Evaluation of Achyranthes japonica Ethanol Extraction on the Inhibition Effect of Hyluronidase and Lipoxygenase (쇠무릎 에탄올 추출물의 DPPH, 히알루로니다아제 및 리폭시게나아제 저해 효과)

  • The 1, 1- diphenyl 2-picrylhyorazyl (DPPH) is a well-known radical and a trap (scavenger) for other radicals. Hyaluronidase (HAase) is an enzyme that depolymerizes the polysaccharide hyaluronic acid (HA) in the extracellular matrix of connective tissue. Lipoxygenase (LOX) enzyme was reported to convert the arachidonic, linoleic and other polyunsaturated fatty acid into biologically active metabolites involved in the inflammatory and immune responses. The purpose of the present study is to evaluate plant extracts as sources of natural antioxidants and to examine whether Achyranthes japonica having significant DPPH, HAase and LOX inhibitory activity. The inhibitory effect of HAase by A. japonica was assayed using a Morgan microplate assay. The antioxidant activity of the A. japonica extracts was measured on the basis of the scavenging activity of the stable 1, 1- diphenyl 2-picrylhyorazyl (DPPH) free radical. DPPH scavenging activity of matured roots of A. japonica was evaluated at 4.0 mg/ml was 87.8% and that of young roots was 86.2% at same concentration. The roots of A. japonica showed maximum inhibition of HAase activity (IC50 = 27.7 μg/ml). The highest LOX inhibition was recorded in the root extract among three vegetative parts. Inhibition of HAase activity of roots may contribute towards the development of herbal medicines. Although percent inhibition of lipoxygenase by Achyranthes japonica for all young and matured groups for leaves, stems, and roots at different concentrations, there were not show a statistically significant difference (p<0.05).

Optimal Conditions of Mycelial Growth and Exopolysaccharide Production in Submerged Culture of Phellinus baumii (Phellinus baumii으로부터 세포외 다당체 생산의 최적화)

  • The polysaccharide isolated from Phellinus species has been known as a folk remedy, including antitumor and immune-stimulating activities. However, there are lacks of knowledge about mycelial growth and exopolysaccharide (EH) production in its submerged culture. We investigated the optimal conditions on mycelial growth and EPS production in Phellinus baumii. The optimal temperature and initial pH for mycelial growth and EPS production in shake flask culture of P. baumii were proved to be 3$0^{\circ}C$ and pH 5.0, respectively. In case of carbon source, cellobiose and maltose were highly efficient for mycelial growth and fructose and mannitol were also relatively favorable for EPS production. Yeast extract was the most suitable nitrogen source for mycelial growth and EPS production. The composition of optimal culture medium was determined to be fructose 20 g/L, yeast extract 20 g/L, and $CaCl_2$ 0.55 g/L, respectively. Under the optimal culture condition, the maximum mycelial biomass and EPS achieved in a 5-L stirred-tank fermenter were 17.43 g/L and 3.6 g/L, respectively. It was found that the EPS was a glycoprotein onsisted of mainly arginine (14.1%) and glycine (12.0 %) in protein moiety and mainly mannose (48.7%) and arabinose (38.4%) in carbohydrate moiety.

Immunostimulating and Anticancer Activities of Hot Water Extract from Capsosiphon fulvescens (매생이 열수추출물의 면역 및 항암 활성)

  • To investigate the immunostimulating and anticancer activities from hot water extract of Capsosiphon fulvescens, tumor cell toxicity, sarcoma-180 growth inhibition activity, complement system-activity, intestinal immune system and oral toxicity were performed. The extract of Capsosiphon fulvescens was prepared by hot water and precipitated by using ethanol. Partially purified extract (CFE) was obtained after dialysis and ultrafiltration. The polysaccharide compositions consisted of xylose(19.1%), fucose(15.3%), mannose(4.2%) and galactose(7.9%). The tumor cell toxicity of CFE slightly showed at high concentrations of 10-30 ${\mu}g/ml$, but inhibition ratio against mouse solid tumor was more increased for CFE of 40.1-59.4% than the control. Blood leukocyte counts increased to a maximum of 83% including liver, spleen and thymic of mouse. Immunoglobulin A binding amounts showed a high level of CFE of $2,454{\pm}113.8-2,670{\pm}133.1{\mu}g/mg$ in comparison with the control of $2,092{\pm}123.0{\mu}g/mg$. Acute toxicity of CFE was not detected at the concentration of 2,000 mg/kg in normal mouse.

Anticoagulant Activity of Sulfated Polysaccharides Isolated from Codium fragile (청각 산추출물에서 정제한 함황다당류의 항응고활성)

  • We have isolated two anticoagulant polysaccharides from an acidic extract of Codium fragile. The purification was conducted using three consecutive chromatographies of DEAE-Toyopearl 650C, Sephadex G-100 (G-75), and Sepharose CL-6B by measuring activated partial thromboplastin time (APTT). The two purified anticoagulant polysaccharides, CF-1-VIa-1 and CF-1-VIIa-1, were found to be nearly homogenous on HPLC using a gel permeation column and appeared to have molecular weights of about 80,000 Da and 40,000 Da, respectively. The polysaccharides consisted mainly of arabinose and galactose in a molar ratio of about 2 : 1, and also comprised 12-13% of sulfates at their constituent sugars. CF-1-VIa-1 and CF-1-VIIa-1 inhibited blood coagulation via both the intrinsic and the extrinsic pathways. The polysaccharides unlike heparin showed an inhibitory activity on thrombin when a pure fibrinogen without antithrombin III was used as a substrate. Structural modifications using sulfation and desulfation affected the anticoagulant activities directly, suggesting that the content of sulfate plays an important role in the blood coagulation cascade. The polysaccharides may inhibit some proteases involved in the blood coagulation cascade, judging from the independence of calcium concentrations in their anticoagulant activity.

Effect of Carbon sources and Vitamins on Mycelial Growth of Tricholoma matsutake DGUM 26001 (송이균사(Tricholoma matsutake DGUM 26001)의 생육에 미치는 탄소원 및 비타민의 영향)

  • The mycelium was isolated from the fruiting body of Tricholoma matsutake collected from Mt. Namsan, Kyongju and it was named as Tricholoma matsutake DGUM 26001. For the mycelial growth of T. matsutake DGUM 26001, the complex media, yeast-malt extract medium and Czapek-Dox medium supplemented with yeast extract, were excellent. The media such as nutrient glucose medium, mushroom complex medium, and Tricholoma matsutake medium (TMM), were effective. However, There was no a mycelial growth in the media used for bacterial cultivation such as colombia medium, brain heart infusion medium, Luria-Bertani medium supplemented with glucose, and brucella medium. When carbohydrate as a carbon and energy source was supplemented in the TMM medium for the mycelial growth, starch as a polysaccharide was best. As a disaccharide, trehalose and maltose were excellent. Sorbitol, xylitol and glucose were excellent carbon sources of monosaccharose. When the mycelia were cultivated for 30 days at $24^{\circ}C$ in the TMM supplemented with 2.0% starch, the dry weight of the mycelia harvested was 8.85 g/L. When organic acid was given as a carbon source, only succinic acid was utilized. As a vitamin source, coconut water and pyridoxine were excellent. After 30 day-cultivation in the TMM medium, the dry weights with coconut water and pyridoxine were 8.65 and 8.32 g/L, respectively.

  • 최근 플라즈마 의학이 발달하면서 제트, 펜, 니들, 토치 등의 다양한 형태의 플라즈마 발생기가 개발되었으며 내부의 가스라인으로 가스의 종류, 유속, 조성 등을 조절하여 생물학적 효과를 극대화 할 수 있고 안정적으로 플라즈마 방전상태를 유지할 수 있으나 처리 면적이 좁아 실제 생물학적 시스템 (세포, 조직, 그리고 박테리아) 적용에 있어 한계점이 존재한다. 이러한 한계점을 극복하기 위해서 유전체격벽방전 (Dielectric barrier discharge, DBD) 방식을 이용한 플렉서블 활성종 발생기를 제작하고 생물학적 시스템에 적용하기 위한 방전 특성 평가를 진행하였으며, 간단한 in vitro 모델인 한천 젤을 이용하여 플라즈마 처리에 따른 전달물질의 침투거리를 확인하였다. 플라즈마 방전 시 생성되는 수산화기 [OH], 과산화수소 [$H_2O_2$], 초산소음이온 [$O_2{^-}$], 오존 [$O_3$], 그리고 산화질소 [$NO_x$]와 같은 산소 및 질소 활성종 (Reactive oxygen and nitrogen species, RONS)은 세포벽 또는 세포막의 주요 구성성분인 다당류와 인지질의 과산화 반응을 통해 구조를 변화시키고 생물학적 시스템의 표면의 pH를 낮춘다. 이러한 RONS의 작용은 살균, 소독 뿐만 아니라 약물의 침투를 돕는다. 일반적으로 한천 겔은 농도에 따라 생체 내 뇌 조직과 물리적 특성이 유사하고, 미생물학 기질, 방사선학 연구를 위한 조직모델로 사용되기 때문에 본 연구에서는 3%와 5% 농도의 한천 젤을 사용하여 침투거리를 확인하였다. 한천 젤은 $2.5{\times}2.5{\times}2.5cm^3$의 크기로 준비되었고 대조군으로 염료가 포함된 에멀젼을 0.01 g 도포하고, 실온에서 30분간 보존 후 단면을 잘라 현미경으로 침투거리를 확인하였으며, 실험군으로 플라즈마 전처리 후 에멀젼을 도포한 시표와 에멀젼 도포 후 플라즈마 처리한 시료에 대해 에멀젼 침투거리의 변화를 확인하였다. 본 연구의 플렉서블 활성종 발생기는 인체에 부착하여 사용되기 때문에 화상, 홍반을 유발을 방지하기위해 $40^{\circ}C$의 온도에서 실험을 진행하였고 이때에 플라즈마 방전조건은 $0.065W/cm^2$ 수준의 전력을 소모하는 1.7 kV의 전압, 16 kHz의 주파수로 10분간 처리하였다. 그 결과 3%의 한천 젤의 경우 침투거리 0.779 mm에서 0.826 mm, 0.942 mm까지 침투거리가 증가하였고 5%의 한천 젤의 경우 0.859 mm, 0.949 mm로 증가하였다. 이러한 침투거리 증가는 젤 표면의 다당류를 구성하고 있는 단량체가 플라즈마 처리시 화확적 구조가 끊어져 결론적으로 약물 침투가 증가된 것으로 판단된다.

In vivo Growth Inhibition of Sarcoma-180 Cells by a β-Glucan from the Mushroom Ganoderma lucidum (영지(Ganoderma lucidum)의 β-Glucan에 의한 Sarcoma-180 육종암 생장 억제)

  • Mushroom-derived ${\beta}$-glucan, a polysaccharide (GLP) isolated from the mycelium of Ganoderma lucidum, was previously shown to have inhibitory effects against tumor-bearing mice in vivo. We investigated the apoptotic effect of mushroom-derived ${\beta}$-glucan in a sarcoma-180 tumor cell- bearing mice model using an ELISA to determine the levels of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in the mice. The morphology of the tumor cells was assessed with transmission electron microscopy (TEM). GLP was injected into the tumor-bearing mice at a dose (i.p.) of 20 mg/kg for 10 days. After 30 days, the tumor mass from the inguinal region was collected, weighed, and assayed using TEM and a TNF-${\alpha}$ ELISA kit. The tumors that developed in the mice treated with GLP were 71.4% smaller than those in the control group, showing the ability of GLP to inhibit tumor growth. The levels of TNF-${\alpha}$ in the serum of the sarcoma-180 bearing mice were 12 times greater than in the serum of the nonbearing tumor mice. An ultrastructural study demonstrated that the GLP-treated sarcoma-180 tumor cells were condensed, with rearranged chromatin. In addition, the marginated chromatin in nucleus induced the nuclear compartment, and there were many vacuolization in the cell. GLP could be an effective apoptosis-inducing compound in sarcoma-type cancers.

Extraction and Component Sugar Analysis of Polysaccharides from Buckwheat (메밀의 다당류 추출과 구성당 분석)

  • This study was conducted to extract and isolate the polysaccharides from buckwheat. Also the sugar composition of the polysaccharides was investigated. The soluble and indigestible polysaccharides were isolated from supernatant and residue after enzyme treatment of raw, roast and steam buckwheat. The yields of low molecular weight soluble polysaccharides(LMS-P: MW<10 Kda) and high molecular weight soluble polysaccharides(HMS-P: MW>10 Kda) were $74.9{\sim}84.2%$ and $5.5{\sim}9.4%$, respectively. The yields of indigestible polysaccharides were low molecular weight insoluble polysaccharides; $0.8{\sim}4.2%$, crude hemicellulose; $3.2{\sim}9.6%$, alcohol insoluble hemicellulose; $0.9{\sim}1.7%$, residue; $2.0{\sim}2.4%$, respectively. The free sugars were detected in the soluble polysaccharides and low molecular weight insoluble polysaccharides but were not detected in the crude hemicellulose, alcohol insoluble hemicellulose and residue. The protein of all fraction were detected and the content was $1.0{\sim}18.9%$. The main sugar of soluble polysaccharides was glucose and the indigestible polysaccharides were composed of glucose, arabinose, rhamnose, xylose, mannose and galactose.

Effect of Brazilin from Caesalpinia sappan L. on the Growth of Streptococcus mutans ATCC 25175 (소목으로부터 분리된 Brazilin이 Streptococcus mutans ATCC 25175의 생장에 미치는 효과)

  • Somok, the heart wood of Caesalpinia sappan is used in traditional Chinese medicine. This study was performed to investigate the effect of growth and culture conditions of brazilin from C. sappan against S. mutans ATCC 25175. The bacteria were cultured in brain heart infusion (BHI) broth, and then incubated under 5% $CO_2$ at $37^{\circ}C$ for 18-24 hours. The effect of brazilin against S. mutans was confirmed under the changes of the culture conditions, such as growth curve and the change of pH, protein, and total carbohydrate. The growth of S. mutans in control medium was the highest at 24 hr, while brazilin-added medium (0.3 mg/ml) showed maximum growth at 32 hr. The pH values of the control medium was 5.25 at 16 hr, but the media supplemented with brazilin (0.3 mg/ml) was 7.0 at 16 hr. The amounts of total carbohydrate of the control medium was 11 mg/ml at 8 hr, but the brazilin-added media (0.3 mg/ml) was 18 mg/ml at 8 hr. In the protein change of the culture medium, the control culture broth and the brazilin supplemented-cultures was 2.4 mg/ml and 2.54 mg/ml at 24 hr, respectively. Polysaccharide contents of the control medium and test media were 3 mg/ml and 2 mg/ml at 8 hr, respectively. Thus, the application of C. sappan can be considered a useful and practical material for the prevention of dental caries.

Chemical Composition and Biological Activities of Immunostimulants Purified from Alkali Extract of Poria cocos Sclerotium (복령 균핵의 알칼리추출물에서 정제한 면역활성 증강물질의 작용과 화학구성)

  • An one percent sodium carbonate extract prepared from sclerotia of Poria cocos activated the proliferation of the T lymphocytes as measured by mixed lymphocyte responses(MLR). The active fraction, PCSC22, was isolated from an one percent sodium carbonate extract by a combination of fractionation procedures, including ethanol precipitation and chromatographies on column of DEAE-cellulose and Sephadex G50. Carbohydrate and peptide contained in PCSC22 were 78 : 22% in ratio. On employing gel filtration high performance liquid chromatography, PCSC22 exhitited a homogeneous peak with an average molecular weight of 8 kDa. The sugar moiety of PCSC22 was composed with mannose (92%), galactose (6.2%) and arabinose (1.3%), which might be indicated as heteromannan. Fifteen amino acids were found in peptide moiety of the polysaccharide and aspartic acid, serine, and valine were major components. PCSC22 activated the primary proliferation of T lymphocytes measured by mixed lymphocyte responses, the antibody production of the B lymphocytes and the secretion of nitric oxide from macrophage cell line, RAW264.7.
