Serum Levels of Xanthine Oxidase Activities in Cyclohexanone-Treated Rats Pretreated with Carbon Tetrachloride

  • To investigate an effect of cyclohexanone (CHO) treatment on the serum levels of xanthine oxidase (XO) in liver damaged animals, the rats were intraperitoneally pretreated with 50% carbon tetrachloride ($CCl_4$) in olive oil (0.1 mL/ 100 g body weight) 14 times every other day. To the $CCl_4$-pretreated rats, CHO (1.56 g/kg body weight) was injected once and then the animals were sacrificed at 4 hours after CHO treatment. The increasing rate of serum and liver XO activities to the control was higher in CHO-treated animals pretreated with $CCl_4$ than the $CCl_4$-pretreated those. Concomitantly CHO injection to the $CCl_4$-pretreated animals showed somewhat higher Vmax and lower Km value in the kinetics of liver XO enzyme. Furthermore, increasing rate of hepatic malonedialdehyde content to the control was also higher in CHO-treated animals pretreated with $CCl_4$ than $CCl_4$-pretreated those. On the other hand, the injection of CHO to the $CCl_4$-pretreated animals showed the more enhanced liver damage on the basis of liver function finding; liver weight per body weight (%), serum levels of alanine aminotransferase activity and hepatic glucose-6-phosphatase activity. In conclusion, injection of CHO to the $CCl_4$-pretreated rats led to more increased activity of serum XO and it may be caused by acceleration of hepatocyte membrane permeability and induction of enzyme protein.

Effect of Carbon Tetrachloride Administration on the Serum and Liver Xanthine Oxidase Activity in Ethanol-Pretreated Rats (Ethanol을 전처리한 흰쥐의 간 및 혈청 Xanthine Oxidase 활성에 미치는 사염화탄소의 영향)

  • In the present study, the comparison of liver damage in carbon tetrachloride (CCl$_4$)-treated rats with that those pretreated with ethanol and an effect of liver injury on the serum and liver xanthine oxidase (XOD) activity were evaluated. The increasing rate of liver weight per body wt., the levels of serum alanine aminotransferase, and the decreasing rate of hepatic glucose-6-phosphatase activity and the protein contents in the liver cell were higher in carbon tetrachloride-treated animals pretreated with ethanol than the carbon tetrachloride-treated group. Especially, the histopathological findings also showed more severe liver damage in the ethanol-pretreated rats than the rats treated with carbon tetrachloride only. In such a experimental condition the xanthine oxidase activity of serum and liver both of carbon tetrachloride-treated rats and those pretreated with ethanol were higher than that of each control group. And the increasing rate of xanthine oxidase enzyme activity to the control group was higher in carbon tetrachloride-treated group pretreated with ethanol than those treated with CCl$_4$. In addition, the heptic uricase activity and the serum levels of uric acid were more increased in carbon tetrachloride-treated group pretreated with ethanol than those in the CCl$_4$-treated rats. On the other hand, there were no statistical differences in hepatic catalase and glutathione S-transferase activities between the CCl$_4$-treated rats and those pretreated with ethanol. In conclusion, it is assumed that the more severe liver damage in ethanol pretreated rats would be due to oxygen free radical produced by the xanthine oxidase system.

  • These studies were carried out to investigate the effect of Jungchuntang water extract on the inhibitory contractile action of acetylcholine in rat. The results of these studies were as follows; 1. Contractile force of acetylcholine from trachea smooth muscle in rat was significantly inhibited by Jungchuntang. 2. Dose-response of acetylcholine pretreated Jungchuntang in rat was not significantly changed. 3. Inhibitory contractile action of acetylcholine pretreated propranolol in rat was decreased. 4. Inhibitory contractile action of acetylcholine pretreated indomethacin was not significantly changed by Jungchuntang. 5. Inhibitory contractile action of acetylcholine pretreated methylene blue was significantly changed by Jungchuntang. 6.   • This study was carried out to investigate the effects of Gooboeum extract on the inhibitory contractile action of acetylcholine in the control and sensitized rat. The results were obtained as follows: 1. The acetylcholine contractile force of the trachea smooth muscle with epithelium was significantly relaxed by Gooboeum. 2. Dose-response of acetylcholine from the trachea smooth muscle pretreated Gooboeum was not changed. 3. Effect of Gooboeum on the inhibitory contractile action of trachea smooth muscle pretreated propranolol was not significantly inhibited. 4. The inhibitory contractile action of acetylcholine of trachea smooth muscle pretreated indomethacin was not significantly changed by Gooboeum. 5. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of the trachea smooth muscle pretreated propranolol was significantly inhibited. 9. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of the trachea smooth muscle pretreated indomethacin decreased. 10. Effects of Gooboeum extract on the inhibitory contractile action of acetylcholine of the trachea smooth muscle pretreated methylene blue was not significant.

Study on the Effects of Fructus Schizandrae on the Cerebral Blood Flow and Blood Pressure (뇌혈유와 혈압에 미치는 오미자의 효능에 대한 연구)

  • The purpose of this present study is to measure the changes of regional cerebral blood flow(rCBF) and blood pressure(BP) in rats. following the intravenous injection of drug. The measurement was continually monitored by laser-doppler flowmeter (Transonic Instrument, USA) and pressure tranducer(Grass, USA) in anesthetized adult Sprague-Dawley rats about for 2h to 2h and a half through the data acquisition system composed of MacLab and Macintosh computer. The result of this experiment was as following. 1. Fructus Schizandrae increased the changes of rCBF in rats, significantly. 2. The rCBF of Fructus Schizandrae did not change by pretreated propranolol. 3. The rCBF of Fructus Schizandrae did not change by pretreated atropine. 4. The rCBF of Fructus Schizandrae did not change by pretreated I-NNA. 5. The rCBF of Fructus Schizandrae was decreased by pretreated methylene blue. 6. The rCBF of Fructus Schizandrae did not change by pretreated indomethacin. 7. Fructus Schizandrae decreased the changes of blood pressure significantly. 8. The BP of Fructus Schizandrae did not change by pretreated propranolol. 9. The BP of Fructus Schizandrae did not change by pretreated atropine. 10. The BP of Fructus Schizandrae did nol change by pretreated I-NNA. 11. The BP of Fructus Schizandrae was decreased by pretreated methylene blue. 12. The BP of Fructus Schizandrae did not change by pretreated indomethacin. These results indicate that Fructus Schizandrae can increase the rCBF and decrease the BP, that related to guanylyl cyclase activity.

Protective Effects of Extracts of Diospyrus kaki Folium against Hepatotoxicity in Carbon Tetrachloride Intoxicated Rats (감잎 추출물이 사염화탄소로 유발된 흰쥐의 간손상에 미치는 효과)

  • This study was done to investigate the protective effects of Diospyrus kaki Folium on carbon tetrachloride intoxicated rats. Male Sprague-Dawley rats (200~230g) was used. Experimental groups were divided into normal group and $CCl_{4}$-treated group and then $CCl_{4}$-treated groups were divided into 7 groups: only $CCl_{4}$-treated group, methanol extract pretreated group, butanol fraction pretreated group and $H_{2}O$ fraction pretreated group, $CCl_{4}$ treated groups were injected with $CCl_{4}$ 0.6 mg/kg (ip). The activities of ananine aminotransferasein hexane, chloroform and $H_{2}O$ fraction pretreated groups were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group and also activity of aspartate aminotransferase and hepatic malondialdehyde content in $H_{2}O$ fraction pretreated group were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group. The activity of alkaline phosphatase in chloroform and $H_{2}O$ fraction pretreated groups were decreased compared to the only $CCl_{4}$-treated group but the difference was not statistically significant. The activity of ${\gamma}$-glutamyltranspeptidase in hexane, chloroform and ethylacetate fraction pretreated groups were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group. The contents of cholesterol and triglyceride were not influenced in any fraction pretreated groups, but HDL-cholesterol content in hexane and chloroform fraction pretreated groups were significantly increased (p<0.05) compared to the only $CCl_{4}$-treated group. Therefore, these results demonstrate a possible hepatoprotective role of $H_{2}O$ fraction of Diospyrus kaki Folium methanol extract in carbon tetrachloride intoxicated rats.

Fish Jelly Forming Ability of Pretreated and Frozen Common Carp and Conger Eel (전처리하여 동결한 잉어 및 붕장어의 어묵원료적성)

  • Changes in fish jelly forming ability of pretreated and frozen common carp and conger eel were examined. Four kinds of pretreated samples were prepared such as round, mechanically separated meat using fishmeat separator, chopped and water washed meat and fish meat paste. The quality of pretreated samples and fish jelly products made from pretreated sample were generally in good condition for three months of frozen storage at $-30^{\circ}C$. Judging from fish jelly forming ability, the round state of common carp was superior to other pretreated samples during frozen storage. In case of conger eel, fish meat paste revealed totter jelly forming ability than any other pretreated samples during frozen storage.

  • The aim of this study was to determine the shear bond properties of four dentin bonding systems to internal cervical dentin, and to investigate the effect of the pretreatment for removing smear layer and position of dentin on shear bond strength of dentin bonding agents. The materials tested in this study were consisted of four commercially available dentin bonding systems[Allbond 2(AB), Clearfil Linerbond 2(CL), Optibond FL(OP), Scotchbond Multi-purpose(SB)], a restorative light-cured composite resin[Z100]J and a chelating agent[RC-prep(RC)]. Fifty-six freshly extracted human molars were used in this study. Dentin specimens were prepared by first cutting the root of the tooth 1mm below the cementoenamel junction with a diamond bur in a high speed handpiece under air-water coolant, and then removing occlusal part at pulp horn level by means of a second parallel section, The root canal areas were exposed by means of cutting the dent in specimens perpendicular to the root axis. Dentin specimens were randomly assigned to two groups(pretreated group, not-pretreated group) based on the pretreatment method of dentin surface. In pretreated group, RC was applied to dentin surface for 1minute and then rinsed with NaOCl. In not-pretreated group, dentin surface was rinsed with saline Each groups were subdevided into four groups according to dentin bonding systems. Four dentin bonding systems and a restorative resin were applied according to the directions of manufacturer. The dentin-resin specimens were embedded in a cold cure acrylic resin, and were cut with a low speed diamond saw to the dimension of $1{\times}1mm$. The cut specimens were divided into three groups according to the position of internal cervical dentin. The shear bond properties of dentin-resin specimens were measured with Universal testing machine (Zwick, 020, Germany) with the cross head speed of 0.5mm/min. From this experiment. the following results were obtained : 1. In case of shear bond strength, there was no significant difference among dentin bonding systems in not-pretreated groups, whereas in pretreated groups, the shear bond strengths of AB and of SB were statistically significantly higher than those of CL and of OP. 2. The shear bond strengths of AB and of SB in pretreated groups were significantly higher than those in not-pretreated groups. 3. The shear bond strengths of radicular layer of OP were higher than those of occlusal layer of OP in not-pretreated groups, and of AB in pretreated groups. The shear bond strengths of radicular layer of AB and of CL in not-pretreated groups were higher than those in pretreated group.

The plasma concentration of diltiazem pretreated with quercetin(pretreated group) were increased significantly (p<0.01) compared to that of control, but those of diltiazem combined with quercetin(combined group) were not affected.
