New Approaches to Increase Skin Efficacy of Chaga Mushroom Extract using High Voltage P ulsed Electric Fields Technology (고전압 펄스 전기장 기술을 이용한 차가버섯 추출물의 피부 효능 증대 방법)

  • In this study in order to develop new approaches we investigated using high voltage pulsed electric fields (PEF) technology to reduce the risks, protect the phyto-constituents and improve skin biological activities. After preparing a Chaga mushroom (Inonotus obliquus) extracts pretreated with PEF, components measurement and skin efficacy evaluation were performed. As a result of the content measurement, the content of polysaccharide and polyphenol were higher in the order of extracts treated with 50 Hz and 25 Hz at 0.5 kV/cm, and the content of protein was the highest in extracts treated with 25 Hz at 0.5k V/cm. Similar to the results of the polyphenol measurements, extracts treated with 25 Hz and 50 Hz at 0.5 kV/cm showed leading DPPH scavenging ability. The cell protection effect against sodium dodecyl sulfate (SDS) and UVB was finest in extracts treated with 25 Hz at 0.5 kV/cm, which had the highest protein content. And the hyaluronic acid synthesis was leading in extracts treated with 50 Hz and 100 Hz at 0.5 kV/cm. Therefore, the active ingredient of the high-voltage PEF pre-treatment Chaga mushroom extract can be developed as a functional material with cell protection and moisturizing effect, and such green technology is expected to be used in various fields of cosmetics and material development.

Antioxidant Compounds and Antioxidant Activities of Fermented Black Rice and Its Fractions (흑미(복합)발효물의 항산화 성분 및 항산화 활성 평가)

  • The aim of this study was to determine the total phenol, total flavonoids, and proanthocyanidin contents of fermented black rice and its fractions, as well as to assess the antioxidant activities. Antioxidative activities were assessed in various in vitro models using 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), reducing power, ferric ion reducing antioxidant power (FRAP), and nitrite scavenging activity (Griess reagent assay). Our results show that the antioxidant activity was significantly higher in the low-molecular fraction of fermented black rice than in the other samples (p<0.05). Among the fermented black rice and its fractions, the low-molecular fraction had the highest total phenol ($109.2{\pm}2.9$ mg GAE, gallic acid equivalent/g), total flavonoids ($39.4{\pm}0.8$ mg RE, rutin equivalent/g), and proanthocyanidin ($32.9{\pm}1.4$ mg CE, catechin equivalent/g) contents, which correlated strongly with its antioxidative activity. Considering the high consumer demand due to the beneficial health effects, fermented black rice and its fractions can be utilized to develop functional food, as well as health-promoting and pharmaceutical agents.

Long-Term Effects of Growth Regulators and Nitrogen Sources on Proliferation and Turnover of Cell Wall Polysaccharides in Suspension Culture of Kidney Bean (Phaseolus vulgaris L.) (강낭콩의 현탁배양시 증식과 세포벽 다당류 전환에 미치는 생장조절제 및 질소원의 장기간 효과)

  • To underatand in vitro regulation of differentiation, the effects of growth regulators and nitrogen source on metabolism of cell wall polysaccharides in suspension culture of kidney bean (Phaseolus vulgaris L.) were investigated. The suspension cells (cell clusters) were directly induced from the epicotyl segments of the seedlings, which were cultivated in MS medium supplemented with 1.0mg/L of 2,4-D and 0.5 mg/L of kinetin. When compared with cell wall sugar contents of the epicotyl segments, the cellulose content of the suspension-cultured cells decreased; while the pectin and hemicellulose content increased; suggesting increases of rhamnogalacturonan I and arabinogalactan IIduring the dedifferentiation, respectively, The effects of growth regulators(2,4-D, 1.0mg/L and kinetin, 0.5mg/L) and nitrogen source (potasium nitrate, 19.0mg/L and ammonium nitrate, 16.5 g/L) in the medium on the proliferation and the turnover of the cell wall polysaccharides were investigated for 30 days. In the medium with growth regulators and without nitrogen source, the proliferation rate was extremely high (16 folds). Growth regulators and nitrogen source increased the pectin content. Analysis of neutral sugar composition of pectin fraction showed that nitrogen source enhanced rhamnose level remarkably, suggesting that rhamnogalacturonan I was the one most likely synthesized. In hemicellulose fraction, growth regulators reduced arabinose level, suggesting that arabinogalactan II was degraded. And nitrogen source reduced galactose level, suggesting that xyloglucan was also degraded.

Development and Transformation of Oocyte' Follicle Cell for Korean Four Sillurid Fishes of Liobagrus and Pseudobagrus (Pisces: Silluriformes) (한국산 동자개속 및 퉁가리속 4종 어류의 난모세포에서 여포세포의 발달과 변형)

  • A histological study on development and transformation of the oocyte' follicle cell for Korean four sillurid fishes, Liobagrus obesus, L. mediadiposalis, Pseudobagrus koreanus, and P. brevicorpus was performed by light and electron microscopes. The follicular layer surrounding the oocyte consisted of an outer theca cell and an inner follicle cell (granulosa cell). The follicle cells of the oocyte were flatten cells at early oocyte but during vitellogenesis they were transformed it to a single layer of cuboidal cell, then to a single columnar cell layer, and finally to a layer covered with a substance secreted by themselves. Although the development and transformation of the follicle cells was similar to four species, the secreted materials, called an adhesive membrane, were divided into two types in its appearance and nature. Firstly, a jelly coat-like type was found in L. obesus and L. mediadiposalis, which they are presumed to be polysaccharides and mucoproteins in its nature and secondly, a granular type in P. koreanus and P. brevicopus, being mucoprotein. A zona radiata with about $0.6{\sim}3.1{\mu}m$ thin was present below the adhesive material secreted by the transformed-follicle cell's activity. The zona radiata was composed of two layers, a thin externa and a thick interna.

Effects of Non-starch Polysaccharide-degrading Enzymes on Nutrient Digestibility, Growth Performance and Blood Profiles of Growing Pigs Fed a Diet Based on Corn and Soybean Meal

  • Two experiments with growing pigs were conducted to investigate the effects of two distinct multienzyme preparations on nutrient digestibility, growth performance and blood profiles. In Exp. 1, a total of 96 pigs ($29.7{\pm}0.69\;kg$) were utilized in a 42-day performance and digestibility trial using four dietary treatments: CON (control diet), ENDO (control+0.10% Endopower), NSPase1 (control+0.10% NSPase) and NSPase2 (control+0.20% NSPase). Endopower was a commercial multienzyme preparation which contained ${\alpha}$-galactosidase, galactomannase, xylanase and ${\beta}$-glucanase. NSPase mainly contained ${\alpha}$-1,6-${\beta}$-galactosidase, ${\beta}$-1,4-mannanase and ${\beta}$-1,4-mannosidase. There were six replication pens per treatment with four pigs per pen. Pigs fed NSPase1 diet had a higher ADG (p<0.05) and G:F (p<0.05) than those fed the control diet. There were no significant differences in growth performance among the multienzyme treatments (p>0.05). Compared with CON, apparent digestibility of DM was increased (p<0.05) by ENDO treatment. N digestibility was improved (p<0.05) in response to multienzyme treatments during the experimental period. Blood urea nitrogen (BUN) was higher (p<0.05) in ENDO treatment than in CON and NSPase1 treatments at the end of the experiment, while the glucose level improved (p<0.05) due to ENDO and NSPase2 treatments. In Exp. 2, four ileal-cannulated, growing barrows ($20.17{\pm}1.31\;kg$) were housed in individual metabolism crates and randomly assigned to 1of 4 treatments (same as Exp. 1) within a $4{\times}4$ Latin square design. Enzyme supplementations improved the majority of apparent ileal amino acid digestibilities (p<0.05). It is concluded that the supplementation of NSPase1 improved growth performance as well as N digestibility and partially improved apparent ileal amino acid digestibility in growing pigs fed a diet based on corn and soybean meal.

By-product of Tropical Vermicelli Waste as a Novel Alternative Feedstuff in Broiler Diets

  • Two experiments were conducted to determine physical and chemical properties of vermicelli waste (VW) and effect of VW inclusion levels on growth performance of broilers. In experiment 1, VW samples were randomly collected from vermicelli industry in Thailand to analyze nutritional composition. Vermicelli waste contained 9.96% moisture, 12.06% CP, 32.30% crude fiber (CF), and 0.57% ether extract (EE), as DM basis. The ratio of insoluble:soluble non-starch polysaccharide (NSP) was 43.4:8.9. A total of 120 chicks (6 pens per treatment and 10 chicks per pen) were fed a corn-soybean meal-based diet or 20% VW substituted diet to determine the apparent metabolizable energy corrected for nitrogen retention ($AME_n$) of VW. The $AME_n$ of VW was $1,844.7{\pm}130.71$ kcal/kg. In experiment 2, a total of 1,200 chicks were randomly allotted to 1 of 4 dietary treatments for 42-d growth assay. There were 300 chicks with 6 pens per treatment and 50 chicks per pen. The dietary treatments contained 0%, 5%, 10%, or 15% VW, respectively. All diets were formulated to be isocaloric and isonitrogenous. From 0 to 18 d of age chicks fed VW diets had higher (p<0.001) feed conversion ratio (FCR) compared with those fed the control diet. No difference was observed during grower and finisher phase (19 to 42 d). Chicks fed VW diets had lower relative weight of abdominal fat (p<0.001) but higher relative weight of gizzard (p<0.05) than those of chicks fed the control diet. Increasing VW inclusion levels increased ileal digesta viscosity (p<0.05) and intestinal villus height of chicks (p<0.001). For apparent total tract digestibility assay, there were 4 metabolic cages of 6 chicks that were fed experimental treatment diets (the same as in the growth assay) in a 10-d total excreta collection. Increasing VW inclusion levels linearly decreased (p<0.05) apparent total tract digestibility of DM and CF.

Anti-oxidative Activity and Moisturizing Effect of Fermented Puer Tea Extract (발효보이차 추출물의 항산화활성 및 보습효과)

  • The fermented puer tea leaves were high concentrated extracted with 50% ethanolic solution in this study. Also, evaluating the anti-oxidative activity and anti-aging effect of this extract, we expected to apply the cosmetic industry. The yield of fermented puer tea extract was 17.9%. The total polyphenol content was 37.5%, tannin content was 7.5%, polysaccharide was 17.9%, unknown compound was 22%, and water content was 8%. Anti-oxidative activity (in-vitro) of fermented puer tea extract by DPPH method was $33.7{\pm}1.8%$ in 30mg/mL, $39.4{\pm}2.2%$ in 50mg/mL respectively. Therefore, we could know that anti-oxidative activity of fermented puer tea extract was effect higher than tocopheryl acetate and greentea extract. The collagen synthesis activity (in-vitro) of fermented puer tea extract was increased with $102.9{\pm}9.9%$ in 1 mg/mL, $111.5{\pm}9.9%$ in 5 mg/mL, $122.7{\pm}12.2%$ in 10 mg/mL, $131.5{\pm}13.7%$ in 30 mg/mL (*p-value£0.05, n=3). Skin moisturizing activity of fermented puer tea extract after application 8 hours was increased 38.5% higher than control samples both tocopheryl acetate and greentea extract. Total moisturizing effect was increased about 32.7% compared to before treatment. Fermented puer tea extract of this study can be applied to the skin care cosmetics industry.

Immunomodulatory Activity of Crude Polysaccharides from Makgeolli (막걸리에서 분리한 다당의 면역자극 활성에 미치는 효과)

  • In this study, the immunomodulatory activities of crude polysaccharides from makgeolli were investigated. Crude polysaccahrides from makgeolli (RWW) were isolated by hot water extraction ($100^{\circ}C$, 30 min), ethanol precipitation (four volumes of 95% ethanol), dialysis (MWCO: 6,000~8,000), and lyophilization. The major constituents in RWW were neutral sugar (87.3%), uronic acid (2.5%), and protein (10.2%). RWW showed potent anti-complementary activity as well as increased cell proliferation of RAW 264.7 macrophages. The immunomodulatory effects of RWW were also analyzed based on cytokine production of macrophages. Macrophages stimulated with RWW produced cytokines such as interleukin (IL)-6, IL-12, and tumor necrosis factor-${\alpha}$ in a dose-dependent manner. These data indicate that RWW may have immunomodulatory effects through activation of the complement system and macrophages, which are a part of natural immunity.

The Study on Optimum Operation Conditions of Ceramic MF Membrane Process in Y Water Treatment Plant (Y 정수장 세라믹막 여과공정 최적 운영인자 평가)

  • This study was performed to discover the optimum operation conditions for the advanced water treatment using the ceramic membrane, introduced the first in the nation at the Y water treatment plant (WTP). The result of investigation to find the optimum operation conditions which can continue preserving the filtration performance as well as satisfying both the economics and the water quality is as follows. In the ordinary water quality condition of the Y WTP, the optimum filtration time(the backwash period), which can minimize the production of backwash waste and preserve the membrane performance was examined to be 4.0 hours on basis of institution capacity ($16,000m^3/day$). Examining the recovery rate of TMP from the chemical cleaning (CIP) discovered that the inorganic contaminants, which cause membrane fouling, such as iron, manganese, aluminum, were removed through the acidic cleaning using citric acid, whereas the membrane recovery rate was found to be low. But, on the other hand, the TMP was recovered to the initial value from the alkali cleaning using the NaOCl. Therefore, the main contaminant causing the fouling was determined to be hydrophilic organic compound( biopolymer). The membrane recovery rate is highly influenced by the temperature of the cleaning chemical. That is, the rate increased with increasing temperature.

  • The Brown-algae polysaccharide consisting of alginate and laminaran is usable as high bio-ethanol production if hydrolyzed to monomer unit. The objective of this study is to produce bio-ethanol from brown-algae using enzymatic saccharification. Bio-ethanol was produced by Saccharomyces cerevisiae KCCM 1129 and Pachysolen tannophilus KCTC 7937 strains. The substrate used Laminaria japonica, Sargassum fulvellum and Hizikia fusiformis. We isolated a new alginate lyase and laminaran lyase producing microorganism for hydrolysis of brown-algae from southern sea of Gijang. The reducing sugar was obtained 1.90 g/L from Laminarin japonica 20 g/L that used enzyme from Bacterium antarctica. In pretreatment of the most suitable brown-algae for ethanol production, ethanol concentration of 0.93 g/L and yield of 4.65% were obtained in condition of Laminaria japonica in medium.
