Central Nervous Depressant Activity of 5-Phenyl-2, 4-pentadienoic Acid Amide Derivatives (5-Phenyl-2, 4-Pentadienoic Acid Amide 유도체의 중추 억제작용에 관한 연구)

  • In order to find out the pharmacologically active but less toxic compounds than piperine, the actions of several amide derivatives of 5-phenyl-2, 4-pentadienoic acid on the CNS depressant activities were examined. The nine amide derivatives were investigated by using ICR mice as an experimental amimals on acute toxicities, anticonvulsant, sedative, analgesic and antipyretic effects. In the case of acute toxicities, all derivatives were weaker than piperine except compound III. Compound I showed strong anticonvulsant activity. On the other hand, compound I, III and IV significantly prolonged the sleeping time induced by hexobarbital in mice. Compound I, III and IV exhibited better analgesic effect than aspirin while compound II, V, VII and IX showed similar antipyretic activity to aspirin.

  • In the previous reports (Park et al., 1991, and 1993), we described the synthesis and analgesic effects of various homovanillic amides as analogs of capsaicin. In the study, we tried to enhance the analgesic actvity of capsaicin by structural modification. Our study has been performed in three directions. First, the amide bond of capsaicin was transposed. Second, a phenyl ring was introduced to replace a double bond of capsaicin. Finally, aminoethylation was performed on 4-hydroxy group of capsaicin to improve oral bioavailability. These studies have led to N-(3-phenylpropyl)homovanillic amide 2 which has high analgesic activity. Our continuing efforts in this area have focused on the introduction of various substituents on the phenyl ring of 2 as well as their pharmacological studies. We report herein the synthesis of homovanillic amide derivatives and their analgesic activity.

Liquid Chromatographic Resolution of Racemic $\alpha$-Amino Acid Derivatives on an Improved $\pi$-Acidic Chiral Stationary Phase Derived from (S)-Leucine

  • A chiral stationary phase derived from (S)-N-(3,5-dinitrobenzoyl)leucine N-phenyl N-alkyl amide (CSP 2) was applied in separating the two enantiomers of various π-basic aromatic derivatives of leucine N-propyl amide in order to evaluate π-basic aromatic groups as an effective derivatizing group for the resolution of a-amino acids. Subsequently N-(3,5-dimethoxybenzoyl) group was found to be very effective as a π-basic aromatic derivatizing group. Based on these results, N-(3,5-dimethoxybenzoyl) derivatives of various a-amino N-propyl amides, N,N-diethyl amides and esters were resolved on the CSP derived from (S)-N-(3,5-dinitrobenzoyl) leucine N-phenyl N-alkyl amide (CSP 2) and the resolution results were compared with those on the CSP derived from (S)-N-(3,5-dinitrobenzoyl)leucine N-alkyl amide (CSP 1). The enantioselectivities exerted by CSP 2 were much greater than those exerted by CSP 1. In addition, racemic N-(3,5-dimethoxybenzoyl)-a-mino N,Ndiethyl amides were resolved much better than the corresponding N-(3,5-dimethoxybenzoyl)-a-mino N-propyl amides and esters on both CSPs. Based on these results, a chiral recognition mechanism utilizing the π-π donor-acceptor interaction and the two hydrogen bondings between the CSP and the analyte was proposed.

A Study on Chemical Cyclodehydration of Aromatic Poly(ether-amide-amic acid)s (방향족 폴리(에테르-아미드-아미드산) 중합체의 화학적 탈수 고리화 반응)

  • A study has been made about some correlations in the chemical cyclization of precursors, poly(ether-amide-amic acid)s by treating in solution a mixture of acetic anhydride and pyridine in the presence of 4,4-dimethyl formamide, with the poly(ether-amic acid)s being respectively reacted between trimellitic anhydride chloride and 3 kinds of diamines, i.e., 4,4'-bis(m-aminophenoxy) benzophenone, 2,2'-bis[4-(m-aminophenoxy) phenyl] propane and 4,4'-bis(m-aminophenoxy) diphenyl sulfone. The cyclization of imide ring in the poly(ether-amide-amic acid)s may be regarded as an intramolecular acylation of amide group by o-carboxyl group. As a result of this reseach, the effects on the conversion to poly(ether-amide-imide)s have been found by changing the ratio of cosolvents in the cyclization mixture.

  • Syntheses and liquid-Crystallinites of thermotropic copoly (ester amide)s were investigated. The three components melt polycondensation of 4,4'-dicarboxy-$\alpha$, $\omega$-diphenoxyalkane as an A component, 4-4'diacetoxybiphenyl as a B, and p-N-acetoxy-aminobenzoic acid as a C gave the thrmotropic copoly(ester amide)s containing a flexible splacer in the polymer backbone. Diacetylated hydroquinone, methyl hydroquinone, chlorohydroquinone, and phenyl hydroquinone were used as anther B components. A polymer(6BPAB) having 10 mol% of C component and hexamethylene space. showed a typical nematic texture between $245^{\circ}C(T_m)\; and\; more\; than\; 360^{\circ]C(T_i)$. The melting points of the members of this series of polymers increased with decreasing methylene spacer. The polymer structure and mesomorphic nature were examined by solid and solution ^{13}C-NMR4 spectroscopy, cross polarizing microscopy with a hot stage, and X-ray diffactometry.

Synthesis and Properties of Novel Flame-Retardant and Thermally Stable Poly(amideimide)s from N,N'-(bicyclo[2,2,2]oct-7-ene-tetracarboxylic)-bis-L-amino Acids and Phosphine Oxide Moiety by Two Different Methods

  • N,N'-(bicyclo[2,2,2]oct-7-ene-tetracarboxylic)-bis-L-amino acids 3a-g were synthesized by the condensation reaction of bicyclo[2,2,2]oct-7-ene-2,3,5,6-tetracarboxylic dianhydride 1 with two equimolars of Lalanine 2a, L-valine 2b, L-leucine 2c, L-isoleucine 2d, L-phenyl alanine 2e, L-2-aminobutyric acid 2f and L-histidine 2g in an acetic acid solution. Seven new poly(amide-imide)s PAIs 5a-g were synthesized through the direct polycondensation reaction of seven chiral N,N'-(bicyclo[2,2,2]oct-7-ene-tetracarboxylic)-bis-L-amino acids 3a-g with bis(3-amino phenyl) phenyl phosphine oxide 4 by two different methods: direct polycondensation in a medium consisting of N-methyl-2-pyrrolidone (NMP)/triphenyl phosphite (TPP)/calcium chloride ($CaCl_2$/pyridine (py), and direct polycondensation in a tosyl chloride (TsCl)/pyridine (py)/N,N-dimethylformamide (DMF) system. The polymerization reaction produced a series of flame-retardant and thermally stable poly(amide-imide)s 5a-g with high yield. The resulted polymers were fully characterized by FTIR, $^1H$ NMR spectroscopy, elemental analyses, inherent viscosity, specific rotation and solubility tests. Data obtained by thermal analysis (TGA and DTG) revealed that the good thermal stability of these polymers. These polymers can be potentially utilized in flame retardant thermoplastic materials.

  • The crystal structure of the potassium salt of penicillin V has been studied by the X-ray crystallographic methods. Crystal data are as follows; potassium 3,3-dimethyl-7-oxo-6-phenoxyacetoamido-4-thia-1- azabicyclo[3.2.0]-heptane-2${\alpha}$-carboxylate, $K^+{\cdot}C_{16}H_{18}N_2O_5S^-$, $M_r$= 388.5, triclinic, Pl, a= 9.371 (1), b= 12.497 (2), c= 15.313 (2) ${\AA},\;{\alpha}= 93.74\;(2),\;{\beta}=99.32\;(1),\;{\gamma}=90.17\;(1)^{\circ},\;V=1765.7\;(2)\;{\AA}^3$, Z=4, $D_m=1.461\;gcm^{-1},\;{\lambda}(Cu\;K{\alpha})=1.5418\;{\AA},\;{\mu}=40.1\;cm^{-1}$, F(000)=808, T=296 K. The structure was solved by the heavy atom and difference Fourier methods with intensity data measured on an automated four-circle diffractometer. The structure was refined by the full-matrix least-squares method to a final R= 0.081 for 3563 observed $[I_0{\geq}2{\sigam}(I_0)]$ reflections. The four independent molecules assume different overall conformations with systematically different orientations of the phenyl groups although the penam moieties have the same closed conformations. There are intramolecular hydrogen bonds between the exocyclic amide nitrogen and phenoxy oxygen atoms. The penam moiety is conformationally very restricted although the carboxyl and exocyclic amide groups apparently have certain rotational degrees of freedom but the phenyl group is flexible about the ether bond despite the presence of the intramolecular N-H${\cdots}$O hydrogen bond. There are complicated pseudo symmetric relationships in the crystal lattice. The penam moieties are related by pseudo 20.5 screw axes and the phenyl groups by pseudo centers of symmetry. The potassium ions, related by both pseudo symmetries, form an infinite zigzag planar chain parallel to the b axis. Each potassium ion is coordinated to seven oxygen atoms in a severely distorted pentagonal bipyramid configuration, forming the infinite hydrophilic channels which in turn form the molecular stacks. Between these stacks, there are only lipophilic interactions involving the phenyl groups.

Fabrication of Amino Acid Based Silver Nanocomposite Hydrogels from PVA- Poly(Acrylamide-co-Acryloyl phenylalanine) and Their Antimicrobial Studies

  • New silver nanoparticle (AgNP)-loaded amino acid based hydrogels were synthesized successfully from poly (vinyl alcohol) (PVA) and poly(acryl amide-co-acryloyl phenyl alanine) (PAA) by redox polymerization. The formation of AgNP in hydrogels was confirmed by using a UV-Vis spectrophotometer and XRD. The structure and morphology of silver nanocomposite hydrogels were studied by using a scanning electron microscopy (SEM), which demonstrated scattered nanoparticles, ca. 10-20 nm. Thermogravimetric analysis revealed large differences of weight loss (i.e., 48%) between the prestine hydrogel and silver nanocomposite. The antibacterial studies of AgNP-loaded PAA (Ag-PAA) hydrogels was evaluated against Escherichia coli (Gram-negative) and Staphylococcus aureus (Gram-positive) bacteria. These Ag-PAA hydrogels showed significant activities against all the test bacteria. Newly developed hydrogels could be used for medical applications, such as artificial burn dressings.

  • Ultrafiltration membranes (UF) were manufactured by the conventional phase inversion method using an additives with N-methyl-2-pyrrolidinone as a solvent. Characteristics of performance could be controlled by the preparation conditions and the operating methods. The fouling resistance was observed by the relative ratio of permeate flux $(J_t)$/pure water flux $(J_o)$. Compared with the anion charged membranes and its original polyamide membrane, fouling resistance to the protein was increasing in proportion to the ion exchange capacity. The relative flux for the bovin serum albumin (BSA) solution increased as pH value further away from isoelectric point of BSA. The hydrophilicity of a membrane, the pH condition, and the operating temperature played the important role in the membrane permeations.
