  • A full-length cDNA encoding ribulose-1,5-bisphosphate carboxylase small subunit (rbcS) has been isolated and its nucleotide sequence determined from root in ginseng plant (Panax ginseng). The rbcS cDNA of ginseng is 790 nucleotides long and has an open reading frame of 549 bp with deduced amino acid of 183 residues (pI 8.37), 20.5 kDa. The deduced amino acid sequence of rbcS matched to the previously reported rbcS protein genes and showed a high similarity with the $78\%$ identity with rbcS of Helianthus annuus (CAA68490). In the phylogenetic analysis based on the amino acid residues, the ginseng rbcS was clustered with H. annuus (CAA68490), C. morifolium (AA025119) and L. sativa (Q40250).

  • We isolated a cDNA clone that encodes a ribulose-1,5-bisphosphate carboxylase small subunit (rbcS) from spinach using a soybean rbcS cDNA as a probe. The small subunit consists of 180 amino acids including a transit peptide of 57 residues. Comparison of the amino acid sequence with those of other plant species shows a maximum of 70-80% identical residues. Southern blot analysis suggests the existence of multiple rbcS genes in the spinach genome. Northern blot analysis indicates that the rbcS gene is expressed predominantly in leaves and that the expression of the gene is induced by light.

  • The software developed for the large scale Programmable Controller consists of the programmer's S/W, the Controller's S/W the RBC's (Remote Base Controller's) S/W and the Analog's S/W. The programmer's S/W, running on the Programmer, includes the editor, the compiler, the communication program, and some other programs for easy use. The Controller S/W, which requires the fast scanning time, consists of the BTI( Block Type Instruction) solving program, the timer service routine, the i/o update program, the communication program and etc. The RBC's S/W includes the communication program, the error recovery program and the i/o processing program. The analog S/W, controlled by the Programmer, includes the PID program. The data communication between the Programmer and the Controller the Controller and the RBC, and the RBC and the Analog are developed.

Isolation of a Rice Genomic Clone Encoding Ribulose-1,5-bisphosphate Carboxylase (리블로스 1,5- 이인산 탄산화효소 유전자의 분리 및 특성규명)

  • To study the light-induced expression mechanism and protein transport into the chloroplast, a rice genomic clone (GrbcS) for the small subunit of ribulose 1,5-bisphosphate carboxylase (rbcS) was isolated and its nucleotide sequence was determined. Nucleotide sequence analysis of GrbcS revealed that the gene consists of two exons interrupted by an intron, encoding a protein of 175 amino acids including a transit peptide of 47 amino acids. These structural features of GrbcS are consistent with those of other rbcS genes from monocot species. Genomic Southern blot analysis suggested that the rbcS genes are present as a relatively small multigene family in the rice genome. Comparison of the nucleotide and deduced amino acid sequences to other rice rbcSs shows close sequence similaritiy. Conserved DNA sequences present in other light-responsive genes are also found in the 5’ upstream region of GrbcS such as G-box, 3AF1-binding site and GATA site. The possible function of these putative regulatory elements are discussed.

Confirmation on Taxonomic Status of Spatoglossum pacificum Yendo (Dictyotaceae, Phaeophyceae) Based on Morphology and Plastid Protein Coding rbcL, rbcS, psaA, and psbA Gene Sequences

  • Phonological, morphological and molecular characteristics of Spatoglossum pacificum Yendo are examined. S. pacificum has an annual life cycle composed of saprophytes with apparent absence of male and female gametophytes in Korea. The seasonal growth of this species explains that the annual growth is closely related to the monthly variation of water temperature. S. pacificum has protruding reproductive structures above the outmost cortical layer. Although this observation is restricted to several species, reproductive structures on the thallus can make S. pacificurn distinguishable from S. crassum and S. lacturn. The morphogenesis of a midrib at the base of S. pacificum in this study is the same as those of Dictyopteris but different from those of S. crassum and S. lacturn, suggesting that S. pacificum is closely related to Dictyopteris. In the comparison of plastid gene sequences among species of Spatoglossum and Dictyopteris, S. pacificum is more similar to D. divaricata and D. undulate than those of S. crassum in rbcL, rbcS, psbA and psaA. This result is congruent with the anatomical characteristic of a midrib at the base of the thallus and the protrusion of reproductive organs on the thallus. The phylogenetic relationship based on these plastid genes also shows that S. pacifism is included in Dictyotpteris Glade and separated from S. crassum. We propose the new combination of Dictyopteris pacifica (Yendo) I.K. Hwang, H.S. Kim et W.J. Lee, comb. nov. based on the differences of anatomical characteristics of the midrib, the existence of reproductive organs on thallus and the molecular analyses.

  • Polysomal polyadenylated mRNAs which were purified from pea leaves were fractionated by sucrose grandient sedimentation. Fractions corresponding to the peak at 11.5S were found to contain mostly mRNA encoding the precursor polypeptide to the small subunit of ribulose bisphosphate carboxylase (rbcS) by in vitro translation in wheat germ extract. Double-stranded cDNA which was synthesized from the 11.5S mRNA was cloned into Hind III site of plasmid pBR 325. A cDNA clone, H24, was identified to code for rbcS. In vitro translation product of the hybridization-selected mRNA was molecular weight 20,000, presumably the precursor of rbcS. The nucleotide sequences of the H24 showed almost complete homology with the sequences encoding the transit peptide of the rbcS-3A gene which was reported by Fluhr et al.(1986).

  • Recently it is reported that RBC membrane fluidity decreases and RBC calcium levels increase with age. The aim of this study was to analyze changes in lipid and calcium metabolism with age, and to seek relationship of diet and metabolism. With clinically normal Korean adults(male 60, female 63), this study was carried out in three phases : 1) to analyze fatty acid percentage of RBC membrane, 2) to analyze calcium levels of RBC with age, and 3) to compare the effects of dietary fatty acid intake on blood fatty acid profiles. The results are as follows : The P/S ratio of RBC membrane fatty acid decreased with age. The RBC calcium content increased according to age, with women having a higher level than men. The higher intake groups of linolenic acid(C18:3) has statistically higher serum linolenic acid levels. But dietary effects of membrane fatty acid were not found. Therefore, the further research to seek the possible relationship of diet and membrane fatty acid should be continued.

Effect of Humenolepis nana infection on immunological responses of mice to sRBC (마우스에 있어서 왜소조충 감염이 sRBC에 대한 면역능에 미치는 영향)

  • In an attempt to investigate the effect of Hymenolepis dana infection on immunological responses to sRBC in ICR strain of mice, cellular and humoral immune responses were chronologically monitored after sensitization with sRBC. Mice weighing about 20 g were allocated into artificial and natural infection groups. The shell-free eggs of H. dana were inoculated into mice on the day 0 (initial) and day 10 in the former group, and praziquantel (25 mg/kg/day) was administered for 3 days to the one half of the mice at the 15th day after the first inoculation and to all of the mice in natural infection group. In artificial infection group, the delayed-type hypersensitivity (DTH) to sRBC was considerably decreased on the day 10 after the first inoculation, and then elevated gradually to normal. Eosinophils in the peripheral blood increased slightly. The hemagglutinin (HA) and hemolysin (HE) titers during the early stage were shown to be more or less higher than those of control. Thereafter, the titers were returned to normal, followed by a transient decrease on the day 15 post-infection. The sRBC rosette and antibody-treated rosette-forming capacities on the day 15 post.infection were temporarily lowered but became higher thereafter. The mucosal mast cells (MMC) in the small intestine were gradually increased to make a peak on the day 10 post-infection and then maintained more or less at lower level. After praziquantel treatment, the DTH and the number of eosinophils were decreased slightly and the MMC number and sRBC rosette-forming capacity were considerably decreased. The titers of HA and HE and antibody-treated rosette-forming capacity, however, were elevated in general. In natural infection group, the DTH, the number of eosinophils, and MMC which were elevated due to H. dana infection were gradually returned to normal after prasiquantel treatment. The titers of HA and HE which were decreased by parasite infection were increased to normal after the treatment. However, the capacities of sRBC rosette or antibody-treated rosette formation were maintained at low levels in spite of the treatment. These results revealed that the immune responses to sRBC were significantly activated during H. dana infection, although they were transiently decreased during the days 10~15 post-infection.

Phylogenetic Relationships of Soranthera ulvoidea (Chordariaceae, Phaeophyceae) on the Basis of Morphology and Molecular Data

  • The brown algal family Chordariaceae sensu lato is a focus of taxonomy because recent studies suggest a broad concept of the family, including genera formerly classified in the Dictyosiphonales. Using morphology, plastid rbcL and nrDNA ITS sequences, we evaluated relationships of the monotyic genus Soranthera (S. ulvoidea), which has been classified in the Punctariaceae. The species occurs in Bering Sea and Aleutian Islands, Alaska to Baja California. Thalli are globose to lobed, hollow, 3-5 cm in diameter, and covered with evenly distributed sori. However, two forms within the species are recognized: f. ulvoidea for globose forms and f. difformis for lobed forms. Plastid rbcL and nuclear ITS region sequences were newly determined in samples of S. ulvoidea from the Pacific coast of the North America. We found little variations in the ITS sequences among samples of S. ulvoidea from five different locations and in the rbcL region from two different locations. These results do not support previous classification of f. ulvoidea and f. difformis within the species. All analyses of our rbcL sequence dataset show that Soranthera was placed in the Chordariaceae s.l., but more related to Botrytella than Punctaria and Asperococcus.

A study on the measurement of the nucleated red blood cell (nRBC) count based on birth weight and its correlation with perinatal prognosis in infants with very low birth weights

  • Purpose: The aim of this study was conducted to investigate the mean nRBC count in very low births weight infants (VLBWIs) and to determine the usefulness of the nRBC as an independent prognostic factors of perinatal complications in VLBWIs. Methods: This study was conducted on 112 VLBWIs who were hospitalized in the neonatal intensive care unit (NICU) of the author's hospital within the period from March 2003 to and May 2008. Based on the infants' nucleated red blood cells (nRBC) counts at birth, on the third day after birth, on the seventh day after birth, in the second week after birth, and in the fourth week after birth in the medical records, the correlation between nRBC or absolute nRBC counts with birth weight, gestational age, and other perinatal outcomes were retrospectively investigated. Results: In VLBWIs, their mean nRBC and absolute nRBC counts were showing a gradual decrease after birth, and they were consisteantly kept at low values since one week after and inversely proportional to the birth weights. The mean nRBC counts based on the stage after birth showed a significant correlation with perinatal death, necrotizing enterocolitis, and severe intraventricular hemorrhage. Conclusion: The increase in the nRBC count showed a significant correlation with having a severe intraventricular hemorrhage, necrotizing enterocolitis, and perinatal death in VLBWIs. If an increase or no decrease in the nRBC count after birth is observed, newborninfant care precautions should be required.
