Induction of FSTL1 Gene in Rat Myometrium by Exogenous Estrogen

  • The aim of this study was carried to the induction of follistatin-like 1 (FSTL1) in rat myometrium tissue by exogenous estrogen. PCR was performed by using estrogen treatment after ovariectomy of myometrium mRNA and only ovariectomy of myometrium mRNA and on ovariectomy of myometrium mRNA in rat normal uterus tissue. The PCR products were visualized on agarose gel (1.2%) electrophoresis. FSTL1 mRNA expression level was significantly higher (**P<0.001 or *P<0.05) in estrogen treatment after ovariectomy than in only ovariectomy, and was significantly higher (**P<0.001) in no ovariectomy than in estrogen with treatment after ovariectomy. In conclusion, Although the mechanisms of FSTL1 gene were uncertain, It also might be inducted by exogenous estrogen. And also it is thought to be related to the estorgen mechanism.

Effect of Crude Saponin Fraction from Korean Red Ginseng on Physiological Events of Ovariectomized Rat (난소절제 흰쥐의 생리적 장애에 미치는 고려홍삼 조사포닌 분획의 영향)

  • This study was carried out to investigate the effect of crude saponin fracton from Korean red ginseng on physiological events of ovariectomized rats. The saponin fractions was prepared by Diaion HP-20 adsorption chromatography and spirit. Total 50 rats were divided into 5 groups : normal control (NC), sham-ovariectomized (SO), ovariectomized (OR), ovariectomized and saponin treated (OS), and normal control treated with saponin (NS). Saponin was intraperitonally administered for 8 weeks. Ovariectomy was carried out at 1 st and 2nd weeks of experiment. Right-side ovary of rat was removed at 1st week, the otehr was done at 2nd week. The body weights fo ovariectomized rats showed no significant changes but that of NS group showed significant increase (p<0.05) compared with NC group. Ovariectomy of rats reduced the weights of femur by 6 to 8% compared with that of NC group. In addition, femur weight of NS group was 5 to 6% higher than that of NC. Uterus weight showed no significant differences by saponin treatment or ovariectomy. Serum testosterone level of ovariectomized rats increased by 60 times compared with that of NC. However, administration of crude saponin to ovariectomized rat attenuated testosterone level to almost that of NC. These results suggest that Korean red ginseng saponin attenuates physiological disorders induced by malfunction of ovary.

  • Osteoporosis is a bone disease associated with reduced bone mineral density resulting in debilitating bone fractures. The present study was carried out to determine the influence of ovariectomy(OVX) on serum level of sex steroid and bone metabolism, as well as bone mechanical property, in OVX subjects in comparison with controls. Body weight and food intake was significantly increased in OVX subjects than in controls. The serum level of estradiol(E2) was significantly lower in OVX subjects than in controls. The serum level of calcium and alkalin phosphatae were significantly increased in OVX subjects than in controls. The bone strength, based on the three point bending test, was not significantly different. Seven weeks after ovariectomy, the cavities in the bone reached 223% of the normal. In conclusion, seven weeks after ovariectomy osteoporo sis was evidently appeared.

Study of acupuncture stimulation on experimental osteopenia

  • In order to study the effect of acupuncture stimulation on bone mineral density (BMD), using the ovariectomized (OVX) rat model, we assessed the degree of osteopenia by dual-energy X-ray absorptiometry, measured the level of locomotor activity using a metabolism measuring system, and performed histological studies of bone tissue. Twenty-four female Wistar rats (8 weeks old, 160 - 180 g)were divided into three groups. Rats in the OVX-A group underwent ovariectomy followed by acupuncture stimulation. The OVX rats in the Vehicle control group were not treated with acupuncture as a control. The rats in the control group received neither ovariectomy nor acupuncture. Acupuncture stimulation for 12 weeks in the OVX-A group inhibited the reduction in BMD of the femoral bones caused by ovariectomy. Moreover, in the two OVX groups, there was no clear difference in the level of locomotor activity between the active and resting phases prior to acupuncture stimulation in each rat, and the pattern of locomotor activity was irregular. After acupuncture stimulation of the OVX-A rats, the pattern of locomotor activity became diphasic with clear active and resting phases, as was observed in the Control group. On histological studies, the continuity of trabecular bone was maintained more favorably and bone mass was higher in the OVX-A group than in the vehicle control group. These results suggest that the increased locomotor activity that had been induced by acupuncture stimulation increased the BMD.

A histochemical study of argentaffin endocrine cells in the gastrointestinal tract of ovariectomized rats

  • The regional distributions and frequencies of argentaffin endocrine cells in gastrointestinal (GI) tract of osteoporotic Sprague-Dawley rat induced by ovariectomy were studied by Masson-Hamperl silver stain. The experimental animals were divided into two groups, one is non-ovariectomized group (Sham) and the other is ovariectomized group (OVX). Samples were collected from each part of GI tract (fundus, pylorus, duodenum, jejunum, ileum, cecum, colon and rectum) at 10th week after ovariectomy or sham operation. Argentaffin cells were detected throughout the entire GI tract with various frequencies regardless of ovariectomy except for the rectum of OVX in which no cells were detected. Most of these argentaffin cells in the mucosa of GI tract were generally spherical or spindle in shape (open type cell) while cells showing round in shape (close type cell) were rarely found in gland regions. Significant decrease of argentaffin cells was detected in OVX compared to that of Sham except for the fundus and jejunum. However, in the fundus and jejunum, argentaffin cells in OVX showed similar frequency compared to that of Sham. In conclusion, the endocrine cells are the anatomical units responsible for the production of gut hormones that regulate gut motility and digestion including absorption, and a change in their density would reflect the change in the capacity of producing these hormones and regulating gut motility and digestion. Ovariectomy induced severe quantitative changes of GI argentaffin endocrine cell density, and the abnormality in density of GI endocrine cells may contribute to the development of gastrointestinal symptoms in osteoporosis such as impairments of calcium and some lipids, frequently encountered in patients with postmenopausal osteoporosis.

THE HISTOLOGIC STUDY OF THE RAT TEMPOROMANDIBULAR JOINT CHANGE FOLLOWING OVARIECTOMY (자성백서의 난소절제시 악관절 변화에 관한 조직학적 연구)

  • This investigation studied the effect of ovariectomy on the temporomandibular joint (TMJ) of rat. Forty female Sprauge-Dawley strain white rats, each weighing about 200grams were used and divided into two groups. The one group was performed ovariectomy, and the other was performed sham operation. Both group were killed at the intervals of 3 days, 1, 2, 4, and 8 weeks postoperatively, paraffin section were stained with H&E, MT and histomorphometric study of the TMJ was performed in a synchronous manner with an sham operated control group The result were as follows: 1. At 4 weeks after ovariectomy, an adhesion between the inferior surface of the articular disc and the condyle was observed in the central portion. At 8 weeks after ovariectomy, a focal thickening of the bone (osteophyte) was observed in anterior portion and there was a marked decrease in the trabecular bone in the posterior region. 2. The thickness of cartilage zone was increased at 2 weeks postopratively in the central portion, and decreased at 4 weeks postopratively in the posterior portion. The thickness of transitional zone was decreased at 1 week postoperatively in the anterior and proliferating zone was decreased postoperatively. The thickness of articular zone was decreased at 3 days postopratively in the anterior portion, and decreased at 1 week postoperatively in the central portion. From the above finding, we could conclue that estrogen deficiency in rats causes alteration of TMJ in marked early stage.

Bone Mineral Density Changes after Ovariectomy in Rats as an Osteopenic Model : Stepwise Description of Double Dorso-Lateral Approach

  • Objective : This study describes a method for inducing osteopenia using bilateral ovariectomy (OVX), which causes significant changes in bone mineral density (BMD) in rats. Methods : Twenty-five 10-week-old female Sprague Dawley rats were used. Five rats were euthanized after two weeks, and BMD was measured in their femora. The other 20 rats were assigned to one of two groups : a sham group (n = 10), which underwent a sham operation, and an OVX group (n = 10), which underwent bilateral OVX at 12 weeks of age. After six weeks, five rats from each group were euthanized, and BMD was measured in their femora. The same procedures were performed in the remaining rats form each group eight weeks later. Results : The femur BMD was significantly lower in the six-week OVX group than in the six-week sham group, and in the eight-week OVX group than in the eight-week sham group. Conclusion : Bilateral OVX is a safe method for creating an osteopenic rat model. The significant decrease in BMD appears six weeks after bilateral OVX.

Effects of Aqueous Extracts of Cynanchum wilfordii in Rat Models for Postmenopausal Hot Flush

  • Menopausal hot flushes (HFs), which manifest as a transient increase in skin temperature, occur most frequently in postmenopausal women, and sometimes negatively influence daily life. We investigated the effect of an aqueous extract of Cynanchum wilfordii (CWW) in a rat model of menopausal HFs, where tail skin temperature (TST) is increased after the rapid estrogen decline induced by ovariectomy. Ten-week-old female rats were ovariectomized and treated with CWW for 1 week. We measured TST and rectal temperatures (RT) and investigated serum estradiol. The TST in ovariectomized (OVX) rats was significantly elevated after ovariectomy compared with control rats, whereas the RT in OVX rats was not elevated. Administration of CWW (200 mg/kg/d for 7 days, p.o.) significantly improved the skin temperature increase in OVX rats. The lower level of serum estradiol in OVX rats was significantly increased by supplying E2, but it was not affected by CWW. The present study indicates a need for future research involving treatment with high concentrations of C. wilfordii and measurement over 24 h.

Study of moxibustion stimulation in the ovariectomized rat model

  • In order to examine the effectiveness of moxibustion stimulation for climacteric disturbance, we administered moxibustion stimulation to ovariectomized (OVX) rats, and compared the tail surface temperature, laboratory parameters, and the level of locomotor activity with those of untreated rats. Twenty-four female Wistar rats (8 weeks old, 160 - 180 g) were divided into three groups. The rats in the OVX-M group underwent moxibustion stimulation after ovariectomy. The rats in the OVX-C group underwent ovariectomy but did not receive moxibustion stimulation. The rats in the Normal group received neither ovariectomy nor moxibustion stimulation. The level of locomotor activity was determined by a metabolism measuring system. The tail surface temperature was significantly lower in the OVX rats before moxibustion stimulation than in the Normal group. In the OVX rats before moxibustion stimulation, there was no clear difference in the level of locomotor activity between the active and resting phases, and the pattern of locomotor activity was irregular. After moxibustion stimulation, the tail surface temperature of the OVX-M group did not significantly differ from that of the Normal group, and the pattern of locomotor activity of the OVX-M group became diphasic with clear active and resting phases, similar to that observed in the Normal group. These results demonstrate that moxibustion stimulation is effective for the treatment of climacteric disturbance.
