Glucose Content and pH of Broiler and Porcine Blood Plasma by Glucose Oxidase or Baker's Yeast Addition (Glucose Oxidase 및 제빵용 효모 첨가에 따른 육계와 돼지의 혈장 포도당과 pH 변화)

  • The effects of GOD and yeast on glucose content and pH of broiler and porcine blood plasma were investigated. The initial glucose concentration of broiler and porcine blood plasma were $150mg/100cm^{3}\;and\;143mg/100cm^{3}$, respectively. Addition of GOD and yeast decreased glucose contents in broiler and porcine plasma. As expected, plasma glucose content decreased as incubation time increased. While 1080 and 1110 min were required to remove glucose from both broiler and porcine plasma at GOD 5 units/g and 480 and 1020 min were required at GOD 10 units/g, respectively; both required 240 min at 0.3% yeast (w/w). The Maillard reaction can be prevented by desugarization. During the removal of glucose, pH of the plasma decreased. As glucose content in plasma leveled off, the pH value of plasma increased. Therefore, pH may be used as an index of desugarization.

Comparison of Li-heparinized Versus Na-fluorinated Plasma for Routine Blood Glucose Determination Using Hexokinase Procedure (Hexokinase 방법을 이용한 Heparin 혈장과 NaF 혈장 시료의 혈당 분석치 비교)

  • In this study, the two sampling methods for blood glucose measurement were evaluated in order to reduce pre-analytical error. For this purpose, glucose levels of Li-heparinized plasma (LHP) and Na-fluorinated plasma (NFP) in blood collected from healthy volunteers were determined by using routine hexokinase procedures. The percentage range of pre-analytical error was quantitated by comparing LHP glucose values with NFP glucose values according to delaying analytical time (0, 60, and 120 min). LHP glucose values were decreased by 6.6% in 60 min., 17.7% in 120 min, whereas NFP glucose values decreased by 1.1% in 60 min, 2.0% in 120 min. Therefore it may be recommended that the NFP sampling method should be used for routine blood glucose determination in diabetes mellitus diagnosis.

Effects of Buckwheat on Organ Weight, Glucose and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats (메밀급여가 Streptozotocin유발 당뇨쥐의 장기무게 및 당질과 지질대사에 미치는 영향)

  • The present study was undertaken to evaluate the effects of 50% buckwheat diet on the body weight, organ weight, urine albumin, urine glucose, plasma glucose and plasma lipid in normal rats and diabetic rats treated with streptozotocin(STZ). The food intake, body weight, the level of urine glucose in diabetic buckwheat groups were not significantly different with diabetic control group. The level of urine albumin was lower in raw and steam buckwheat group than in the diabetic control group. Compared to the normal control group, liver and kidney weights were heavier in the diabetic groups. Pancreas weight was heavier diabetic buckwheat groups than in normal and diabetic control groups. Fasting plasma glucose level of diabetic buckwheat groups significantly decreased by 18-37% compared with the diabetic control group. Plasma triglyceride level of diabetic buckwheat groups significantly decreased by 34-50% compared with the diabetic control group. Plasma total cholesterol level of diabetic buckwheat groups decreased by 15-27% compared with the diabetic control group. The level of HDL-cholesterol was not affected by buckwheat diet. These results indicate that buckwheat is an effective therapeutic regimen for the control of metabolic derangements in diabetics.

Effect of Oolong Tea Extracts on Plasma Glucose Level and Antioxidant System in Diabetic Rats

  • The present study was conducted to investigate the effect of oolong tea extract on blood glucose level and antioxidant system in diabetic rats. The Sprague-Dawley rats were fed on AIN-76 based experimental diets containing 1 % oolong tea extract for 6 weeks. They were induced to be diabetic by receiving streptozotocin (45mg/kg BW) intramuscularly. Blood glucose, blood and hepatic concentration of vitamins A and E, and antioxidant enzyme activities were measured. Oolong tea extract feeding decreased the plasma glucose in diabetic rats. Dietary supplementation of oolong tea extract did not affect antioxidative enzyme activities such as superoxide dismutase, glutathione peroxidase and catalase in diabetic rats. The plasma level of retinol was increased in diabetic rats by feeding oolong tea extract. Plasma and hepatic levels of ${\alpha}$-tocopherol were higher in diabetic rats fed oolong tea extract. In conclusion, these results suggest that oolong tea extract consumption might reduce the plasma glucose in diabetic rats and protect the oxidative damage from diabetic stress to some extent.

  • The seasonal trends of plasma protein and glucose concentrations in marbled soles(Limanda yokohamae) and greenlings(Hexagrammos otakii), and the influence of transport stressor on those levels were investigated. Total plasma protein levels of marbled soles and greenlings in late spring and summer were significantly higher than those in winter(January). Plasma glucose levels were consistently increased according to elevation of water temperature both in marbled soles and greenlings. Transport stressor gave rise to decrease of plasma protein levels and increase of blood glucose levels.

  • The early plasma gastrin responses to single oral glucose or casein solution were studied in the same normal subjects on different days. After an overnight fast, glucose or casein solution was ingested within few minutes at the breakfast time. The plasma gastrin responses to these solutions were compared and contrasted with the concentration when the subjects received glucose solution intravenously. Results were as follows: 1) Rapid intravenous glucose infusion did not produce any changes in the plasma gastrin concentration. 2) Plasma gastrin concentration rose and peaked within 10 minutes after an oral liquid ingestion and then decreased substantially by 15 minutes, but remained slightly above fasting levels at 60 minutes. 3) There was no significant difference between the mean plasma gastrin concentrations after glucose of casein ingestion, but each fluid produced a significant increase in serum gastrin above fasting levels. 4) The subjects who produced high plasma gastrin response to glucose solution did so to casein solution. Conversely a low response to one solution reflected a low response to the other solution. 5) From the above results, authors discussed that individual responsibility rather than the sorts of meals is the factor in the determination of the magnitude of the early gastrin response.

Correlation between the Plasma Insulin and Glucose Concentration in Normal Korean Adults (한국인 혈장 Insulin과 혈당량의 상호관계에 관한 고찰)

  • The correlation between the plasma insulin, and glucose concentration was studied in healthy Korean adults consisting of 20 males and 22 females of 16 to 38 years of age. The blood samples of above subjects were obtained through cubital vein at arbitrary times, during their usual working hours. Plasma insulin was assayed by means of double antibody system of radioimnmunoassy technics, and blood glucose was determined by means of Van Slyke-Folch method. Results were as follows: 1. There were no differences in the blood sugar levels in relation to the plasma insulin concentration either by sex or age. 2. In the case, when the plasma insulin concentration was within $200m{\mu}D/ml$, the correlation between the insulin, and glucose concentration existed, the ratio of which was expressed as; Plasma glucose concentration(mg/dI)=$91.9+0.08{\times}Insulin$ concentration r=0.62 3. Insulinogenic index was 12.4%, which was somewhat higher than other reports. 4. It is suggested that the correlation between plasma insulin and glucose concentration could be determined at arbitrary times instead of fasting times.

Effect of Dietary Conjugated Linoleic Acid on Plasma Levels of Glucose and Lipids and Hepatic Lipogenic Enzyme Activity in Otsuka Long Evans Tokushima Fatty Rats (OLETF 비만쥐에서 CLA첨가 식이가 혈장의 포도당과 지질농도 및 간조직의 Lipogenic Enzyme 활성도에 미치는 영향)

  • The study was designed to observe whether the conjugated linoleic acid supplemented to diet could reduce plasma levels of glucose and lipids which were increased in 27-weeks old Otsuka Long Evans Tokushima Fatty(OLETF) rats. Twenty male OLETF rats of 7 weeks old were fed an experimental diet containing 4.5%(w/w) total fat including 1% CLA and six of twenty rats were sacrified at 6 weeks feeding. The rest of OLETF rats was divided into 2 groups, one group was continuously fed for 14 weeks more the same experimental diet containing 1% CLA and the other group was fed control diet which eliminated CLA. CLA did not significantly reduce food intake and body weight gain in OLETF obese rats. Plasma triglyceride and total cholesterol level were significantly increased at older age of OLETF obese rats, but CLA could significantly reduce plasma cholesterol and triglyceride increased in obese rats. However, CLA was not strong enough to reduce the increased plasma glucose level and hepatic lipogenic enzyme acitivies. CLA was mostly deposited in epididymal fat pad and could be incorporated into hepatic microsomal membrane and did interfere the conversion of C18 : 0 into C18 : 1 in liver. In conclusion, CLA could have anti-atherogenic effect by reducing plasma cholesterol and triglyceride which was increased in genetically obese rats, but CLA(1%) was not good source of dietary fatty acid to reduce body fatness and plasma glucose which was increased by obese gene in older rats.

Effect of Guar gum levels in Backsulgies on Plasma Glucose and Insulin in Healthly Men (백설기에 첨가된 Guar gum 수준이 혈장 포도당과 인슐린에 미치는 영향)

  • Backsulgies containing various levels of guar gum(0g, 2.5g or 5g) were ingested by ten healthy male subjects after 12-h fast. Guar backsulgies was given a time in a week a time in a week for 3 weeks with 75g glucose. The Sensory qualities of backsulgies containing guar gum powder of different levels(0g, 2.5, 5g) were also evaluated using a hedonic scoring technique. On each test day 4ml samples of venous blood were taken from the fasted subjects. Further 4ml post-prandial blood samples were taken from the subjects 30, 60, 90 and 120min after guar backsulgies had commenced. Result were as follows: Plasma glucose and insulin levels were not sifnificantly reduced according to guar gum levels in backsulgies. But plasma glucose and insulin levels of the case which ingested 5g guar backsulgies tended to be lower than those of the case which ingested 0g or 2.5g guar backsulgies. Therefore it is recommended that 5g guar backsulgies is most effective snack of diabetics. KEY WORDS: guar backsulgies plasma glucose, plasma insulin, snack.

Effects of Exercise on Pasma Glucose and Cholesterol Level in Streptozotocin-induced Diabetic Rats Fed Buckwheat Diet

  • The ability of exercise and buckwheat diet to modify plasma glucose and cholesterol levels in streptozotocin-induced diabetic rats has been studied. Diabetic rats were fed corn starch as a control diet or buckwheat as an experimental diet for 4 weeks. One group of rats were exercise-trained to swim for 60 min/day, 6 days a week. Plasma glucose levels of sedentary rats both on the control diet and on the buckwheat diet were significantly increased from 367.0$\pm$33.6 mg/dl to 545.0$\pm$38.7 mg/dl and from 380.3$\pm$18.9 mg/dl to 540.5$\pm$36.6 mg/dl, respectively. However, this large increase of plasma glucose was not seen in exercised rats on the buckwheat diet (from 345.0$\pm$25.6 mg/dl to 391.4$\pm$34.7 mg/dl). The total plasma cholesterol level was not affected by either diet or exercise. The HDL-cholesterol level tends to increase due to the buckwheat diet or an exercise, but not with a significant increase. Our results suggest that the buckwheat diet is beneficial in lowering the plasma glucose level only when diabetic rats ar exercised.
