  • The effects of pulp beating degree and bulky promotor concentration on the properties of handsheet including a bulk and strength have been investigated during manufacturing of low density paper (high bulk paper) with cationic fatty acid bulky promotor. It was found that paper bulk increased with increasing cationic fatty acid bulky agent concentration, while tensile strength decreased. On the other hand the opacity of handseets also increased with increasing cationic fatty acid bulky agent concentration, while brightness decreased slightly. With increasing pulp beating degree, paper bulk as well as tensile strength increased, while opacity decreased. Brightness did not show a significant difference with increasing pulp beating degree. The highest bulk and strength values were observed when 1.5% (SwBKP) and 2% (HwBKP) of bulky promotor was treated into the 450 mL CSF pulp slurry.

Overproduction and High Level Secretion of Glucose Oxidase in Saccharomyces cerevisiae (Glucose Oxidase의 Saccharomyces cerevisiae에서의 대량생산 및 고효율 분비)

  • The overproduction and high level secretion of Glucose Oxidase (GOD) from A. niger in S. cerevisiae was carried out by cloning GOD gene. For this purpose, using two different strong promoters (ADH1 promoter, GAL10 promoter) and signal sequences (${alpha}$-MF signal sequence of S. cerevisiae and ${alpha}$-amylase signal sequence of A. oryzae) and GAL7- and GOD terminator, four expression vectors were constructed. All the expression vectors were transformed in S. cerevisiae 2805 using auxotroph method. By the flask culture, transformants of pGAL expression vector series containing GAL 10 promotor showed much higher GOD productivity than transformants of pADH expression vector series containing ADH1 promoter Transformants of pGALGO2 containing GAL10 promotor and ${alpha}$-amylase signal sequence has shown the best productivity of GOD ($GOD_{total}$: 10.3 unit/mL, $GOD_{ex}$: 8.7 unit/mL) at 115 hr. This value was three fold higher than that of pGALGO1 containing GAL 10 promotor and ${alpha}$-MF signal sequence, even if the same promotor was involved. Through the ${alpha}$-amylase signal sequence of A. oryzae, GOD was secreted much more than the case of ${alpha}$-MF signal sequence from S. cerevisiae. These results suggest that signal sequence may play a important roles in not only the secretion but also the overproduction of foreign protein. Secretion rate of GOD in pGALGO1 and pGALGO2 was 89% and 84%, respectively, Because of the overglycosylation in S. cerevisiae the molecular weight of recombinant GOD in S. cerevisiae was much larger (250 kDa) than that of nature GOD in A. niger (170 kDa).

  • We have prepared the silicone sealant using non silicone oil without silicone oil for solvent and using adhesion promotor against various substrate. We are replaced the silicone oil by the mineral oil and the normal adhesion promotor by mixed adhesion promotor at the multi purpose silicone sealant.

  • Objectives : Sasang Constitutional Medicine is based on the diversity of human beings and medically developing a variation of responses to diseases and medicines. This diversity is categorized into four concerning morphology, physiology, pathology, and pharmacology. It is supposed that sasang constitutional medicine is related the genetic diversity of individuals. Single nucleotide polymorphism is the basic tool to research genetic polymorphism as a landmark of genomes. Each constitution has different processes of pathophysiology and metabolisms to herb medications. In clinical research, the stroke incidence is significantly different by constitution. Methods : We researched whether the polymorphic expression of CCK (rs=2241997) depends on sasang constitution. The [c/t] polymorphism site of promotor region of CCK gene on 3p22-p21.3 was investigated. Results : The allele frequency of [c/t] polymorphism of CCK promotor region was different in constitution groups compared to the average allele frequency of SNP DB. The allele frequencies of Soeumin and Soyangin groups were (c:0.70/t:0.30). and (c:0.71/t:0.29), that of Taeumin group was (c:0.57/t:0.43) and of Taeyangin group was (c:1.00/t:0.00) Conclusions : It was regarded the [c/t] polymorphism of CCK promotor region is available to classify the constitution. However, it is necessary to research about CCK gene polymorphism and more constitution population groups. It is also necessary to research the more functional gene's polymorphism and sasang constitution.

  • The samples used for this work were low grade c1ay($Fe_2O_3$ : 2.90%, $TiO_2$ : 1.65%) produced from Youngsan-po district, and for the iron-bearing impurities removal having a bad influence on Kaoline sample was carried out column floatatoin using Oleic acid, Potassium oleat, Aero 840 promotor, Aero 870 promotor and Keroseneas collectors. For removal of the iron-vearing impurities from the kaoline, investigated first chemical composition size distribution of Kaoline sample, also performanced a basic study on pulp density, collector and pH change on a sort of frother. The flotation separation of impurities from kaoline can be achieved by using both oleic acid and Aero 840 promotor as collectors within narrow acid limits(pH 2-3). Pine oil was more effective frother than MIBC with olelic acid as aa collector.

A plasmid vector faciliting gene expression in both yeast and mammalian cells

  • A plasmid vector with combined features of yeast shuttle vector and mammalian expression vector was constructed to facilitate expression of cloned gene in both cell-types. All necessary elements required for plasmid maintenance and selection in E. coli, yeast and mammalian cells were size-economically arranged in this plasmid. The numan cytomegalovirus (CMV) immediate early promoter and yeast GAL1 promoter were sequentially placed in front of the gene to be expressed. The synthetic splicing donor and acceptor sequences were inserted into the immediate upstream and downstream of the GAL1 promotor, allowing the CMV promotor to direct the expression of a given gene in mammalian cell environment by splicing out the interfering GAL1 promotor sequence. When the resulting vector containing LacZ as a gene was introduced into yeast and mammalian cells, both cells efficiently produced .betha.-galactosidase, dimonstrating its dual host usage.

  • The effect of PSOD expression on lacZ-sodP fusion (pYK4) was explored in Escherichia coli sodA sodB mutants (QC774) under oxidative stress. In this system, although .betha.-galactosidase activity was not fully induced by isopropyl-1-thio-.betha.-galactosidase (IPTG) and was inhibited by glucose, functional PSOD was under lacZ promotor control and was induced by IPTC, lactose, PQ and copper isons, finally, the results show that higher PSOD expression leel was consistently importnat in defending against superoxide radicals.

  • Purpose : The purpose of this study was to investigate the polymorphisms of the TNF-alpha promotor gene, its susceptibility to Kawasaki disease (KD) and to assess whether the TNF-alpha promotor gene polymorphism was related the risk of coronary artery lesions (CALs). Methods : From January 2003 to January 2007, 51 children (30 boys and 21 girls) with KD and 48 children forming an age-matched control group were studied. DNA from the peripheral blood of all the children was sampled, and the DNA polymorphisms of the 5' flanking regions of the TNF-alpha promoter gene at position -308 [guanine (G) to adenine (A)] were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Then, the relationship between KD and the TNF-alpha promotor gene polymorphisms was evaluated. Results : The A allele frequency of the -308 site of the TNF-alpha promotor gene was 17.6% (9/51) for children with KD and 6.8% (3/48) for the control group children, but this result was not statistically significant. Twenty-four patients experienced CALs within 60 days after the onset of symptoms. KD children with TNF-alpha -308 A allele had lower frequencies of CALs (12.5% versus 22.2%, P>0.05). Conclusion : The DNA polymorphism of the -308 site TNF-alpha gene was not associated with susceptibility to KD and a risk of CALs. Multicenter, large-scale randomized controlled trials are needed for further study.

Effect of Color Developing by Alkali and Heating of Cotton Fabrics Dyed with Persimmon Extract (알칼리와 열처리에 의한 면직물의 감즙염색 발색효과)

  • The purpose of this study is to improve the method of color developing with alkali solution as a promotor of color developing for feasible use. Cotton fabric was dyed with persimmon extract ranged with 0~3% alkali component with 5 types of strong to mild alkali solution. Heat treatment for color developing was applied to fabric dyed with persimmon extract and alkali mixing solution. Tests were carried out to analyze the change of surface color, ${\Delta},Ea^*b^*$, and water repellent of the dyed cotton fabric. The alkali mixing sample showed higher ${\Delta},Ea^*b^*$ value than control one without alkali mixing on the base of dyed fabric due to high color developing by alkali in the initial step of dyeing process. As alkali concentration increased, deeper dark color appeared on the fabric. The fabric color was changed to more dark in the application of sodium hydroxide, sodium carbonate, potassium carbonate in the initial step of dyeing process but color was not changed by increased heating time. However, the fabric showed a slight dark color with sodium acetate and more color change than that of the fabric dyed with persimmon extract without alkali. Therefore, sodium acetate seemed to a suitable promotor for color developing in persimmon extract dyeing. Property of water repellent was showed after color developing by heating with low concentration of alkali treatment.

THE HYPERMETHYLATION OF E-CADHERIN GENE IN ORAL SQUAMOUS CELL CARCINOMA (구강 편평세포암에서 E-cadherin 유전자의 과메틸화)

  • Loss of E-cadherin (E-cad) expression has been found in multiple cancers and is postulated to facilitate tumor cell dissociation and metastais. Promotor methylation may provides an alternative pathway for loss of gene function. This study evaluated the role of hypermethylation in the down-regulation of E-cad in oral squamous cell carcinoma (OSCC). We examined the E-cad expression by immunohistochemical staining and detected methylation status by methylation-specific polymerase chain reaction (MSP) in 20 OSCC tissues. Overally, 12 (60%) cases of hypermethylation of E-cad were detected and we found there were no correlation between methylation and age, histologic grade, lympn node metastasis, tumor size and clinical stage. However, Eleven (73.3%) of 15 samples which was negative for E-cad staining showed hypermethylation of E-cad promotor region. On the other hand, only one (20%) of 5 E-cad positive sample was observed with methylated status. The underexpression of E-cad was found to be related to promotor hypermethylation (p=0.035). In conclusion, we suggest that hypermethylation play a role in inactivation of E-cad gene and may be a appreciable biomarker for diagnosis and treatment of OSCC.
