Effect of Sucrose and Boric Acid on In Vitro Pollen Germination of Boxthorn (Lycium chinense M.) (자당과 붕산이 구기자 화분 발아에 미치는 영향)

  • This study was carried out to investigate the effect of sucrose, boric acid, and calcium nitrate in optimum pH and relative humidity on pollen germination and pollen tube growth of boxthorn (Lycium chinense M.). Medium containing 100mg/L of $H_3BO_3$ showed maximum pollen germination and pollen tube elongation, 64.8% and $920{\mu}m$. Additionally supplied calcium nitrate to the medium lowered pollen germination rate and pollen tube elongation. Here we report the optimum conditions for the pollen germination and pollen tube development was observed at 10% sucrose, 1.0% agar, pH 6.0, and 100% relative humidity.

Factors Influencing Pollen Germination in vitro of Codonopsis lanceolata (더덕의 花粉發芽에 관한 溫度와 培地條件)

  • The present study has been undertake to obtain the fundatmental data of optimum germination condition and to establish storage time for artificial pollinationin Codonopsis lancelata pollen. In vitro condition for germination of freshly collected and stored pollen were examined. The optium temperature for germination of fresh pollen was $25^{\circ}C$. The optium sucrose concentration in the medium ranged from 30 to 40 % and optium pH 6.0% for pollen germination. The rate of pollen germination accelerated considerably in the medium with 1% agar. 30% sucrose, and pH 6. C. lanceolata pollen remained viable for 15 days when stored at 5$^{\circ}C$ with silica gel as desiccant.

Optimal Condition for Pollen Germination of Rare and Endangered Forsythia saxatilis (희귀.멸종위기 산개나리(Forsythia saxatilis) 화분의 최적 발아 조건)

  • Optimal condition for pollen germination was suggested as a basic research of flowering physiology in order to identify the characteristics of flowering and seed setting of rare and endangered Forsythia saxatilis Nakai. Pollen samples were collected during flowering time from the end of March to the beginning of April. First, a suitable germination temperature, medium sucrose concentration and germination time were determined for pollen germination and pollen tube elongation in vitro, and then an optimal pH of culture medium. Pollen germination and tube elongation were significantly different among the levels of germination temperature, sucrose concentration and germination time. Interactive effects were observed between germination temperature and time, germination temperature and sucrose concentration, germination time and sucrose concentration. Pollen germination was the highest at $10^{\circ}C$ and increased with the increase of sucrose concentration, whereas it had no relation with germination time. In addition, pollen germination and tube elongation did not increase at more than 15% of sucrose concentration and 24 hours later. Pollen germination was the highest at pH 5 (20.8%) and the lowest at pH 6 (3.8%). In conclusion, $10^{\circ}C$, 15% sucrose and pH 5 were proposed as the optimal condition for pollen germination 24 hours later of pollen culture.

In vitro Pollen Performance of Pinus densiflora and P. rigida: Temperature and Medium Nutrient Effects

  • Effects of environmental factors on in vitro pine pollen performance were investigated. Pinus densiflora and P. rigida pollen grains collected at Mt. Kwanak, Korea were used. Three environmental factors, such as pollen storage temperature, pollen culture temperature and nutrient condition in medium, were tested. To determine the storage temperature effects on pollen viability, pine pollen was stored at $-70^{\circ}C$, $-12^{\circ}C$, $4^{\circ}C$ and $22^{\circ}C$. Pollen viability was substantially extended at the storage temperatures of $-12^{\circ}C$ and $4^{\circ}C$ for more than 300 days. To elucidate the culture temperature effects on pine pollen germination and tube growth, pollen grains were cultured at the temperatures from $5^{\circ}C$ to $40^{\circ}C$ at $5^{\circ}C$intervals. The germination rate and tube growth were highest at $25^{\circ}C$ and decreased above $30^{\circ}C$. To investigate boron and sucrose effects on pollen tube growth, the pollen was cultured at different sucrose and boric acid concentrations. Germination rate was optimal in germination medium containing 3 or 5$\%$ sucrose with 0.01 $\%$ boric acid. These results indicate that the pine pollen can be stored for considerable length of time without noticeable loss of viability at storage temperature below or near $0^{\circ}C$. Optimal germination medium conditions were established for pine pollen. Therefore, pine pollen can be used for many biological and environmental monitoring researches.

Effect of Germination Temperature, Spermine and Putrescine on Pollen Viability of Strawberry (발아온도 및 Spermine, Putrescine이 딸기의 화분 발아에 미치는 영향)

  • This study was conducted to determine the optimum conditions for the treatment of polyamine to promote pollen germination and improve the fruiting rate by overcoming fertilization defects in low-temperature strawberry cultivation. The optimum temperature for pollen germination of strawberries was 30 ℃ for Seolhyang and 25 ℃ for Maehyang, and Seolhyang had a higher pollen germination rate than that of Maehyang. The addition of polyamines, namely spermine and putrescine to the medium improved the pollen germination rate and spermine treatment showed a better effect than putrescine treatment. The proper polyamine type and treatment concentration for enhancing the potency of pollen germination was 500 μM of spermine for both Seolhyang and Maehyang, which improved the pollen germination rate by 19-23% compared with that of the control. However, combined treatment of spermine and putrescine, resulted in a lower germination rate lower than that of the single treatment. Our results indicated that the treatment of polyamines during flowering in protected cultivation of strawberrise can improve the fruiting rate by overcoming the problem of poor pollen germination due to low temperature.

Effect of Air Temperature and Relative Humidity during Flowering on Pollen Germination of Oriental Melon (Cucumis melo L. var. makuwa Makino) (개화 시 기온 및 상대습도가 참외의 화분발아에 미치는 영향)

  • This study was conducted to examine the effect of temperature and relative humidity (RH) that might affect the pollen germination rate and pollen tube length in oriental melon. The experiment was performed using three cultivars namely 'Mallijangseong', 'Ohbokggul', and 'Joeundae' wherein environmental conditions were controlled by three different temperatures $15^{\circ}C$, $25^{\circ}C$, and $35^{\circ}C$ and also three RH levels 30%, 60%, and 90%. Based on the results in terms of temperature, true to all cultivars, plants exposed to $25^{\circ}C$ had the highest pollen germination rate while plants exposed to $15^{\circ}C$ had the lowest. Among the three cultivars, 'Mallijangseong' had the highest in terms of pollen germination rate, followed by 'Ohbokggul', and lastly was 'Joeundae' that showed the lowest germination rate. In the case of pollen tube length, pollen exposed under $35^{\circ}C$ showed the longest tube length, followed by pollen under $25^{\circ}C$, and pollen under $15^{\circ}C$ remarkably had the shortest tube length. Across the three cultivars 'Ohbokggul' had the longest pollen tube length. In the case of RH, pollen germination response among three cultivars depends on the humidity conditions but based on the results, there was no significant differences although high germination rate was observed in highest humidity condition. Between each cultivar, 'Ohbokggul' had the lowest pollen germination rate compared with 'Mallijangseong' and 'Joeundae' after exposure to different RH conditions wherein pollen germination showed significant differences among treatments. The pollen collected under 60% RH had the longest pollen tube length, followed by 30%, and the shortest was under 90% RH. In general, 'Ohbokggul' had the longest pollen tube length while 'Joeundae' had the shortest among the three cultivars.

Effect of Boric Acid on In Vitro Pollen Germination in Transgenic Plants Expressing Monoclonal Antibodies (단일항체를 발현하는 형질전환 식물체의 In Vitro 화분발아에 대한 Boric Acid의 영향)

  • Pollen germination viability is an essential factor to produce seeds from pollination and fertilization, which are required to maintain plant generation. In this study, we tried to identify the effect of boric acid on pollen germination and tube grouch in non-transgenic and transgenic plants expressing monoclonal antibodies (anti-colorectal cancer mAb CO17-1A, anti-breast cancer mAb BR55, and anti-rabies virus mAb57). The pollen of non-transgenic plant was treated with different concentration of boric acid (0, 5, 10, 15, 20, $40{\mu}g/mL$) in germination buffer to investigate its effect on in vitro pollen germination. At $20{\mu}g/mL$ of boric acid, tile pollen germination rate was the highest (49.5%) compared to other concentrations. In general, the germination rate significantly increased 3-10 folds in boric acid ($20{\mu}g/mL$) treated group in non-transgenic and transgenic plants. Also, the pollen tube length increased in boric acid ($20{\mu}g/mL$) treated groups. In the treated group, the pollen tube length increased until 3 h boric acid treatment and decreased after the 3 h, indicating that the 3 h is the most appropriate incubation time period. Western blot analysis showed that the mAb transgene expression was more stable in leaf than pollen in transgenic plants. This study suggested that $20{\mu}g/mL$ of boric acid is ideal concentration to induce in vitro pollen germination of transgenic plants expressing therapeutic monoclonal antibodies, indicating stable pollination and fertilization in transgenic plants.

Effect of Various Fungicides on Pollen Germination and Pollen Tube Growth in Some Fruit Trees (각종 살균제(殺菌劑)가 주요과수(主要果樹)의 화분발아(花粉發芽) 및 화분관신장(花粉管伸長)에 미치는 영향(影響))

  • The experiment was carried out in order to choose fungicides which are proper to be used in flowering time of fruit trees. The effect of fungicides on pollen germination and pollen tube growth was investigated in vitro. The results obtained are as follows : 1. Difolatan-Wp, Lime-sulfur, Captan-Wp and Bordeaux mixture greatly inhibited pollen germination and pollen tube growth. 2. The effect of Daconil-Wp and Zimaneb-Wp on pollen germination and pollen tube growth was not so great as that of Difolatan-Wp, Lime-sulfur, Captan-Wp or Bordeaux mixture. 3. Polyram-combi-Wp, Topsin-Wp and Benlate-Wp did not affect pollen germination but these fungicides slightly inhibited pollen tube growth. 4. Polyoxin-Wp significantly increased pollen tube growth but did not show promotive effect on pollen germinaiton.

An Increment of Crossing Efficiency with Consideration of Pollen Viability Analysis in Rose (장미 교배 효율 증대를 위한 화분 임성 검정)

  • Current rose cultivars are all composed of heterozygous genome due to long history of out crossing including interspecific hybridization. It has been adapted by artificial selection and crossing by breeders that mainly based on the crossing with fertile pollen derived from inter- or intra-specific hybridization. Pollen viability and germination ability tests provide valuable information for the designing of parentage for more successful breeding efficacy. In this study, we tested the pollen viability and germination ability in seven rose cultivars to find any relationship among several factors including pollen size, ploidy levels, and crossing compatibility. The pollen viability showed wide ranges from 39% 'Pinocchio' as minimum to 82% 'Scarlet Mimi' as maximum, whereas pollen germination rate were from 1% 'Mini Rosa' to 41% 'Scarlet Mimi' as a highest. Pollen size ranged from 41.3 to $45.4{\mu}m$ in large sized pollen and 30.7 to $37.4{\mu}m$ in small sized pollen. The mean diameter of large sized pollen is approximately 10-40% bigger than that of small sized pollen. There are positive relationships among ploidy level, total chromosome length, and pollen size. Crossing list showed that seed setting ratio and seed germination were related to pollen viability, pollen germination, and ploidy level.

In Vitro Culture and Transformation by Agroinfiltration of Lisianthus (Eustoma russellianus) Pollen (Lisianthus 화분의 기내배양 및 Agroinfiltration에 의한 형질전환)

  • Optimized conditions for Agrobacterium-mediated lisianthus pollen transformation were adjusted using various factors such as temperature, pH and sucrose concentration. Pollen tube growth was successfully achieved in a medium (pollen germination medium; PGM) containing $7-15\%$ sucrose with pH in the range of 5.5-7.0 at temperature of $20-27^{\circ}C$. Lisianthus pollen was vacuum-infiltrated with Agrobacterium cell suspension for 20 min, and transformed pollen was confirmed by GUS histochemistry and Southern hybridization following RT-PCR. Transgenic pollen system may be utilized for establishing an area of plant transient expression systems based on the convenient pollen transformation procedure presented in here.
