Effects of Temperature and Plant Growth Regulators on Bud Sprouting of Stem Cutting of Hibiscus syriacus 'Honghwarang' (온도(溫度)와 생장조절물질(生長調節物質)이 무궁화(Hibiscus syriacus ''Honghwarang') 삽수의 맹아(萌芽)에 미치는 영향)

  • Various temperatures and plant growth regulators were assessed to investigate the nature of the dormancy and bud sprouting of stem cutting of Hibiscus syriacus 'Honghwarang'. The bud sprouting was promoted as temperature increased. Under $30^{\circ}C$ condition, stem cuttings began sprouting at 4days after cutting and showed 100% sprouting at 6 days after cutting. With GA or BA treatment, the cuttings started sprouting at 6 days after cuttings under $25^{\circ}C$ condition and reached 65% sprouting, which was not different from the results without hormone treatments. However, ABA dipping under below $25^{\circ}C$ condition resulted in low sprouting, whereas under over $25^{\circ}C$ condition they started sprouting at 6 days after cutting and reached 93% at 12 days after cutting, which indicated that ABA is not effective on inhibition of sprouting under high temperature condition.

  • To develope the soybean sprouting equipment(a type of cabinet) for medium amount(one bucket; 10~13kg) production of soybean sprouts, the watering system was made of four siphons and four the bottle necked water tank. Diameter of four siphons used for development of the watering system were 8, 13, 17 and 21mm, respectively and their water exhausting volumes were 40, 92, 154, 524ml/sec, respectively. The watering system can be automatically watered 524ml/sec without electric supply. The soybean sprouting equipment is consisted of four the bottle necked water tanks with the four siphons and six sprouting buckets with net shaped bottom for water draining. The soybean sprouting equipment with the watering system may be produced more than one box a day in the condition of 6~7 watering times a day at 25~30oC.

Sprouting Condition of Crown Bud and Plug Seedling Production in Yacon

  • The objectives of this study were to find the sprouting condition and to establish the optimum production methods of plug seedlings in yacon (Polymnia sonchifolia Poepp. & Endl.). The sprouting ratio was greatest at $30\pm 1^{\circ}C$ at 20 days after planting. Crowns with single buds were more effective than those with two or more buds for sprouting, which might be due to the apical dominance. Planting the shoots separated from crown after sprouting in the single- and double-layer polyethylene-covered greenhouses reduced seedling period with 25% and 50%, respectively. Planting the shoots after sprouting was more effective than planting the crown buds. Double-layer polyethylene-covered green-house was good for plug seedling production than open field or single-layer polyethylene-covered greenhouse. The bed soils composed of clay loam : compost or sand : compost (1:l=v:v) were more effective to produce plug seedlings than only clay loam, sand or compost. Seedlings could be produced at 30 days after planting in our studies.

Studies on the Cause and Control for Non-sprouting Bud in the Utility Reclaimed Hill Side Mulberry Field. (개간지 상전에 발생하는 발아불량 현상의 원인 및 방제에 관한 연구)

  • These studies are conducted to find out inducing factors for the non-sprouting bud which appears in the utility reclaimed hill mulberry held and to establish control method for it since 1971. The results are as follows: 1. In spring, winter bud does not germinate at all on the top of branches or does wither suddenly after or during its germination. Necrosis and browning are appeared in cortex and phloem of non-sprouting bud branch. In autumn, the deterioration of leaves, the abnormal leaves on the top of branch, and browning of veins or petioles are also observed. 2. The soil of non-sprouting bud mulberry Held were found to be sand loom derived from granite rocks. The sub soil of it was very poor in development of root system because it had hard soil and lower pore space ratio. Especially, the sub soil to bring about severe non-sprouting bud had been easily appeared with deficiency of moisture content because of high density solid. 3. Content of soil moisture was significantly lower in the severe non-sprouting bud soil than in the healthy field. The sub soil o( the healthy field contained proper moisture content. On the other hand, the sub soil of the severe non-sprouting bud held contained almost critical moisture content for wilting. 4. The depth of available soil was shallow in the non-sprouting bud than in the healthy fold. The more rate of the non-sprouting bud was severe, the more available soil depth was sallow. 5. Available boron content in soil was affected by moisture content in soil. There was. lower moisture content in the non-sprouting bud field than that in healthy fold during 5, June to 5, September. 6. There was no significant correlation between soil pH and available boron content. On the other hand, the correlation of the content of organic matter and available boron content appeared to be highly positive significance. 7. The quantity of boron was significantly contained more in healthy mulberry field (0.34∼0.43ppm) than in non-sprouting bud field (0.10∼0.28ppm). 8. Boron content in leaves and barks was significantly lower in the non-sprouting bud trees than in healthy trees. 9. The symptom of non-sprouting bud induced from boron free sand culture was similar with that arised in the non-sprouting bud field. 10. The rate of non-sprouting bud was high by the increased application of lime. Considering the facts mentioned the above, author may conclude that the non-sprouting bud of mulberry tree is caused by boron deficiency, but also it is affected by the parent rocks, organic matter, soil moisture content and lime application. 11. The non$.$sprouting bud may be completely controlled by the application of 6∼9kg borax per 10a mulberry field twice a year in spring and summer.

The variation of Sugar and Sprouting stem in Gibberellin Treated seed potato (Gibberellin 처리가 종서의 당함량 및 붕아경의 생장구조에 미치는 영향)

  • This study aimed to contribute fundamental study of sprouting in Gibberellin treated potatoes, and studied about Glucose and Sucrose amount and its sprouting stem structure in it sprouting seed potatoes with Gibberellin treatment. The results obtained are as follow; The quantity of glucose in gibberellin treated seed potatoes showed remarkable increase in comparison with that of those untreated, and the increment went up as the seeding date pass. As to the concentration of gibberellin, the 10 ppm plot contained more glucose than 5 ppm plot but 14th after seeding, it was found that this tendency was reversed. The increasing tendency of the quantity of sucrose in gibberellin treated seed potatoes were similar to that of glucose. The sprouting stems of the gibberellin 10 ppm and 5ppm plot were all longer than the controls. And after 14 days on account of thin ing growth by gibberellin treatment, the sprouting stems of the 5ppm plot were longer than 10ppm plot. The microscopic vertical and cross section of the gibberellin treated sprouting stems showed larger cells than those of untreated. The cells of sprouting stem treated by gibberellin 10 ppm were larger than those of the stems treated 5ppm.

Influence of Rainfall During the Ripening Stage on Pre-Harvest Sprouting, Seed Quality, and Longevity of Rice (Oryza sativa L.)

  • The influence of rainfall during the ripening stage on pre-harvest sprouting, seed viability, and seed quality was investigated in two Korean rice cultivars, Shindongjin and Hopum. When the rainfall was artificially treated in a greenhouse, HP started to pre-harvest sprouting at three days of rainfall treatment (DRT), but Shindongjin did not show pre-harvest sprouting at 40 DAH treatment and just 0.3~0.8% at 50 DAH, which was much lower than 15.3~25.8% of Hopum in the same treatment. After harvest, the seed germination of Hopum decreased about 10~25% compared to non-treated seeds, but that of Shindongjin decreased much little rate than that of Hopum. The seed longevity tested by accelerated aging decreased with prolonged rainfall period in both cultivars, but the varietal difference was clear; Shindongjin could withstand longer accelerated aging than Hopum. Shindongjin maintained its germination (>50%) ability after 15 days of accelerated aging regardless of the rainfall treatment period and time, but Hopum dropped below 50% germination ability after only 5 days of accelerated aging. In conclusion, rainfall during the ripening stage induced not only pre-harvest sprouting, but also reduced seed quality and longevity during storage, which varied between two cultivars.

Changes in Isoflavones Contents and Germination Characteristics of Germinated Soybeans [Glycine max.] under Light Condition (명조건에서 발아시킨 콩의 발아특성 및 isoflavone함량 변화)

  • This research was conducted to investigate changes in isoflavone contents and sprouting characteristics of germinated soybeans [Glycine max.] under light condition. The relative wet weight of soybeans increased as germination progressed, and it reached $291.1{\pm}14.3%$ at 60 h after beginning of sprouting. On the other hand, relative dry weight slowly decreased as germination progressed. The length of sprouting rapidly increased as germination progressed. The sprouting rate sharply increased from 12 h to 48 h and reached $96.4{\pm}2.5%$ at 60 h after sprouting. In terms of color, L value did not change until 24 h after the beginning of germination, but it rapidly increased from 24 h to 36 h and did not change thereafter. The a value rapidly increased at 24 h after the beginning of germination and decreased thereafter. The b value rapidly increased at early and last stages and reached $9.0{\pm}0.7$ at 60 h after the beginning of sprouting. Total isoflavone reached a maximum level at 12 h after the beginning of germination and consistently decreased thereafter. Among isoflavone compounds, m-genistin had the highest content followed by daidzin and m-daidzin.

Studies on the Sprouting Characteristics of Scirpus planiculmis and Storage Methods of Corm for the Herbicide Screening (새섬매자기(Scirpus planiculmis)의 출아특성(出芽特性)과 제초제(除草劑) 스크리닝을 위한 구경(球莖)의 보관방법(保管方法))

  • The sprouting characteristics of Scirpus planiculmis and storage methods of corm for the herbicide screening were investigated under both laboratory and greenhouse conditions. Sprouting and elongation of S. planiculmis corms were continued during storage at low temperature ($5^{\circ}C$) and wet condition. However, sprouting of corm was failed when the surface water was removed from the corm, and elongation was reduced by the treatment of 0.5-1.0% sodium chloride(NaCl) solution. Alternate temperature was more effective on the sprouting of S. planiculmis corm than constant temperature. Optimum temperature for the sprouting of S. planiculmis corm was alternation between $25^{\circ}C$ to $35^{\circ}C$. Storage at $-20^{\circ}C$ for 10 min. or 14.7% dehydration made the corm of S. planiculmis to fail for sprouting. S. planiculmis corm was more tolerant to sodium chloride than Echinochloa crus-galli or Oryza sativa(Donginbyeo) seeds. Sprouting rate was decreased by cutting of the corm. Sprouting percent by vertical cuttings, horizontal cuttings, and vertical plus horizontal cuttings were 72, 56, and 28%, respectively.

Microbiological Evaluation in situ of Each Process in Seed Sprouting (새싹채소 생산현장에서 재배공정별 미생물학적 위해 평가)

  • The consumption of raw sprouts has increased in popularity worldwide because the food is natural and healthy. However, in Korea, nothing is known on the safety standards of sprout producers or changes in the microbial populations of sprouts during sprouting. We evaluated the microbial safety and quality of sprouts during each step in the sprouting process. Bacteriological analysis showed that seeds had a Total Plate Count (TPC) ranging from 3.04 - 5.21 log CFU/g and coliform counts ranging from 1.80 - 3.86 log CFU/g. TPC and coliform counts increased rapidly during the sprouting process to attain values of 6.99 - 8.26 and 3.70 - 7.15 log CFU/g, respectively, regardless of decontamination of seeds with commercial sanitizer. TPC and coliform counts were on high level after sprouts were washed. Escherichia coli was detected in samples of domestic radish sprouts at all stages from seed to storage, rape sprouts in the stages from soaked seed to storage, and red radish sprouts during sprouting, and no sanitizer was used in any of these processes. Untreated red radish sprouts were also positive for Bacillus cereus at all processing steps and Listeria monocytogenes after germination. However, pathogens were not detected at any sprouting stage of seeds treated with sanitizer. It is necessary to carefully control commercial sprouting, and to develop HACCP guidelines applicable to all sprouting processes, commencing at the first step in raw seed production.

Effects of cold and room temperature storage on the sprouting and marketability of early-grown spring potatoes

  • Three spring potato varieties (Atlantic, Chubaek, and Superior) were cultivated for 80, 90, and 100 days and compared in terms of their total and marketable yields and specific gravities. Subsequently, changes in the quality parameters (weight loss, sprouting rate, disease incidence, dry weight, and specific gravity) of potatoes cultivated for 100 days were monitored during two-month storage in improved and semi-underground warehouses at 4℃ and room temterature. The productivities of Atlantic, Superior, and Chubaek were maximized at cultivation periods of 100, 100, and >90 days, respectively. In all cases, cold storage resulted in <5.3% weight loss and no marketability loss. However, two-month room temperature storage resulted in pronounced marketability loss due to weight loss (6.2, 7.3% and 10.9% for Atlantic, Superior, and Chubaek, respectively) and sprouting (in ~13%, >75%, and 99% of Atlantic, Superior, and Chubaek tubers, respectively). The marketability of Atlantic, Superior, and Chubaek during cold storage was maintained for >2, ~2, and <2 months, respectively. All varieties were characterized by rapid sprouting, low dry weight and specific gravity, and rapid marketability loss during storage.
