Comparative Study of Food Components and Sensory Properties of Common Porphyra yezoensis and Functional Porphyra yezoensis (일반 김과 기능성 김의 식품성분과 관능평가 비교 분석에 관한 연구)

  • This study is to compare food components and the sensory properties of common and functional Porphyra yezoensis, Pyroligneous liquor, Salicornia herbacea L. and Salicornia herbacea L. treated with oak charcoal. The samples used in this study were provided by a local manufacturer. We analyzed their proximate composition, mineral content, heavy metal content and amino acids. We conducted the sensory evaluation before and after grilling Porphyra yezoenis. Common components are more plentiful in functional Porphyra yezoenis than in common except for carbohydrate and crude fiber. In the case of mineral content, generally functional Porphyra yezoensis contains much more mineral than common except for magnesium and zinc. Especially Salicornia herbacea L. treated with oak charcoal contains abundant iron. Also, Salicornia herbacea L. contains a lot of calcium potassium, copper and sodium. There is a little more heavy metal except for chromium, cadmium and lead in Salicomia herbacea L. treated with oak charcoal than in common that contains a lot of chromium, cadmium and lead, although a wide difference was not discovered between them. In the case of composed amino acid, there are much glutamic acid, glycine, isoleucine and phenylalanine in both Porphyra yezoensis. Especially alanine and phenylalanine are trebled in functional Porphyra yezoensis. Also, there are far more serine and lysine in Salicornia herbacea L. and Salicornia herbacea L. treated with oak charcoal than in common Porphyra yezoensis and Pyroligneous liquor. In the case of the sensory properties, there isn't a big difference before grilling Porphyra yezoensis; however, after grilling, most functional Porphyra yezoenis had good sensory properties results, especially Salicornia herbacea L. were better than Pyroligneous liquor.

Relationship between Phycoerythrin and Nitrogen Content in Gloiopeltis furcata and Porphyra yezoensis

  • Seasonal changes of pigment compositionin two intertidal red algae, Gloiopeltis furcata (Postels et Ruprecht) J. Agardh, and Porphyra yezoensis Ueda, were investigated. Chlorophyll α and phycoerythrin levels were high during winter, but decreased in late spring or summer, with accompanying discoloration from deep red to green or yellow. This discoloation corresponded closely to the fluctuationof phycoerythrin content. Nevertheless, photosynthesis capacity was maintained by the increasing water temperature in the field, suggesting that large amounts of phycoerythrin are not necessary for photosynthesis. Phycoerythrin conten correlated significantly with nitrogen content in both species when the nitrogen level was greater than the level of critical content (1.30% DW in G. furcata, and 2.26% DW in P. yezoensis), indicating that phycoerythrin plays a more important role in the nitrogen status as a nitrogen pool than that of nitrogen critical content. Furthermore, the dependence level of the alage on phycoerythrin as a nitrogen pool was greater in P. yezoensis than in G. furcata because of the remarkable increase of phycoerythrin content in P. yezoensis with increasing jnitrogen content.

Toxicity of Cryoprotectants to Gametophytic Thalli of Red Algae Porphyra yezoensis

  • We assessed the toxicity of cryoprotectant agents (CPAs) to gametophytic thalli of red alga Porphyra yezoensis at room temperature. The CPAs used were: dimethyl sulfoxide (DMSO), ethylene glycol (EG), glycerol (GC), 1,2-butanediol (1,2-BD), 1,3-butanediol (1,3-BD), 2,3-butanediol (2,3-BD), 1,3-propanediol (1,3-PD) and propylene glycol (PG). CPA concentrations of 10, 15, 20, 25, 30, 35, 40, 45, and 50% were employed with 30 or 60 s immersion. The toxicity of the eight CPAs to gametophytic thalli of P. yezoensis was in the order: 1,3-BD < DMSO ${\approx}$ 2,3-BD ${\approx}$ PG ${\approx}$ EG < GC < 1,3-PD ${\approx}$ 1,2-BD. All thalli were more sensitive to high CPA concentrations, and most (>75%) thalli survived exposure to 10-25% CPA for 60 s. These data will facilitate selection of the optimal cryoprotectant concentration for cryopreservation of P. yezoensis thalli.

  • During in door or outdoor mass culture, Porphyra have been easily contaminated with bacteria, protozoa and microalgal species. Several axenic treatments for Porphyra thalli have been published (Polne-Fuller and Gibo 1984; Chen and McCracken 1993), but axenic techniques for neutral spores and conchocelis we not developed. DNA Sequences and Identification of Porphyra Cultivated by Natural Seeding on the Southwest Coast of Korea (한국 남서해안 자연채묘 양식 김의 DNA 염기서열과 종 동정)

  • Nuclear SSU and ITS1 rDNA and plastid rbcL sequences were determined to identify the seven samples of Porphyra cultivated by means of natural seeding on the southwest coast of Korea and analyzed to access the phylogenetic relationships of them with the natural populations of P. tenera and P. yezoensis from Korea and Japan. SSU, rbcL and ITS1 data from 18, 21 and 31 samples, respectively, including previously published sequences were investigated in the study. Results from our individual and combined data indicated that the seven samples were all P. yezoensis and the entities except one from Muan 2 aquafarm strongly grouped together with the natural populations of P. yezoensis from the south and the west coast of Korea. The sample from Muan 2 seems to be derived from a strain of P. yezoensis introduced from Japan by Porphyra farmers, based on DNA sequence data.

A Simple Screening Method for Anti-attachment Compounds Using Monospores of Porphyra yezoensis Ueda

  • We measured the anti-attachment activity of allelochemical and antifouling substances using monospores from Porphyra yezoensis Ueda as an assay. Methanol or aqueous extracts ($20/{\mu}g/mL$) from 32 seaweeds were added to monospore suspensions. Methanol extracts of Corallina pilulifera, Ishige sinicola, Sargassum horneri, and Sargassum sagamianum inhibited attachment $by\;>90\%$ compared to the reference. Phenolic compounds fractionated from S. sagamianum caused the most potent inhibition. P. yezoensis monospores also showed significant sensitivity to known allelochemical and algicidal compounds.

Construction and Analysis of cDNA Library from Porphyra yezoensis (방사무늬 김의 cDNA Library 제조 및 분석)

  • As an attempt to preserve resources in marine biological organisms, we first constructed a cDNA library from the red alga Porphyra yezoensis. The library construction method from P. yezoensis consists of three steps; those include protoplast presparation, RNA isolation, and phage library construction. Protoplast was prepared in order to remove much of the carbohydrate compounds which are characteristics of algal cell walls. Carbohydrate contamination in the purified RNA may inhibit further enzyme reactions, those carbohydrates should be removed. RNA samples prepared from protoplast still seemed to contain residual amount of carbohydrate because mRNA isolation with conventional method failed. We therefore developed a method with Poly ATtract mRNA isolation system. The constructed phage library was tested by analyzing cDNA insert in phage vector from randomly picked ten independent white plagues. All of the phages contained cDNA inserts with sizes ranging 0.5kb and 2.0kb.
