Effect of Sodium Acetate on the Production of Sisomicin by Micromonospora inyoensis (Micromonospora inyoensis에 의한 Sisomicin 생산에서 Sodium Acetate의 영향)

  • This study was carried out to enhance the production of sisomicin by Micromonospora inyoensis ATCC 276000. The effect of various sugars and organic acids as the supplements to a basal mineral medium was tested for the sisomicin production by fully grown mycelium. Among the substances tested, acetate and citrate greatly increased the production of sisomicin at the sixth day of culture. Especially, in the basal mineral medium containing 0.1M sodium acetate, sisomicin production was 6.7 times more than that in the same medium with glucose. When 0.1M sodium acetate was added in the fermentation medium initially, the cell growth was inhibited by sodium acetate, although specific productivity was higher than that in the same medium without sodium acetate. On the contrary, when sodium acetate was added to the culture after three days, the sisomicin production and specific productivity were 1.6 times higher than those in the same medium without sodium acetate. The results suggested that sodium acetate was a stimulating substance of sisomicin production by M. inyoensis.

The Effect of Sodium Acetate in Alkaline Treatment of Acetate Fabrics (아세테이트 직물의 NaOH 처리시 무기염 첨가에 따른 영향)

  • The effect of sodium acetate to reduce the fiber damage and hardening of acetate fabrics during alkaline treatment is studied. The optimal condition is controlled concentration 2%, at $50^{\circ}C$ for 6 minutes and at $70^{\circ}C$ for 2 minutes through the result of weight loss, shrinkage and tensile strength. Alkaline treated acetate fabrics under optimal condition show softer than untreated acetate fabrics. Alkaline treatment with sodium acetate brings the reduction in hardening and shrinkage in internal fiber of acetate fabric. Also, alkaline treatment with sodium acetate improves the tensile strength of acetate fabrics compared with only alkaline treatment. The moisture regain of acetate fabrics is also improved by alkaline treatment under optimal condition.

  • 1. The optimum temperature of amylase activity produced by Bacillus subtilis var. M-181 was $50^{\circ}C$, and its activity was lost by heating to $70^{\circ}C$, 10 minutes without addition of salts. 2. Addition of sodium salts effects for heat resistance of the amylase affected differently by kinds of the salt. Among organic sodium salts monosodium glutamate, sodium acetate as sodium propionate affected on heat resistance of the amylase relatively better effects. 3. Addition of 10mg of sodium sulfate per ml of enzyme solution $({D_{30}}^{40^{\circ}}\;1250/ml)$, showed maximum affect on the neat resistance. 4. Coexistence of calcium acetate and sodium acetate, affected on the hear resistance, remarkably.

  • Experiments were carried out to develop of mutant strain from Pichia ciferrii ATCC 14091 and investigate the effect of sodium acetate on the production of tetraacetylphytosphingosine (TAPS). A high-TAPS producing mutant was simply selected by staining with sudan black B and colony shape after the treatment of UV and NTG. Sodium acetate enhanced the TAPS production.

  • 본실험의 목적은 탄산기에 방사성동위원소 C14를 표식한 sodium acetate ($CH_3$C14 OONA)를 사용하여 염소의 위내에 존재하는 acetate 의 위벽으로부터의 흡수율과 위의 acetate의 평균함량을 측정하는데 있다. C14 로 표식된 sodium acetate(specific activity 1.35$\times$108 cpm./g.)를 급사 3 시간후의 염소의 위내에 주입하고 주입 2분후부터 약 2분간격으로 위 내용물을 추울하여 column chromatograpy를 이용하여 acetate를 분리정량한 후 그의 specific activity를 측정하였다. 주입후 3 분경까지는 위내에 존재하는 acetate에 의한 표식 acetate의 희석으로 말미암아 specific activity 는 급격히 감소되어 갔고 3 분후부터는 감소도가 비교적 완만하였으나 역시 계속적으로 감소되어갔다. 희석완료후의 이 specific activity 감소는 위벽을 통한 acetate를 흡수와 위 내용물로부터의 acetate 생성으로 인한 것으로서, 이 감소율로부터 acetate의 위벽흡수속도를 추정할 수 있다. 상기 specific activity의 감소 graph 로부터 추정된 위내 acetate의 량은 본실험의 제조건하에서 약 30 g이었으며 위내 acetate 의 specific activity가 1/2 로 감소되는데 요하는 평균 시간은 약 4 분이었다. 이는 위내에 존재하는 acetate량의 약 절반은 4 분동안에 위벽을 통과함을 의미한다.

Pavement Impact Evaluation of Basic Materials of Airport Airside Deicers (공항 airside용 제설제의 기본물질에 대한 포장 영향성 평가 연구)

  • OBJECTIVES : This is a basic research for the domestic production of airport-airside deicers. This research selected basic materials for deicers appropriate for the pavement of domestic airports by evaluating the deicing performances of basic materials used in international-standard airport deicers and their impacts on pavements. METHODS : Laboratory investigation was conducted to evaluate the asphalt surface tensile strength, concrete scaling impact, ASR impact, and deicing performances of sodium formate (NaFm), potassium formate (KFm), sodium acetate (NaAc), and potassium acetate (KAc), which are the basic de-icing materials commonly used at international airports, approved by the FAA. In addition, the analyses were also performed on the airside deicer urea, which is currently used in domestic airports. RESULTS : Laboratory investigation confirmed that sodium formate, potassium formate, sodium acetate, and potassium acetate had superior surface tensile strength, concrete scaling impact, and deicing performance compared to airside urea, but they also had greater impacts on concrete ASR. Among these materials, sodium formate had the best asphalt surface tensile strength, concrete scaling impact, and deicing performance, while also having the greatest impact on ASR; hence, mitigation plans for ASR were needed, if it were to be used as airport-airside deicer. CONCLUSIONS : It is necessary to consider additional additives to prevent ASR of concrete pavements when developing airport-airside deicers using sodium formate, potassium formate, sodium acetate, and potassium acetate.

Effect of Color Developing by Heat Treatment on Cotton Fabrics Dyed with Catechu Extract (아선약염색 면직물의 열발색 공정에 의한 발색효과)

  • In order to clarify the availability of heat treatment for catechu dyeing, effect of color developing by heating process on cotton fabrics dyed with catechu extract was investigated. Dyed fabrics were heated various time(min) at $150^{\circ}C$. The cases of non-mordanting and mordanting were compared, and examined the effect of sodium acetate as an agent to promote the color developing to fabrics. Experiments with after-mordanting method showed that various colors can be obtained using catechu. K/S values of dyed fabrics with Al, Cu, Fe mordant were higher than the case of no mordant. The fabric by Cu mordanting showed 3 times to the case of no mordant. Fabrics dyed with catechu by mordanting lowered $L^*$ value while there was little change in $a^*$, $b^*$ value as the heating process progressed. Therefore, K/S values were increased due to heating process. The effects of adding sodium acetate in dyeing bath on dyed fabrics were shown higher K/S values, and the $L^*$ values were lower than those without sodium acetate according to heating time increase. The result show sodium acetate had a promoting effect on the color developing to the fabrics by heating process, it was very effective.

Phosphate and Carbon Source Regulation of Alkaline Phosphatase and Phospholipase in Vibrio vulnificus

  • In this study, the effects of phosphate concentration and carbon source on the patterns of alkaline phosphatase (APase) and phospholipase (PLase) expression in Vibrio vulnificus ATCC 29307 were assessed under various conditions. The activities of these enzymes were repressed by excess phosphate (4 mM) in the culture medium, but this repression was reversed upon the onset of phosphate starvation in low phosphate defined medium (LPDM) containing 0.2 mM of phosphate at approximately the end of the exponential growth phase. The expressions of the two enzymes were also influenced by different carbon sources, including glucose, fructose, maltose, glycerol, and sodium acetate at different levels. The APase activity was derepressed most profoundly in LPDM containing fructose as a sole carbon source. However, the repression/derepression of the enzyme by phosphate was not observed in media containing glycerol or sodium acetate. In LPDM-glycerol or sodium acetate, the growth rate was quite low. The highest levels of PLase activity were detected in LPDM-sodium acetate, followed by LPDM-fructose. PLase was not fully repressed by high phosphate concentrations when sodium acetate was utilized as the sole carbon source. These results showed that multiple regulatory systems, including the phosphate regulon, may perform a function in the expression of both or either APase and PLC, in the broader context of the survival of V. vulnificus.

Study on the Latent Heat Characteristics of the Organic Compound, $C_{28}H_{58}$ and the Inorganic Compound, $CH_3COONa{\cdot}3H_2O$ (유기잠열재, $C_{28}H_{58}$과 무기잠열재, $CH_3COONa{\cdot}3H_2O$의 잠열특성연구)

  • In this research, Octacosane($C_{28}H_{58}$) and Sodium Acetate Trihydrate($CH_3COONa{\cdot}3H_2O$) were selected as latent heat storage materials to store off-peak electricity or waste heat of an industrial plant. Experimental analyses were performed in terms of the variation of phase change temperature and latent heat, phase change stability for the long term utilization. The results were as follows. 1. The phase change temperatures of industrial grade Octacosane and Sodium Acetate Trihydrate were $60.7^{\circ}C$ and $57.4^{\circ}C$, the latent heat were 60.6kcal/kg and 51.1kcal/kg respectively. 2. The latent heat quantity of Octacosane was decreased with the increasing number of phase change cycles. It decreased from 60.6kcal/kg to 47.2kcal/kg upto 200 cycles and then no variation was observed after 200 cycles. 3. To prevent the supercooling of Sodium Acetate Trihydrate, the nucleating agent, Sodium Pyrophosphate Decahydrate of 3 wt% was added, and then the supercooling temperature (Tm-Tsc) was decreased from $25.7^{\circ}C$ to $1^{\circ}C$. The phase separation was disappeared by the addition of CMC-Na of 3 wt% as a thickener. It was found that the optimal quantity of nucleating agent and thickener was 4wt% considering the stability of SAT as a latent heat storage material. 4. The phase change temperature of Sodium Acetate Trihydrate($CH_3COONa{\cdot}3H_2O$) was adjusted from 57.4 to $46.2^{\circ}C$ by the addition of UREA. And then the latent heat quantity was decreased from 51.1 to 38.3kcal/kg. 5. When the heat storage capacities between the sensible and latent heat storage materials were analyzed and compared in heating process from 30 to $90^{\circ}C$, the heat storage capacity of Octacosane was 2.45 times larger than water and 12.5 times than granite at $60.7^{\circ}C$, and the heat storage capacity of Sodium Acetate Trihydrate was 2.53 times larger than water and 12.91 times than granite at $57.4^{\circ}C$.

Effect of Sodium Acetate and Calcium Chloride on Characteristics of Kakdugj (Sodium Acetate와 Calcium Chloride를 첨가한 깍두기의 특성)

  • This study was undertaken to examine if sodium acetate and/or calcium chloride would increase the firmness and/or retard rancidity of Kakdugj (Korean seasoned pickles of cubed radish roots) during short $(at\;20^{\circ}C)$ or long $(at\;4^{\circ}C)$ period of fermentation During the fermentation periods, pH and acidity were higher in Kakdugis containing 0.3 or 0.6% Na-acetate than in the other samples at both storage temperatures. Firmness was increased with addition of 0.05% Calcium chloride and synergistic effect on firmness was observed when Calcium chloride and Na-acetate were added together. Sourness did not decreased in samples containing 0.3% Na-acetate at either fermentation periods but decreased in those containing 0.6% Na-acetate and stored for long. Saltiness was increased slightly with the addition of Calcium chloride. Compression test gave closer result to sensory evaluation than puncture test In the measurement of firmness with Instron.
