Extraction Characteristics of Polysaccharide from Fomitopsis pinicola Jeseng Mushroom (소나무 재생버섯(Fomitopsis pinicola Jeseng) 다당류의 추출 특성)

  • This study was conducted to investigate the extraction characteristics of the polysaccharide from Jeseng mushroom (Fomitopsis pinicola Jeseng). Yields of the polysaccharide extracted from powdered mushroom by autoclaving(120, 30 min) with water at different pH and salt concentration were 8.2~9.2% in pH 5~11, 4.7~5.5% in 1~5% salt solution, respectively. The yield by the 0.05~1.0 N KOH-extraction was ranged 3.45~13.20%, while that by HAS-extraction(homogenizing after KOH swelling) using 1~2.5 N KOH 73.6~78.4%. Content of carbohydrate, protein, lipid and ash of the crude polysaccharide extracted from fruits body and its cultured mycelium by method of water extraction, KOH extraction(0.005~1N) and HAS-extraction were ranged 86.5~92.6%, 2.3~13.1%, 0.1~4.2% and 0.1~1.7%, respectively. The polysaccharide were composed of 62.0~77.8 g/g of pentose, 138.0~187.8 g/g of hexose and 21.2~117.3 mg/g of protein. From these results, the polysaccharide extracted was supposed to be a protein-bound polysaccharide.

Immunomodulating Activity of Crude Polysaccharide from Inonotus obliquus Sclerotia by Fractionation including MeOH Reflux

  • To obtain the immunomodulating polysaccharide from chaga mushroom (Inonotus obliquus sclerotia, IO), crude polysac- charide fractions (IO-M-CP and IO-CP, respectively) prepared from hot-water extract (IO-W) of I. obliquus by EtOH precipitation after MeOH reflux or not. After IO-W was re-dissolved in water followed by EtOH addition in the case without MeOH reflux, EtOH mixture was fractionated into EtOH-soluble (IO-E) and crude polysaccharide (IO-CP). In the meanwhile, MeOH-soluble fraction (IO-M) was separated from IO-W after MeOH reflux. The residue was dissolved in water and was added by EtOH, and then EtOH mixture was also fractionation into EtOH-soluble (IO-M-E) and crude polysaccharide (IO-M-CP). As a result of the macrophage stimulating activity of these fractions, IO-CP and IO-M-CP showed significantly increased cell proliferation and cytokines production than IO-W. Particularly, IO-M-CP promotes the production of IL-12 more than IO-CP. In the splenocytes proliferating activity and intestinal immune system modulating activity through Peyer's patch, both of 2 crude polysaccharide fractions were significantly promoted in cell proliferation and cytokines production than IO-W, and IO-M-CP was more potent than IO-CP in IL-2 production from splenocytes and GM-CSF production ($10{\mu}g/mL$) in Peyer's patch cells. In addition, immunomodulating polysaccharide fractions (IO-M-CP and IO-CP) prepared from IO-W by EtOH precipitation with or without EtOH reflux showed no significant difference in the chemical composition and component sugar. These results suggested that MeOH reflux might exclude low-molecular weight materials from IO-W and consequently increase the immunomodulating activity of IO-M-CP. Therefore, it was confirmed that immunomodulation of polysaccharide prepared from hot-water extract of chaga mushroom was enhanced by fractionation including MeOH reflux and EtOH precipitation.

  • Schizophyllum commune belongs to Basidiomycetes is able to produce an extracellular polysaccharide, beta-glucan, which has been well known as an effective immune stimulator. In other to obtain the optimum conditions for the polysaccharide production, Schizophyllum commune was cultivated in different cultivation media containing various carbon and nitrogen sources. For the cell growth and the polysaccharide production, the optimum synthetic medium was developed with YMP medium an a basal midium. The flask culture conditions for the polysaccharide production were $27^{\circ}C$ and 150rpm with the initial pH 5.5.

  • A bacterium synthesizing extracellular polysaccharide was isolated from soil and identified as Enterobacter agglomerans. The polysaccharide was found to be glucan polymer containing glucose and galactose in a molar ratio of 1 : 1.1. The aqueous solution was very viscous. The viscosity of 1% solution was 264 mPa.s. at $42\;sec^{-1}$ and yield stress was 4.89 Pa. The polysaccharide solution did not have thermal stability but pH and salt stability.

  • Effects of Allolobophora caliginosatrapezoides (Ac) polysaccharide fractions on the inflammation and hypersensitivity were studied in vivo. It showed that Ac polysaccharide fractions have the significant inhibitory activities of inflammation and hypersensitivity; They inhibited significantly the carrageenin-induced paw edema and acetic acid-induced writhing syndrome. They also inhibited significantly the Arthus reaction and delayed hypersensitivity in the sheep red blood cell-sensitized mice in accordance with the inhibition of haemaglutinin titer, haemolysin titer, plaque-forming cells and rosette-forming cells. They also improved markedly the oxazolone-induced dermatitis in rats dose-dependently. As the above results, it exhibited that Ac polysaccharide fraction inhibited not only humoral immune response, but also cell-mediated immune response. It seemed that methanol and ether extracts have also another physiological active agents.

  • An acidic polysaccharide H-III was extracted from fruiting bodies of Lyophyllum shimeji with hot water. Acid hydrolysis and gas chromatography analysis showed that the polysaccharide was almost exclusively composed of glucose with a very small amount of mannose and galactose. Uronic acid of 8.36% was also detected in H-Ill. Its molecular weight was estimated to be $1{\times} 10^6 Da.\;By\;^{13}C-NMR$ analysis, some repeating units of disaccharide were detected in the polymer H-III. The polysaccharide showed a strong mitogenic activity in a dose-dependent manner.

  • A hot-water extract from fruit body of Auricularia auricula was purified by ethanol precipitation and subsequent dialysis. The polysaccharide showed a typical IR spectrum similar to $\beta$-glucan and was composed of glucose and mannose in a molar ratio of 16.8:83.2 indicating that it is a glucomannan. A glucomannan-like polysaccharide was also identified from Tremella fuciformis. Both of the polysaccahrides showed the presence of acetyl groups and presented colorimetric responses for $\beta$-1,4-glucomannan indicating the acetylated $\beta$-glucomannan similar to the typical polysaccharide found in Aloes.

  • Water-soluble crude polysaccharide (TM-1) prepared from the leaves of Teucrium viscidum var. miquelianum was fractionated into three polysaccharide fractions, TM-2, TM-3 and TM-4 by the addition of cetyltrimethylammonium bromide. The major polysaccharide fraction, TM-2, consisted of glucose, galactose, rhamnose, mannose, glucuronic acid and galacturonic acid, and all fractions contained galactose and glutose as the major neutral sugars. TM-4 showed the highest anti-complementary activity. When TM-4 was futher fractionated by anion exchange chromatography, TM4- II a and TM4-II b showed the most Potent anti-complementary activity. TM4- II a was composed mainly of galactose, arabinose and glucose in the molar ratios of 2.13 : 0.94 : 1.00 respectively, and contained a small amount of galacturonic acid and glucuronic acid.

  • In the previous report, we described the marked antitumor and immunomodulatory activities of PVp, a protein-polysaccharide fraction of a Korean wild mushroom Psathyrella velutina. In this study, a protein-polysaccharide fraction, PVMP, was prepared from the shake-cultured mycelia of the same mushroom and its immunoactivities as well as chemical compositions were investigated. At 200 $\mu\textrm{g}$/ml, PVMP weakly stimulated the BALB/c mouse splenic lymphocytes to form lymphoblasts and upregulated the expression of CD25 molecules, but failed to stimulate peritoneal macrophages. In chemical analysis these two protein-polysaccharide fractions were found to be quite different in that the carbohydrate contents of PVMP and PVP, respectively, was 85.3% and 41.2%. These results reveals that PVMP, unlike PVP, is a moderate immunostimulator on the immune system.

  • To examine the chemical properties of the polysaccharide from Phellinus linteus, we obtained the polysaccharide treated by the alkali and hot water. The carbohydrate and protein contents of each polysaccharide 81.1%, 37.4% and 6.2%, 21.8%, respectively. The carbohydrate consisted of six monosaccharides and the protein contained 16 amino acids.
