Composition and Structural Characteristics of Polysaccharide from Hot Water Extraction of Auricularia auricula (흑목이 버섯 열수추출 다당의 성분 및 구조적 특성)

  • A hot-water extract from fruit body of Auricularia auricula was purified by ethanol precipitation and subsequent dialysis. The polysaccharide showed a typical IR spectrum similar to $\beta$-glucan and was composed of glucose and mannose in a molar ratio of 16.8:83.2 indicating that it is a glucomannan. A glucomannan-like polysaccharide was also identified from Tremella fuciformis. Both of the polysaccahrides showed the presence of acetyl groups and presented colorimetric responses for $\beta$-1,4-glucomannan indicating the acetylated $\beta$-glucomannan similar to the typical polysaccharide found in Aloes.

  • To examine the chemical properties of the polysaccharide from Phellinus linteus, we obtained the polysaccharide treated by the alkali and hot water. The carbohydrate and protein contents of each polysaccharide 81.1%, 37.4% and 6.2%, 21.8%, respectively. The carbohydrate consisted of six monosaccharides and the protein contained 16 amino acids.

  • An acidic polysaccharide H-III was extracted from fruiting bodies of Lyophyllum shimeji with hot water. Acid hydrolysis and gas chromatography analysis showed that the polysaccharide was almost exclusively composed of glucose with a very small amount of mannose and galactose. Uronic acid of 8.36% was also detected in H-Ill. Its molecular weight was estimated to be $1{\times} 10^6 Da.\;By\;^{13}C-NMR$ analysis, some repeating units of disaccharide were detected in the polymer H-III. The polysaccharide showed a strong mitogenic activity in a dose-dependent manner.

  • Effects of Allolobophora caliginosatrapezoides (Ac) polysaccharide fractions on the inflammation and hypersensitivity were studied in vivo. It showed that Ac polysaccharide fractions have the significant inhibitory activities of inflammation and hypersensitivity; They inhibited significantly the carrageenin-induced paw edema and acetic acid-induced writhing syndrome. They also inhibited significantly the Arthus reaction and delayed hypersensitivity in the sheep red blood cell-sensitized mice in accordance with the inhibition of haemaglutinin titer, haemolysin titer, plaque-forming cells and rosette-forming cells. They also improved markedly the oxazolone-induced dermatitis in rats dose-dependently. As the above results, it exhibited that Ac polysaccharide fraction inhibited not only humoral immune response, but also cell-mediated immune response. It seemed that methanol and ether extracts have also another physiological active agents.

Enhancement of Anti-tumorigenic Polysaccharide Production, Adhesion, and Branch Formation of Bifidobacterium bifidum BGN4 by Phytic Acid

  • The polysaccharide (BB-pol) extracted from Bifidobacterium bifidum BGN4 showed growth inhibitory effects on several colon cancer cell lines such as HT-29 and HCT-116. To increase the yield of polysaccharide, B. bifidum BGN4 was cultured in various culture media with different compositions. When B. bifidum BGN4 was cultured in modified MRS broth containing phytic acid, the cells showed increased branch formation and enlarged morphology. The content of total carbohydrate and the ability of adhesion to intestinal epithelial cells were also increased by phytic acid. The polysaccharide obtained from the cells grown in the presence of phytic acid inhibited the proliferation of cancer cell lines such as HT-29 and MCF-7 cells but not normal colon cell line, FHC. Taken together, Bifidobacterium grown in the presence of phytic acid may confer enhanced beneficial function for the host.

  • A bacterium synthesizing extracellular polysaccharide was isolated from soil and identified as Enterobacter agglomerans. The polysaccharide was found to be glucan polymer containing glucose and galactose in a molar ratio of 1 : 1.1. The aqueous solution was very viscous. The viscosity of 1% solution was 264 mPa.s. at $42\;sec^{-1}$ and yield stress was 4.89 Pa. The polysaccharide solution did not have thermal stability but pH and salt stability.

  • In the previous report, we described the marked antitumor and immunomodulatory activities of PVp, a protein-polysaccharide fraction of a Korean wild mushroom Psathyrella velutina. In this study, a protein-polysaccharide fraction, PVMP, was prepared from the shake-cultured mycelia of the same mushroom and its immunoactivities as well as chemical compositions were investigated. At 200 $\mu\textrm{g}$/ml, PVMP weakly stimulated the BALB/c mouse splenic lymphocytes to form lymphoblasts and upregulated the expression of CD25 molecules, but failed to stimulate peritoneal macrophages. In chemical analysis these two protein-polysaccharide fractions were found to be quite different in that the carbohydrate contents of PVMP and PVP, respectively, was 85.3% and 41.2%. These results reveals that PVMP, unlike PVP, is a moderate immunostimulator on the immune system.

Studies on the Blood Anticoagulant Polysaccharide Isolated from Hot Water Extracts of Hizikia fusiforme (톳 열수추출물로부터 분리한 혈액 항응고성 다당류에 관한 연구)

  • This study was focused on the purification, characterization and promotion mode of an anticoagulant polysaccharide from Hizikia fusiforme. The anticoagulant crude polysaccharide(HF 0) was obtained by using hot water extraction at 100oC for 3 hrs after homogenizing desalted Hizikia fusiforme. The anticoagulant polysaccharide(HF 2 3 1a) was purified from the crude extract(HF 0) through stepwise gradient ethanol precipitation(HF 2), DEAE Toyopearl 650C(HF 2 3), Sephadex G 75(HF 2 3 1), Sepharose CL 6B(HF 2 3 1a) chromatography and HPLC to homogeneity. HF 2 3 1a was estimated at 5.3$\times$105 Da molecular weight and composed of fucose(51.92%), galactose(19.34%), mannose(13.92%), xylose (7.14%), arabinose(3.95%) and rhamnose(3.78%), and comprimised 29.7 % sulfate residue. The sulfated anticoagulant polysaccharide from HF 2 3 1a was proposed to inhibit via the intrinsic pathway and common pathway in the blood coagulation. The HF 2 3 1a exhibited the anticoagulant activity by activating an antithrombin III and the activity depended on the concentration of HF 2 3 1a. Acute toxicity of HF 2 in mice was not detected. Only 14 of 33 control mice(11.4%) that had taken saline survived for 30 min after injecting thrombin(100 NIH unit/ml).

Polysaccharide from Polygonatum Inhibits the Proliferation of Prostate Cancer-Associated Fibroblasts Cells

  • Inhibition of cancer-associated fibroblasts (CAFs) may improve the efficacy of cancer therapy. Polysaccharide extracted from polygonatum can selectively inhibit the growth of prostate-CAFs (p<0.001) without inhibiting the growth of normal fibroblasts (NAFs). Polysaccharides from polygonatum stimulate autophagy of prostate-CAFs. 3-methyl-adenine(3-MA) is an autophagy inhibitor. 3-MA was added to prostate-CAFs with polysaccharide from polygonatum to determine whether autophagy plays an important role in the restrained effect. Finally, polysaccharide from polygonatum treatment significantly increased the activation of Beclin-1 and LC3, key autophagy proteins. Polysaccharide from polygonatum stimulates autophagy of prostate-CAFs and inhibits prostate-CAF growth, indicating that a novel anti-cancer strategy involves inhibiting the growth of prostate-CAFs.

Quality Characteristics of Mixed Polysaccharide Gels with Various Kiwifruit Contents by Freeze-Thawing (키위 첨가 다당류 흔합겔의 냉동ㆍ해동에 따른 품질 특성)

  • This study was performed to determine the quality characteristics of mixed polysaccharide gels made from kcarrageenan and locust bean gum with various kiwifruit content after freeze-thawing. The syneresis, rupture properties, TPA properties and sensory properties of the mixed polysaccharide gels with various kiwifruit contents after freeze-thawing were measured. The syneresis of the gel with various kiwifruit contents by freeze-thawing was not significantly different, whereas repeating freeze-thawing resulted in remarkable increase in the syneresis of the mixed polysaccharide gels with 5%, 20% and 40% kiwifruit. In terms of the rupture properties, the rupture stress, rupture energy and rupture strain decreased with increasing kiwifruit content after freeze-thawing. In terms of the TPA properties, the adhesiveness, hardness and chewiness increased and the cohesiveness decreased with increasing kiwifruit content after freeze-thawing. The rupture properties, springiness and cohesiveness of the gel by freeze-thawing were lower than those of the gel stored in the refrigerator. In contrast, the gumminess, hardness and chewiness of the gel after freeze-thawing were higher than those of the gel stored in the refrigerator. The results showed that the gel became crumbly and tough as a result of freeze-thawing. In the sensory evaluation, He overall acceptability of the gel after freeze-thawing was highest at the 30% kiwifruit content, and differences in the sensory properties between the gel after freeze-thawing and that stored in the refrigerator were small. Therefore, mixed polysaccharide gels with kiwifruit after freeze-thawing could be useful despite the small decrease in quality.
