  • Winter crops have been recognized as an alternative to soil management for fertility and crop productivity in paddy soil. Recently, rape has been produced at winter season and there is little research results on reduction of N fertilizer by adding rape residues for rice cultivation. In this study, we investigated the productivity and quality of rice by applying with 0, 27, 63, $90kg\;ha^{-1}$ of N fertilizer after input of rape residues into soil for two years. The highest yield of rice was average $4.68Mg\;ha^{-1}$ at the treatment applied with the $90kg\;N\;ha^{-1}$ with rape residue. It was reduced to N rate from 31.6 to $43.2kg\;N\;ha^{-1}$ in comparison to $4.53Mg\;ha^{-1}$ of maximum yield in treatment added $90kg\;N\;ha^{-1}$ with rape. Amylose content in rice was similar among treatments with/without rape residue, but protein content was lower in treatment with rape residue than in those without rape residue. In conclusion, input of rape residue in rice cultivation could be alternative to reduction of N fertilization and improvement of quality by adjusting rice productivity in paddy soil under cropping system.

  • Nuclear Factor 1 (NF1) proteins are a family of transcriptional factors consisting of four different types: NF1-A, -B, -C, and -X. Some NF1 transcription factors contain a heptasequence motif, SPTSPSY, which is found as a repeat sequence in the carboxy terminal domain (CTD) of the largest subunit of RNA polymerase II. A similar heptasequence, SPTSPTY, is contained in rat liver NF1-A at a position between residues 469 and 475. In order to investigate the roles of the individual amino acids of the heptasequence of rat liver NF1-A in transcriptional activation, we systematically substituted single and multiple amino acid residues with alanine residue(s) and evaluated the transcriptional activities of the mutated NF1-A. Substitution of a single amino acid reduced transcriptional activity by 10 to 30%, except for the proline residue at position 473, whose substitution with alanine did not affect transcriptional activity. However, changes of all four serine and threonine residues to alanine or of the tyrosine residue along with the serine residue at position 469 to alanine reduced the activity to almost background levels. Our results indicate that multiple serine and threonine residues, rather than a single residue, may be involved in the modulation of the transcriptional activities of the factor. Involvement of the tyrosine residue is also implicated.

  • Peptide fragments, isolated from proteolytic cleavage of thyroglobulin at specific sites, were examined for the iodination of tyrosine residues. The 50 kDa polypeptide, which was prepared from digestion of bovine thyroglobulin and continuous preparative SDS-PAGE, was subjected to reduction with DTT and alkylation with iodoacetic acid to generate S-car-boxymethylated peptide derivative, which was further hydrohysed by endoproteinase-Asp-N. Peptide products were separated by RP-HPLC, and each fraction was analyzed by LC/ESI-MS and MALDI-MS analyses. Based on the specificity of endoproteinase-Asp-N andthe mass spectra data, a peptide fragment turned out to correspond to a peptide, DALCCVKCPEGSYFQ (1438-1452), characterized by the presence of a thioredoxin box (CVKC) and a tyrosine residue. In addition, another peptide fragment (1453-1465) containing a thioredoxin box (CIPC) and a tyrosine residue was also observed. However, any evidence of iodination of the tyrosine residue present in these peptides was not provided. Meanwhile, tyrosine residues in the peptides, DVEEALAGKYLAGRFA (1366-1381) and DYSGLLLAFQVFLL (1290-1303) were found to be iodinated; mono- or diiodinated tyrosine residues, characteristic of a hormogenic site, existed in both peptides. In addition, the tyrosine residue in the peptide (1218-1252), corresponding to a hormonogenic site was also iodinated. Thus, there was a sharp difference of the susceptibility to oxidative iodination between the tyrosine residue in a hormonogenic site and that in a thioredoxin region. From these results, it is suggested that polypeptide region adjacent to tyrosine residues may govern the susceptibility of tyrosine to oxidative iodination.

  • Food safety or sanitation are terms broadly applicable to procedures designed to ensure that food quality is high and free of factors which may adversely affect human health. These factors include zoonotic diseases and acute and chronic effects of ingesting natural and human-made xenobiotics. Use of drugs in animal production for the treatment and control of animal diseases, to promote growth rate, and to improve feed conversion efficiency has expanded year by year, thus increasing the possibilities for occurrences in animal products of residues harmful to humans. Governmental agencies have made efforts to control or prevent residue problems. The Korean Food and Drug Administration (KFDA) is charged with the responsibility of establishing tolerances for veterinary drugs, pesticides, and mycotoxins and other non-pharmaceutical substances. The Department of Veterinary Service is responsible for establishing guidelines regarding withdrawal times of drugs, approval of drugs, their uses, and sanitation enforcement of livestock products. The authors describe the toxicological basis for the establishment of tolerance levels for xenobiotics and the pharmacokinetic basis for establishing withdrawal time for veterinary drugs. The regulatory tolerance levels of chemicals in pork and swine feed, Korean regulations on the use of feed additives, rapid residue test methods, the National Residue Program, and the Food Animal Residue Avoidance Databank are discussed. Rapid EIA methods that are under development for the screening of live animals are described These methods predict tissue residues from an examination of blood samples taken from pigs before they are slaughtered.

  • This study conducted to investigate the effects of dietary Korean medical herb residue on egg quality and serum cholesterol of laying hens under heat stress. One hundred forty four, 41 weeks old ISA brown commercial layer, were used in a 28 d growth assay with a 7d adjustment period. Dietary treatments Included 1) Control (basal diet), 2) KMHR1.0 (basal diet+1.0% korean medical herb residue), 2) KMHR2.0 (basal diet+2.0% Korean medical herb residue). For overall Period, laying hens fed KMHR1.0 diet were higher hen-day egg production than laying hens fed Control and KMHR2.0 diets with significant difference (quadratic effect, p<0.01). Egg weight, egg shell breaking strength and egg shell thickness were not influenced by Korean medical herb residue supplementation (P>0.05). As adding level of Korean medical herb residue increased in the diets, yolk color tended to increase, however, the differences failed to reach significance (P>0.05). Also, egg yolk index was not influenced by korean medical herb residue supplementation (P>0.05). Total-, HDL- LDL+VLDL-cholesterol and triglyceride concentrations in serum tended to increase as the concentration of Korean medical herb residue in the diets was increased without significant difference (P>0.05). In conclusion, supplementing 1.0% Korean medical herb residue to a corn SBM meal diet for laying hens increased hen-day egg production.

  • BACKGROUND: This research has investigated the residue patterns of insecticide flubendiamide on three species of peaches with different surface forms, and the residue amounts of them when mixed with a spreader. METHODS AND RESULTS: Pesticide used for field application on peaches was 20% flubendiamide of suspension concentrate(SC) and was sprayed at a recommended rate. The residue amounts of flubendiamide in peach were analyzed by HPLC equipped with UV detector. After the observation with a microscope, the rank of fuzz amount on peach's surface was Kurakatawase, Wolmi in descending order and Cheonhong did not have any fuzz. The residue amounts of flubendiamide were 0.54 mg/kg for Kurakatawase, 0.43 mg/kg for Wolmi and 0.10 mg/kg for Cheonhong, respectively. When flubendiamide was used with a spreader, polyoxy ethylene methylpoly siloxane, the residue amount for Kurakatawase barely changed at 0.55 mg/kg regardless of mixing with a spreader, and at 0.53 mg/kg for Wolmi. In Cheonhong, the residue amount was 0.48 mg/kg, which increased by 4.8 times due to the use of a spreader. CONCLUSION: This result indicates that the residue amounts of flubendiamde were affected by the surface forms of peaches, and in the presence of a spreader the residue amount did not increase in fuzzy species, but was affected greatly for species without fuzz.

  • BACKGROUND: The present study was carried out to identify the residue patterns of insecticide pyridalyl and fungicide fluopicolide in watermelon and calculate the biological half-lives for establishing the pre-harvest residue limits (PHRLs). METHODSANDRESULTS:The watermelon samples for residue analysis were harvested 7 times during 0~10 days (Field 1) and 0~20 days (Field 2) after treatment of pesticides on watermelon in two different fields at the recommended dose, respectively. The residue analysis was conducted with HPLC/UVD. The method limit of quantitation (MLOQ) were set at 0.05 and 0.02 mg/kg, respectively, and overall mean recoveries were 81.2~90.5% for pyridalyl and fluopicolide. The residues in sample were stable for 43~47 days. The initial residue amount in field 1 and 2 were 0.12~0.16 mg/kg for pyridalyl and 0.23~0.24 mg/kg for fluopicolide, which were below maximum residue limit (MRL). The biological half-lives in field 1 and 2 were 26.9 and 17.9 days for pyridalyl and 16.6 and 94.2 days for fluopicolide, respectively. CONCLUSION: The PHRL for watermelon were estimated as 0.21 and 1.03 mg/kg for pyridalyl and flopicolide at 10 days before harvesting. The residue patterns of pyridalyl and fluopicolide were characterized by a very slow decrease of residue levels in watermelon.
