Structural Alterations of Wogonin Significantly Reduce the Inhibitory Activity against COX-2 Catalyzed PGE2 Production from LPS-Induced RAW 264.7 Cells

  • Structural alterations of wogonin were conducted to observe the role of each functional group at the A-ring of wogonin on the inhibitory activities against COX-2 catalyzed $PGE_2$ production from LPS-induced RAW 264.7 cells. Serial deletion of the functional groups at the A-ring of wogonin exhibited low to comparable bioactivity. The present study validated that all the functional groups at the A-ring of wogonin are important factors to exhibit the optimum inhibitory activities.

  • Atratoxin $B_1$ which was found to inhibit the growth of Candida albicans caused structural and morphological alteration of the cells. Increased accumulation of vesicles and membranous bodies in the cytoplasm, and alterations of the cell membrane and cell wall were most obvious. Sequential lytic events of the cells eventually resulted in complete disintegration of the cytoplasmic structures. These results suggested that atratoxin $B_1$ functioned by either blocking the biosynthetic step during cell wall synthesis, altering cell wall metabolism or dissolution of the cell organelles. These changes caused a progressive destruction of the cell wall leading to cell lysis.

  • It is generally accepted that the surface quality of wafer edge is mostly damaged by the vibrations of the edge grinding machine. The surface quality of wafer edge is supposed to be the most dominant factor of the cracks, scratches, burrs and chips on the edge surfaces, which are the main defects of the wafers. In this study, the structure of a wafer edge grinder has been investigated through the frequency response experiment and the computer simulation to find ways to suppress the vibrations from the structure. The main reasons of the structural vibrations were analyzed. And further the design alterations were deduced from the results of the experiment and the simulation, and applied to the machine to check the effects of those alterations and to eventually improve the structural stability. The result shows that the machine can have much improved stability with relatively simple design changes.

  • The modern industry requires the precision machining as well as the high productivity. The machine tool structure should be evaluated in aspects such as durability, static stability, precision rate and the dynamic stability which is one of the most critical characteristics in determining the magnitude of vibrations. In this study, the dynamic properties of a pipe cutting machine were investigated to analyze the structural vibrations of the machine, and further to improve the structural stability and precision machining. Frequency response test and computer simulation have been utilized for the analysis and the design alterations. And the result shows that proposed design alterations can reduce the vibrations of the machine substantially.

  • Laser welding is widely used for precision welding because of superior mechanical properties and high productivity. Generally the accuracy of the welding is determined by the distribution of the bead which is affected by the structural vibrations of the equipment. This study was originated to stabilize a laser welding machine to minimize the bead distribution for the precise joining. The structural properties of the laser welding machine have been investigated to analyze the major factors of the vibrations to cause the bead distribution. The ideas for the design improvement have been applied to the simulation model to identify the effects and further to achieve the stability design and to minimize the bead distribution. The result shows that a few simple design alterations can substantially suppress the structural vibrations and improve the welding accuracy. The procedure used for this study can also be applied to similar welding equipments for improving the structural stability and the welding accuracy.

Structural Design of a Draw Tower for the Improved Stability and the Suppressed Wire Breakage (단선억제 및 가공정도 향상을 위한 Draw Tower의 구조 안정화 설계)

  • Wire breakage in drawing, which is the most critical process in glass fiber manufacturing, is caused by numerous factors such as height, drawing speed, and air flow. The vibrations of the draw tower, however, is the most dominant factor to create the wire breakage. In this study, the structure of a draw tower has been analyzed through experiments and computer simulations to figure out the main reasons of the vibrations. And the design alterations were applied to the machine to identify the effects of those alterations. The result shows that design alterations of the draw tower structure can suppress the 87% of the vibrations, and further can prevent the wire breakage.

  • The aim of this study was to detect longitudinal alterations on lumbar vertebral trabecular bone quality (microarchitecture and degree of mineralization) and bone mineral density (BMD) during pregnancy Virginal eighteen mice were used. Then, twelve mice were mated. Mice lumbar vertebrae were scanned before mate, at 7 days of pregnancy (early pregnancy, 6 mice) and immediately after delivery (late pregnancy, 6 mice) by using in-vivo micro-computed tomography Structural parameters, degree of mineralization and BMD were measured. During early pregnancy, there were no significant alterations on structural parameters, degree of mineralization and BMD. At late pregnancy, Tb.Th (11.8%) and BMD (12.7%) were significantly decreased and Tb.N (6.3%), Tb.Pf (43.0%) and BS/BV (15.1%) were significantly increased (p<0.05). Additionally, the lower degree of mineralization was increased, although, the higher degree of mineralization was decreased. These results indicated that the quality and BMD might be not affected during early pregnancy. At late pregnancy, however the bone quality and BMD were likely to be negatively affected.

  • Strengthened glass is widely used in the display industry for the smart phone and the tablet PC's, the market of which shows steady growth. The strengthened glass is prone to the fracture due to the machined surface defects such as crack and notch. The surface defects are caused mostly by the vibrations of the laser cutter. In this study, the vibrations of the laser cutter were investigated through the frequency response experiment and the computer simulation. The main reasons of the structural vibrations were analyzed, and further the design alterations were deduced and applied to the machine to check the effects of those alterations and to eventually improve the structural stability. The result shows that simple design modifications without major structural change can substantially suppress the vibrations, and improve the quality of the machined surface.

FE model updating method incorporating damping matrices for structural dynamic modifications

  • An accurate finite element (FE) model of a structure is essential for predicting reliably its dynamic characteristics. Such a model is used to predict the effects of structural modifications for dynamic design of the structure. These modifications may be imposed by design alterations for operating reasons. Most of the model updating techniques neglect damping and so these updated models can't be used for accurate prediction of vibration amplitudes. This paper deals with the basic formulation of damped finite element model updating method and its use for structural dynamic modifications. In this damped damped finite element model updating method, damping matrices are updated along with mass and stiffness matrices. The damping matrices are updated by updating the damping coefficients. A case involving actual measured data for the case of F-shaped test structure, which resembles the skeleton of a drilling machine is used to evaluate the effectiveness of damped FE model updating method for accurate prediction of the vibration levels and thus its use for structural dynamic modifications. It can be concluded from the study that damped updated FE model updating can be used for structural dynamic modifications with confidence.

  • The powerful data mapping capability of computational deep learning methods has been recently explored in academic works to develop strategies for structural health monitoring through appropriate characterization of dynamic responses. In many cases, these studies concern laboratory prototypes and finite element models to validate the proposed methodologies. Therefore, the present work aims to investigate the capability of a deep learning algorithm called Sparse Autoencoder (SAE) specifically focused on detecting structural alterations in real-case studies. The idea is to characterize the dynamic responses via SAE models and, subsequently, to detect the onset of abnormal behavior through the Shewhart T control chart, calculated with SAE extracted features. The anomaly detection approach is exemplified using data from the Z24 bridge, a classical benchmark, and data from the continuous monitoring of the San Vittore bell-tower, Italy. In both cases, the influence of temperature is also evaluated. The proposed approach achieved good performance, detecting structural changes even under temperature variations.
