영지(Ganoderma lucidum) 균사체의 액체배양에 의한 세포외 수용성 다당류의 분획 및 항암활성

  • Exo-polysaccharide (BWS) obtained from submerged cultivation of Ganoderma lucidum mycelium was fractionated. Antitumor activity of their fractions was investigated in comparison with the mycelial polysaccharide fractions. Eight kinds (BWS-DN, BWS-DA, BWS-DN-GI, BWS- DA-GI, MWS-DN, MWS-DA, MWS-DN-GI and MWS-DA-GI) of polysaccharide fractions were obtained by DEAE-cellulose ion exchange chromatography and Sepharose CL-4B gel chromatography from BWS and MWS, which were isolated from culture fiuid and mycelial cell, respectively. The anticomplementary activities (ITCH$_{50}$%) of the exo-polysaccharide fractions showing 15% to 30% were lower than those of mycelial polysaccharide fractions showing 15% to 70%. The acidic fractions of BWS-DA and BWS-DA-GI fractionated from BWS, showed the highest activity of 30%. In the MTT assay, BWS-DN and MWS against mouse leukemia L1210 exhibited high inhibition ratio of 86 and 89%, respectively at the concentration of 600 $\mu$g/ml. High inhibition ratio of 50% (IC$_{50}$) was achieved for BWS, BWS-DA and MWS-DA fractions against human colon adenocarcinoma COLO-205 and for BWS-DA, BWS-DN and MWS-DN fractions against human leukemia HL-60 at the concentra- tion of 300 $\mu$g/ml among the six polysaccharide fractions, respectively.

  • Effect of Mylabris sidae(MS) and Epicauta gorhami(EG) polysaccharide fractions on the inflammation and immune responses were studied in vivo. MS and EG contained cantharidin about 0.61 and 0.65%, respectively. It was shown that MS and EG polysaccharide fractions at a oral dose of 100 mg/kg have the significant anti-inflammatory and analgesic activity; They inhibited significantly the carrageenin-induced inflammation and acetic acid-induced writhing syndrome. They accelerated significantly the carbon clearance and the phagocytosis of colloidal carbons by Kupffer cells in liver, but they at a oral dose of 100 mg/kg suppressed significantly the Arthus reaction in the sheep red blood cell(S-RBC)-sensitized mice in accordance with the inhibition of haemaglutinin titer, haemolysin titer and plaque-forming cells. On the other hand, they at a oral dose of 200 mg/kg accelerated slightly the oxazolone-induced dermatitis in rats and delayed hypersensitivity in the S-RBC-challenzed mice in consistent with the increase of rosette forming cells. As the above results, it exhibited that MS and EG polysaccharide fractions inhibited the humoral immune responses, but they accelerated the function of macrophages and cellular immune responses. EG polysaccharide fraction had more active than MS polysaccharide fraction.

  • Toxohormone-L is a lipolytlc factor, found in ascites fluid of sarcoma 180-bearing mice and of patients with hepatoma. A substance that inhibited the lipolytic action of Toxohormone-L was isolated from Korean red ginseng powder. This substance had a pectin-like a-1,4-polygalacturonan backbone with some acetoxyl groups, and so was an acidic polysaccharide. Acidic polysaccharide was found to inhibit significantly toxohormone-L-induced lipolysis at its concentration of 10$\mu\textrm{g}$/ml.

  • This investigation was aimed at the study of the antidiabetic activity and effect on hepatic glutathione metabolism of polysaccharide from Trichosanthes kirilowii in hyperglycemic rats with aboxan (175 mg/Kg, i.p.). As the results, the polysaccharide inhibited the increase of blood glucose, triglyceride level and lactate dehydrogenase activity, but cholesterol not changed. And it increased protein bound-SH, nonprotein bound-SH, glutathione level and inhibited the decrease of glutathione S-transferase.

  • This study was conducted to investigate the extraction characteristics of the polysaccharide from Jeseng mushroom (Fomitopsis pinicola Jeseng). Yields of the polysaccharide extracted from powdered mushroom by autoclaving(120, 30 min) with water at different pH and salt concentration were 8.2~9.2% in pH 5~11, 4.7~5.5% in 1~5% salt solution, respectively. The yield by the 0.05~1.0 N KOH-extraction was ranged 3.45~13.20%, while that by HAS-extraction(homogenizing after KOH swelling) using 1~2.5 N KOH 73.6~78.4%. Content of carbohydrate, protein, lipid and ash of the crude polysaccharide extracted from fruits body and its cultured mycelium by method of water extraction, KOH extraction(0.005~1N) and HAS-extraction were ranged 86.5~92.6%, 2.3~13.1%, 0.1~4.2% and 0.1~1.7%, respectively. The polysaccharide were composed of 62.0~77.8 g/g of pentose, 138.0~187.8 g/g of hexose and 21.2~117.3 mg/g of protein. From these results, the polysaccharide extracted was supposed to be a protein-bound polysaccharide.

  • 식용버섯으로부터 항혈전제제 개발의 소재화연구로 항응고활성의 검색과정에서 높은 활성을 보였던 구름버섯에서 항응고성 다당류를 정제하여 정제다당의 화학적 특성을 검토하였다. 정제는 구름버섯의 알칼리 추출물(CV-0)을 메탄올환류, 에탄올침전 후 3개의 연속적인 DEAE-Toyopearl 650C 이온교환, Sepadex G-100 및 Sepharose CL-6B 겔여과 크로마토그래피와 HPLC에 의하여 실시하였으며 정제다당 CV-40-Va-1의 분자량은 $7.2{\times}105$으로 측정되었다. CV-40-Va-I은 구성당 잔기에 19.32%의 황산기를 함유하는 함황성다당으로서 구성당조성은 fucose:mannose:glucose:galactose가 1.00:0.22:0.20:0.11의 몰 비율로 존재하였다. CV-40-Va-1의 항응고활성은 농도의존성을 보였으며 특히 내인성경로를 통하여 혈액응고를 저해하는 것으로 확인되었다.

  • 참당귀 세포(Angelica gigas H4)를 현탁배양하여 면역 증강성 다당을 생산하기 위한 최적배지 설계 및 생산조건 최적화 연구를 수행하였다. 우선 세포의 성장에 가장 좋은 배지로서 Schenk and Hildebrandt(SH) 배지를 선택하여 배양한 결과 $25^{\circ}C$, 암배양조건에서 각각 15.8g DCW/l와 0.85 g/l의 세포 및 다당을 얻을 수 있었다. 한편 다당 생산용 배지의 개발을 위하여 여러 가지 식물세포배양용 배지 중에서 다당 생산능이 가장 높은 배지로서 Gamborg B5 배지를 선정하였으며 B5 배지를 변형하여 면역증강성 다당생산을 위한 최적배지(PPM)를 개발하였다. 그리고 최적배지를 이용한 당귀 세포증식과 다당 생산을 위한 최적 pH는 6.0-6.5, 배양온도는 $20^{\circ}C$, 그리 고 광조건보다는 암조건이 좋은 것으로 밝혀졌다. 상기 최적조건에서의 최대 세포증식과 다당생산은 각각 14.8 g/l, 1.5 g/l 이었다.

Aigeok polysaccharide의 겔화에 미치는 칼슘농도와 온도의 효과 (The effect of calcium concentration and temperature on the gelation of Aigeok Polysaccharide)

  • 정제된 Aigeok Polysaccharide의 분자량은 $5.5{\times}10^4$ Da였고, 분자량 분포는 단분산도에 가까웠으며, FT-IR 스펙트럼으로부터 메칠에스테르화된 정도는 낮은 것으로 확인되었다. Aigeok polysaccharide의 겔화 속도는 칼슘이온과 온도에 크게 영향을 받으며 4.08mM 칼슘 이온이 첨가될 때까지는 탄성률의 세기가 증가하였다가 그 이상이 되면 저장탄성률과 손실탄성률 모두 감소하였다. Aigeok polysaccharide의 화학적 구조와 junction zone은 적절한 양의 칼슘이온에 의해 안정화됨을 알 수 있었고 과량의 칼슘농도하에서는 그물구조 형성이 방해를 받아 탄성률은 감소하였다. 겔화 속도 상수는 온도가 증가함에 따라 감소하였고 이는 Aigeok 분자간에 수소결합이 주된 역할을 한다는 것을 알 수 있었다. Aigeok polysaccharide의 겔화 능력은 4.08mM의 칼슘 첨가와 $5^{\circ}C$에서 가장 안정한 겔을 형성함을 알 수 있었고, 반응식이 겔화를 추정하는데 좋은 표준이 됨을 알 수 있었다.

  • Schizophyllum commune의 균체 생육 및 다당체 생성에 미치는 여러 가지 배지성분의 영향을 조사하였다. 탄소원으로는 산업적인 경제성과 높은 다당체 생성량을 나타낸 glucose를 선택하였고, 무기질소원은 유기질소원에 비해 현저히 낮은 다당체 생성량을 나타내었으며, 그 결과 균체량과 다당체 생성량에서 높은 효과를 나타낸 yeast extract를 질소원으로 선택하였다.

  • Optimal culture conditions for efficient in vitro propagation and polysaccharide production of Orostachys japonicus were established. The highest growth yield was achieved in 1/2 MS medium, while the lowest growth yield was obtained in 4 MS medium. The patterns of polysaccharide formation were a little similar in all cases, but on MB5 medium, the po]ysaccharide contents of plant were higher than others. Among the modified nitrate levels, effective growth level were obtained in 1/4 N and 1/2 N. High contents of polysaccharide were obtained in 4 N. Different concentration of potassium and calcium did not improve the growth and polysaccharide production. The micropropagated shoots were successfully acclimatized artificial soils.
