The Fruiting Body Formation of Oudemansiella radicata in the Sawdust of Oak (Quercus variabilis) Mixed with Rice Bran

  • To screen additives and their mixed ratio suitable for the mycelial growth and fruiting body formation of Oudemansiella radicata in the oak sawdust, additives such as rice bran, fermented soybean powder and wheat bran were used. Generally, the mycelial growth of O. radicata has been stable on oak sawdust mixed with rice bran of $5{\sim}20%$. In case that O. radicata was cultured for about 30 days at $22{\pm}1^{\circ}C$ under the illumination (350 lux) of 12 hours and moisture condition of $90{\pm}5%$, the primordia have been formed gradually from red-brown crusts covering the surface of oak sawdust media. Based on the experimental results from 9 strains of O. radicata, fruiting bodies were produced widely on oak sawdust medium mixed with rice bran of 5 to 30%. Even though fruiting bodies of O. radicata have been produced well on oak sawdust media mixed with rice bran, fruiting bodies of O. radicata were produced intensively on oak sawdust media mixed with rice bran of 10%. Therefore, this result will provide a basic information for commercial production of fruiting body of wild O. radicata. This result is the first report associated with an artificial fruiting body formation of O. radicata in Korea.

Optimum mixing rate of used media for saving the production cost of Flammulina velutipes (팽이버섯(Flammulina velutipes) 생산비 절감을 위한 폐배지 적정 혼합비율)

  • These experiments were conducted to find the optimum mixing rate of used media for saving the production cost of Flammulina velutipes. The materials for media formation was used needle-leaf tree sawdust, media that finished 1th cultivation of F. velutipes, corncob meal, and rice bran, wheat bran as nutrition source. We inoculated the F. velutipes in 14 kinds of different media types and checked the spawn growth speed, fruit body quality and quantity. Two nutrition agents, which is rice bran and wheat bran, did not affected the incubation period, but the effective stem number, quality and quantity of fruit body was better at rice bran than wheat bran. The quality of fruit body produced at mixed 20% of used media (needle-leaf tree sawdust 60% + used media 20% + rice bran 20%) was similar to control plot (needle-leaf tree sawdust 80% + rice bran 20%), but the yield was improved 10% than control plot 130g. According as the used-media mixing amount increases, quality and quantity of fruit body became low remarkably. Therefore, the optimum mixing amount of used-media was 20% and it increased 10% of fruitbody yield.

Combined application of oil cake and rice bran reduced the number of weeds and increased the yield of paddy rice in a paddy field incorporated with white clover

  • The combined application of oil cake and rice bran into the soil surface was found useful for weed control in our previous pot study. The present study was undertaken to evaluate the performance of white clover (Trifolium repens L.) while incorporated in the paddy field and effects of combined fertilizer on weed control and rice yield. A plot was divided into two parts i.e. white clover incorporated and not incorporated. The nitrogen content of the incorporated white clover was $12.5gm^{-2}$. Chemical fertilizer and combined fertilizer plots were compared with non-fertilizer conditions. The mixed ratio of combined fertilizer was oil cake 1.35 and rice bran 1.0. Combined fertilizer was applied to the soil surface, and chemical fertilizer was mixed in the soil. Nitrogen application rate was $8gm^{-2}$ for any fertilizer. The weed numbers were significantly reduced in the white clover plot irrespective of application condition both at heading and harvest time. Also, weed control ability was improved by the use of combined fertilizer. In the not incorporated plot, the number of weeds was suppressed about 90% by applying combined fertilizer. The rice yield was markedly increased by the incorporation with white clover under all fertilization conditions. Contribution rates of increased rice yield by white clover and combined fertilizer were about 55% and about 25%, respectively. The rice yield was increased by the incorporation with white clover, and the number of weeds remarkably decreased as well. Also, these effects were improved due to combined application of oil cake and rice bran.

Xylanase Production by Bacillus sp. A-6 Isolated from Rice Bran

  • A Bacillus sp. A-6 strain that produced xylanase was isolated from rice bran. The optimal temperature and pH for xylanase activity of the culture supernatant of Bacillus sp. A-6 were 40$^{\circ}C$ and pH 7, respectively. The optimal temperature and pH for xylanase production in the xylan medium were 30$^{\circ}C$ and pH 9, respectively. The optimal concentrations of oat spelt xylan and peptone for xylanase production were 0.5% and 1.5%, respectively. The best nitrogen sources for xylanase production was beef extract, but xylanase production was also supported comparably by tryptone and peptone. The bacterial growth in the optimal xylan medium reached stationary growth phase after 12 h of incubation. The xylanase production in the culture supernatant increased dramatically during the initial 12 h exponential growth phase and then remained constant at 23.8-24.5 unit/ml during the stationary growth phase. The pH of the culture medium decreased from 8.8 to 6.7 during the exponential growth phase and subsequently increased to 8.1 during the stationary growth phase. Rice bran, sorghum bran, and wheat bran as well as oat spelt xylan induced xylanase production. The xylanase production was repressed when glucose was added to the xylan-containing medium.

Characteristics of Seedling Establishment and Yield of Platycodon grandiflorus by Ridge Width and Mulching Materials (이랑폭과 피복재에 따른 도라지의 입모율 및 수량특성)

  • This study was carried out to investigate the seedling establishment and yield of direct-sown Platycodon grandiflorus seeds cultured by three ridge width, 0.4, 0.8, and 1.2 m and four mulching materials, rice straw, rice bran, black plastic film, and no mulching in upland. Seedling establishment rate (70%) was the highest in rice straw mulched plot of 500 seeds/$m^2$ sowed and root yield after one year was also the highest in rice straw mulched treatment and followed by rice bran, no mulching, and black plastic film treatment. In this result, ridge width 120cm and rice straw mulching combined treatment was best for getting the highest seedling establishment ratio and seedling numbers per area. However, ridge width and mulching materials should be considered soil moisture content and weed population for saving labor cost.

Enhanced Production of ${\gamma}$-Aminobutyric Acid Using Rice Bran Extracts by Lactobacillus sakei B2-16

  • An efficient and simple fermentation process was developed for the production of ${\gamma}$-aminobutyric acid (GABA) by Lactobacillus sakei B2-16. When the L. sakei B2-16 was cultivated in the rice bran extracts medium containing 4% sucrose, 1% yeast extract, and 12% monosodium glutamate, the maximum GABA concentration reached 660.0 mM with 100% conversion yield, showing the 2.4- fold higher GABA concentration compared with the modified MRS medium without the rice bran extracts. The GABA production was scaled-up from a laboratory scale (5 l) to a pilot (300 l) and a plant (5,000 l) scale to investigate the application possibility of GABA production to industrial fields. The production yields at the pilot and plant scales were similar to the laboratory scale using rice bran extracts medium, which could be effective for the low-cost production of GABA.

Xanthan Gum Production from Hydrolyzed Rice Bran as a Carbon Source by Xanthomonas spp.

  • The aim of this study was to utilize rice bran, the main waste product of paddy processing, in xanthan gum production by Xanthomonas campestris fermentation. Deffated rice bran was enzymatically hydrolyzed using cellulase, gluco-amylase, alpha-amylase and xylanase at various pHs and temperatures within 0-12 h. The highest sugar content reached at $35^{\circ}C$, pH 5.5 in 6 h with 41.66%. The enzymatic hydrolysate was used as the carbon source for xanthan gum production by X. campestris NRRL B-1459 and X. campestris pv. campestris. The highest productivities obtained were 21.87 and 17.10 g/L, respectively. Viscosity measurement for the obtained xanthan gums and commercial gum was carried out in gum solutions at various pHs and temperatures. The highest viscosity was reached with 1% gum solutions at $20^{\circ}C$ and pH 5.5 for all gums with viscosity values of 470, 131 and 138 mPa sec, respectively. This work has provided relevant scientific information about the use of rice bran, an abundant agroindustrial residue, to produce xanthan gum.

Isolation of Major Microflora Bacillus coagulans from Rice Bran (쌀겨발효로부터 분리된 우점종 미생물 Bacillus coagulans의 분리)

  • It is known that temperature during solid fermentation using rice bran is increased upto 60-70 degree. To investigate the major microflora regarding temperature maintenance of rice bran bathing, we first isolated predominant microbes using various media by a limiting dilution method. The RNA of isolated strains were purified and sequenced. The rRNA sequencing revealed that the selected strains were similar to Bacillus coagulans according to their taxonomical relationships. Together, these results indicate that Bacillus coagulans is a major kind of microflora during solid fermentation using rice bran.

Performance and Emissions Characteristics of a Diesel Engine with Some Bio-Oil Fuels

  • The performance and exhaust emissions of a diesel engine using light oil, heated, rice-bran oil , heated rice-bran oil treated with ultrasonic wave, used frying oil, use frying oil treated with ultrasonic wave, used frying oil, used frying oil treated with ultrasonic wave, methyl esters of rice-bran oil and used frying oil have been compared. All the fuels performed satisfactorily in a precombustion chamber-type diesel engine without injection pump recalibration or any engine modification at the range of engine speed from 1600 to 2800 rpm at its full load during a sort period , with the rice-bran oil and rice-bran oil treated with ultrasonic wave requiring somewhat preheating when ambient temperature was below 15$^{\circ}C$. General performance and emission characteristics of light oil and bio-oils were comparable , with the bio-oil based fuels giving very low SO$_2$ and lower smoke readings.

Effect of Different Pretreatment Methods on the Bioconversion of Rice Bran into Ethanol

  • The efficiency of acid, enzyme and microbial pretreatment of rice bran was compared based on the content of cellulose, hemicellulose, reducing sugars and xylose in the substrate. An isolate of Aspergillus niger or a strain of Trichoderma viride(MTCC 800) was employed for microbial pretreatment of rice bran in solid state. Acid pretreatment resulted in the highest amount of reducing sugars followed by enzyme and microbial pretreatment. A. niger showed a higher rate of hydrolysis than T. viride. The rice bran hydrolysate obtained from the different methods was subsequently fermented to ethanol either by Zymomonas mobilis(NCIM 806) or by Pichia stipitis(NCIM 3497). P. stipitis fermentation resulted in higher ethanol(37% higher) and biomass production($76{\sim}83%$ higher) than those of Z. mobilis. Maximum ethanol production resulted at 12h in Zymomonas fermentation, while in Pichia fermentation, it was observed at 60h. Microbial pretreatment of rice bran by A. niger followed by fermentation employing P. stipitis was more efficient but slower than the other microbial pretreatment and fermentation.
