Serological characterization of bovine viral diarrhea virus isolates

  • Bovine viral diarrhea viruses (BVDVs) were isolated from cattle with respiratory and diarrhea signs as well as persistently infected cattle. These isolates were analysed serologically to characterize serogroups and to compare serological relationship with reference viruses of type I and II. Most isolates from calf diarrheal cases and persistently infected individuals showed a significant difference in cross-neutralization test with the viruses isolated from nasal discharges showing severe respiratory signs. Serologically most of the commercial vaccine strains could be classified into classical BVDV (type I) such as NADL strain. This serological difference among BVDV isolates suggested the need for new vaccines to protect cattle from both respiratory and enteric BVDV infections in field. The immunogenicity of BVDVs which showed a good propagation capability in MDBK cells and high rates of neutralizing activity (isolate : KD26-1, PHG, B5 and 95002) against all viruses used in this study, was confirmed in guinea pig when treated as single or combined groups.

  • Borna disease(BD) virus 특이항체검출에 대한 세 가지 혈청진단법(간접형광항체법, 세포효소면역반응법, 혈청중화시험)의 정확도를 비교하기 위해 BD virus를 실험적으로 감염시킨 273수의 토끼의 혈청으로 시험하였다. 혈청중 간접형광항체법에 의하여 판정된 123혈청들은 모두 세포효소면역반응법에 의해서 양성으로 판정되었으나 혈청중화시험법에 의해서는 단지 27혈청에서만 양성으로 판정되었다. 혈청중화시험법은 간접형광항체법과 세포효소면역반응법에 비하여 민감도가 훨씬 낮게 나타나 BD virus의 혈청학적 연구에는 간접형광항체법 및 세포효소면역반응법의 활용도가 높을 것으로 생각된다.

수입 소의 검역검사 수준에 따른 블루텅 검출 확률 비교 (Comparison of Probability of Detecting Bluetongue in Quarantine Testing for the Imported Cattle with Special Focus on the Sampling Scenario)

  • In view of free from bluetongue (BT) in the domestic cattle population in Korea, the key of quarantine testing for BT virus (BTV) infection is detection of cattle previously exposed to the virus. The objective of this study was to estimate the probability of detecting a cattle infected with BTV using a stochastic modeling analysis of existing quarantine testing data. Three testing scenarios were considered in this study: serological testing of all animals in all imported lots (scenario 1), serological testing of a sample of cattle from all imported lots (scenario 2), and serological testing of 50% of imported lots (scenario 3). In scenario 2 and 3, it was assumed that cattle were sampled (sample size) within each lot to detect 5% of the cattle in each lot with a 95% confidence, taking into account diagnostic sensitivity of the ELISA (enzyme-linked immunosorbent assay). The model output was the total number of BTV-infected cattle and the prevalence of BTV infection in imported cattle from the US, Australia, Canada and Japan. Compared to the scenario 1, the probability of detecting a BTV-infected cattle was estimated to be 19% and 1.6% in scenario 2 and 3, respectively. Furthermore, the analyses showed a 95% confidence that BTV prevalence was less or equal to $9.7{\times}10^{-4}$ (median = $1.5{\times}10^{-5}$), indicating that, for the scenario 2 and 3 with serological testing for a sample of cattle, the risk of introducing an exotic strain of BTV into Korea through the importation of live cattle would not be acceptable.

A comparison of single dose efficacy of Mycoplasma hyopneumoniae bacterin in swine farms with different serological patterns of PRRSV and PCV2

  • This study was to evaluate the efficacy of single dose Mycoplasma hyopneumoniae (M. hyo)-vaccination in the swine farms which had different serological patterns of porcine reproductive and respiratory syndrome virus (PRRSV) and porcine circovirus type 2 (PCV2). A minimum of 240 pigs from each farm was applied, allocating M. hyo vaccinated and control groups. The PRRSV and PCV2 infections were analyzed by serological method (commercial ELISA kit). After administrating pigs a single dose of M. hyo vaccine or control saline at 3 weeks of age, serum antibodies to M. hyo, PRRSV and PCV2 were monitored at 4, 10, 16 and 22 weeks of age. Mortality, weight changes, feed conversion ratio (FCR) and lung score were also evaluated. A single-dose vaccination of M. hyo bacterin was efficacious to reduce mycoplasmal lung lesions and induce good humoral immune response. However, FCR was improved only in one of the three farms where showed seronegative status to both PRRSV and PCV2 in the period from 4 to 16 weeks of age. These results might imply that M. hyo vaccine alone could not overcome the PRRSV and PCV2 infection-associated wasting in the field condition. Therefore, the control of PRRSV and PCV2 should be considered to obtain the better effects of M. hyo vaccination.

가금인플루엔자 바이러스 항체검출을 위한 혈청학적 진단법 비교 (Comparison of serological methods for detection of avian influenza virus antibodies)

  • An enzyme-linked immunosorbent assay (ELISA) using purified hemagglutinin of swine influenza virus (H1N1) as antigen was developed for detection of antibody to avian influenza virus (AIV). The sensitivity and specificity of a developed and commercial available ELISA kits were compared with those of agar gel precipitation (AGP) test and hemagglutination inhibition (HI) test using sera collected from chickens under condition of field exposure. The concentration of antigen, serum dilution and concentration of enzyme-conjugated secondary antibody in developed ELISA (S-ELISA) were 0.5ug/100ul, 1:200 and 0.03ug/100ul, respectively. The correlation coefficients between S-ELISA and commercial ELISA and HI titers were 0.419 and 0.533, respectively. A significant correlation (p < 0.01) was not found between HI and ELISA titers. The S-ELISA was found to be as more sensitive and specific than the AGP test, showing 86.8% sensitivity and 85.3% specificity. It is suggested that the ELISA using the SIV as antigen may be useful method as an investigating tool for AIV serological surveillance.

Investigation of serum biomarkers for neuropathic pain in neuromyelitis optica spectrum disorder: a preliminary study

  • Background: We aimed to investigate candidates for serological biomarkers of neuropathic pain in individuals with neuromyelitis optica spectrum disorder (NMOSD). Methods: We analyzed 38 sera samples from 38 participants with NMOSD in National Cancer Center. Neuropathic pain was evaluated using the painDETECT questionnaire. Pain with neuropathic components (painDETECT score ≥ 13) was observed in 22 participants, among whom 17 had definite neuropathic pain (painDETECT score ≥ 19). The remaining 16 participants had non-neuropathic pain (painDETECT score < 13). Serum glial fibrillary acidic protein (GFAP) levels were assessed using a single-molecule array assay. Several cytokines, including tumor necrosis factor-alpha (TNF-α), interleukin (IL)-6, IL-10, and IL-17A, were measured by a multiplex bead-based immunoassay. Results: In comparison of NMOSD participants with neuropathic pain components (or definite neuropathic pain) and those with non-neuropathic pain, the absolute values of serum GFAP, TNF-α, IL-6, and IL-10 levels were higher in participants with neuropathic pain components (or definite neuropathic pain), but these findings did not reach statistical significance. Conclusions: Further larger-scale investigations to find reliable serological biomarkers for neuropathic pain in NMOSD are warranted.

혈청학적 진단을 위한 돼지 편충의 체항원, 배설/분비항원의 분리 및 비교 (Isolation and comparison of somatic and excretory-secretory antigens for serological diagnosis in Trichuris suis)

  • Swine whipworm(Trichuris suis) is cosmopolitan nematode which can cause serious pathology in immature stage(larva2~larva5) of infected pigs, such as anorexia, diarrhea, anemia, and death in heavy infections. In this larval stages, it is very difficult to diagnose the infection of whipworm and to differentiate from other common swine gastrointestinal disorders such as 21 day scours which are associated with TGE virus, rota virus, coccidium, and the stress of weaning. In this experiment, the isolated antigens of Trichuris spp. were carried out to examine the structure and specificity of antigens and to select the reasonable antigens which would be used in serological diagnosis by electrophoresis, Western blotting, ELISA. The results of this experiment were as follows; 1. The common fractions of each Trichuris suis antigen were identified 28,32,45, 80kDa by SDS-PAGE with silver stain and four major fractions could be detected in positive swine sera by Western blot analysis. 2. The OD(optical density) values of somatic and excretory-secretory antigens which were reacted against positive(negative) sera from pigs infected with Trichuris suis by ELISA reader were; 1) OD values($mean{\pm}SD$) of adult somatic antigen against positive(negative) sera were $0.30{\pm}0.12(0.09{\pm}0.006)$ and third-stage larva of somatic antigen were $0.28{\pm}0.038(0.10{\pm}0.005)$. And OD values of excretory-secretory antigens of adult and third-stage larva were $0.24{\pm}0.031(0.11{\pm}0.005)$ and $0.08{\pm}0.013(0.10{\pm}0.003)$, respectively. 2) OD values of adult somatic, larval somatic antigen and adult excretory-secretory antigen response to positive sera were significantly (p<0.01) associated with negative swine sera. And the Cut-off OD values(minimum positive value) were determined to be mean negative value plus 3 SD that would minimized the risk of false positives. 3. The OD values of somatic antigens of T suis and T vulpis against swine positive(negative) sera were $0.30{\pm}0.120(0.09{\pm}0.006)$ and $0.25{\pm}0.141(0.09{\pm}0.003)$. These data mean that the somatic antigens of T suis and T vulpis were able to diagnose T vulpis infection in dogs as well as T suis infection in pigs. These results suggest that somatic antigen of third-stage larva and excretory-secretory antigen of adult T suis could be used the diagnostic antigen by serological test(ELISA) in immature Trichuris spp. infection.

돼지 생식기호흡기증후군바이러스의 농장단위 방역대책 수립을 위한 혈청학적 및 바이러스학적 감염유형 분석법 적용 및 비교 (Comparison of Serological and Virological Analysis for Infection Patterns of Porcine Reproductive and Respiratory Syndrome Virus to Establish a Farm Level Control Strategy)

  • Porcine reproductive and respiratory syndrome virus (PRRSV) has plagued pig populations worldwide causing severe economical impacts. In order to establish effective strategies for prevention of PRRS, infection patterns on the herd level are primarily evaluated. In the present study, therefore, serological and virological analyses were conducted in 20 pig farms suffering from PRRS. Seroprevalence levels in each farm were grouped into 3 patterns: SN (Stable sow groups/Not infected piglet groups, SI (Stable sow groups and Infected piglet groups), and UI (Unstable sow groups and Infected piglet groups). The rates of each serological pattern were 15% (n=3), 10% (n=2), and 75% (n=15), respectively. In addition, the pattern analysis was extended to virological monitoring on the same farms that further included suckling pig groups. As a result, the infection pattern was classified into 4 categories: SNI (Stable sow groups/Not infected suckler groups/Infected piglet groups), SII (Stable sow groups/Infected suckler groups/Infected piglet groups), UNI (Unstable sow groups/Not infected suckler groups/Infected piglet groups), and UII (Unstable sow groups/Infected suckler groups/Infected piglet groups). The rates of each viroprevalence were estimated at 50% (n=10), 30% (n=6), 10% (n=2), and 10% (n=2), respectively. PRRSV viroprevalence results of suckling pig groups revealed that 8 farms were considered virus positive. In 2 farms among these farms, PRRSV appeared to be transmitted vertically to suckling piglets from their sows. In contrast, piglet-to-piglet horizontal transmission of PRRSV seemed to occur in sucking herds of the remaining farms. Thus, this virological analysis on suckling piglets will provide useful information to understand PRRSV transmission routes during the suckling period and to improve a PRRS control programs. Our seroprevalence and viroprevalence data found that infection patterns between sow and piglet groups are not always coincident in the same farm. Remarkably, 15 farms belonging to the UI seroprevalence pattern showed four distinct viroprevalence patterns (SNI; 7, SII; 4, UNI; 2 and UII; 2). Among these farms, 11 farms with unstable seroprevalence sow groups were further identified as the stable viroprevalence pattern. These results indicated that despite the absence of typical seroconversion, PRRSV infection was detected in several farms, implying the limitation of serological analysis. Taken together, our data strongly suggests that both seroprevalence and viroprevalence should be determined in parallel so that a PRRS control strategies can be efficiently developed on a farm level.

  • 류마티스 관절염은 주로 관절의 활막 세포의 증식, 병리학적 면역 반응과 관절의 진행성 파괴를 특징으로 하는 만성 염증성 질환이다. 이 질환은 중요한 사회적 건강 문제로 대두된다. 이 논문을 통해 분자수준에서 변화와 류마티스 관절염의 진단 또는 질병의 진행에 대한 발병 기전을 새로운 관점에서 발병기전을 제공한다. 또한 조기 진단과 류마티스 관절염의 예후에 도움이 될 것으로 보인다. 새로운 혈청학적 및 면역학적 바이오마커에 초점을 맞추고 있다. 따라서 넓게 질병 진행의 위험 환자를 식별하는 혈청학적, 생체 면역학적 요인 등을 규명하고 적용시키는 것을 도출할 수 있다. 진단 검사 의학을 기반으로 하는 증거는 환자에게 최선의 결과를 제공할 수 있다. 마지막으로. 최근의 연구 데이터는 이 접근을 통해서 치료를 최적화하기 위해 조기 진단 및 치료에 도움이 되는 새로운 접근 방식으로 궁극적인 유용성을 정립하는데 도움이 될 것으로 사료된다.

개 혈액 재료에서의 Brucella 검출을 위한 진단방법의 비교 (Comparison of diagnostic methods for detection of Brucella species in dog blood samples)

  • Canine brucellosis produce abortions and infertility in dogs and is currently diagnosed by serological methods such as rapid slide agglutination test with 2-mercaptoethanol (2-ME RSAT) and immunochromatographic assay (ICA). Bacterial isolation is considered gold standard for Brucella diagnosis and the polymerase chain reaction (PCR) is an alternative method to bacterial isolation. A total of 36 whole blood samples were collected from dogs reared in area of Chuncheon and were subjected to serology (2-ME RSAT and ICA for B. canis, Rose Bengal test and C-ELISA for B. abortus), blood culture and 3 types of PCRs (BSCP31, 16s rRNA, and OMP-2). All blood samples were negative by serology and blood cultures. The BCSP31 and the OMP-2 PCR detected 5 samples were positive whereas the 16S rRNA PCR detected all samples were negative as serological methods and blood culture did. From the results observed in the present study, we conclude that 16S rRNA PCR could be used for direct PCR for canine blood samples.
