samples of the eleven populations in Korea to estimate leaf variation within species. The 35 morphological characteristics allowed us to resolve well-supported fixed characteristics and variable characteristics. Sixteen of twenty-three quantitative characteristics and five of twelve qualitative characteristics showed significant differences among populations. We argued that the population differentiation can accounted for the variation in phenetic characteristics such as spine in this species, but were less successful in accounting for variation in other traits. Within a polystatistic leaf structure, their morphological differences could be modulated by ecological pressure such as temperature, altitude, duration of sunshine, and solar radiation. The phenogram showed two distinct clades. One was a group in central Korea and the other was a group in the southern regions of Korea. If morphological characteristics in R. crataegifolius populations reflect their ecological niche, morphology should be regarded with caution as the main criterion for population studies in R. crataegifolius.

Genetic Diversity of Korean Cyanobacteria determined by DNA polymorphisms within the Phycocyanin Locus (Phycocyanin locus내의 DNA Polymorphism에 의한 한국산 Cyanobacteria의 유전적 다양성)

  • The genetic diversity among Korean cyanobacteria was assessed by restriction fragment length polymorphism(RFLP) analysis of PCR products from the phycocyanin locus. Strains of all the genera tested were successfully amplified, and the size of amplified fragments was approximately 700bp. The restriction patterns generated by AluI, MspI, and HaeIII were conserved for strains within each of genera studied and were specific to the genus level. Intrageneric delineation of strains was revealed by the enzyme, CfoI for members of genera Anabeana and Synechocystis. Phenogram derived from the different RFLP patterns revealed a coherent cluster among Anabeana, Chlorogloea, and Synechocystis strains. PC-RFLP methods provided useful tools for classification of cyanobacteria.

Genetic relationship of Aloe vera 'Saengjang', a new forma, based on cpDNA and ITS sequence variation (cpDNA와 ITS 염기변이에 근거한 신품종 생장알로에 유전적 상관관계)

  • This study was carried out to understand the genetic relationship of three Aloe spp. cultivated in Korea, A. saponaria, A. vera and A. arborescens and a new variant in Korea based on three plastid (matK, trnL-F, rbcL) and one nuclear (ITS regions) DNA barcode markers. A total of 2,420 bp sequence was amplified. Two indels were detected in the trnL region, and also several species specific nucleotide loci were detected in all 29 parsimonious informative sites, and 148 variable sites were detected among four taxa studied while 170 variable and 75 parsimonious sites were detected when other Aloe spp. in worldwide were used. An UPGMA phenogram with 10,000 bootstrap replication showed that the new variant was closest to A. vera. The variant was not morphologically and genetically concurrent with any reported species so far. The clustering of Aloe species were broadly in agreement with previously reported results.

  • RAPD analyses were performed to investigate genetic relationships and useful molecular maker for 3 species and their 17 regional populations of the Pueraria lobata and related taxa. The length of amplified DNA fragments ranged from 200 to, 2,800 bp. Two hundred and eight scorable polymorphic makers and three scorable monomorphic makers were found from the PCR reactions with 15 random oligo primers, and those were analyzed by Nei's genetic distance coefficient. Based on the UPGMA phenogram from RAPD analyses, two major groups (9 populations from Korea; 3 populations from foreign countries) were recognized. And it showed distinct genetic differences from related taxa. The RAPD results was very useful to define the samples by geographical distribution and to discuss the relationships among the populations and their related taxa of the Pueraria lobata.

Numerical taxonomy of Rhus sensu lato (Anacardiaceae) in Korea (한국산 광의의 붉나무속(Rhus L. sensu lato)의 수리분류학적 연구)

  • Numerical analysis based on the 67 morphological characters from 28 populations of 6 species of Korean Rhus sensu lato (Anacardiaceae) was performed for the taxonomic delimitation. Based on the results of PCA with 47 quantitative characters, the sum of contributions for the total variance of three major principal components was 77,9% (PCl 35.2%, PC2 22.5% and PC3 20.2%). The sum of contributions for the total variance of three major principal components were 90,7% (PCl 37.7%, PC2 33.0% and PC3 20.0%) based on the results of PCA with 20 qualitative The characters. Two dimensional plotting from PCA results recognized six distinct species. UPGMA phenogram based on simple matching coefficient method recognized clear taxonomic delimitations among six taxa. On the cluster analysis, qualitative characters were more useful for grouping the species treated. Numerical analysis was very valuable to delimit the Korean taxa of Rhus s.l.
