Simultaneous analysis of sulfonamides in beef and pork by high performance liquid chromatography and electrospray ionization mass spectrometry (HPLC와 LC/MS에 의한 식육내 잔류 설파제의 동시 분석법)

  • A multiresidual analysis was performed to determine 12 sulfonamides(sulfacetamide, sulfadiazine, sulfisomidine, sulfathiazole, sulfapyridine, sulfamerazine, sulfamethazine, sulfamonomethoxine, sulfisoxazole, sulfamethoxazole, sulfaquinoxaline, and sulfadimethoxine) in beef and pork simultaneously. The multiresidual analysis for the sulfonamides currently used was able to analyze 5 kinds of sulfonamides at the same time. The method of this 12 sulfonamides multiresidual analysis in this study was matrix solid-phase dispersion(MSPD) by high performance liquid chromatography (HPLC) and liquid chromatography mass spectrometry (LC/MS). The recovery rate of the materials was measured by MSPD method with 3 different extraction solvents; Dichloromethane, DCM: Ethylacetate(3:1), DCM:EA(9:1). Also, samples (84 beef and 205 pork samples) which were positive by EEC-4 plate test from 2001 to 2003 were tested to investigate the kinds of sulfonamides using HPLC. The results from the study were as follows; 1. The recovery rate of the materials was measured by MSPD method with 3 different extraction solvents; Dichloromethane, DCM:Ethylacetate(3:1), DCM:EA(9:1). The method of extraction solvent with DCM:ethyl acetate(9:1) was the most excellent(87.7∼99.3%) in separation and reappearance. 2. In the LC/MS analysis. of sulfonamides, signal to noise ratio was showed relatively high in the positive mode and special ion in the quality analysis was determined via [M+H]$\^$+/ and m/z 156. A spectrum of sulfonamides was showed from all 12 sulfonamides. 3. The samples positive by the EEC-4 plate, a screening test method, were categorized by sulfonamides through Charm II and confirmed the kinds of sulfonamides through HPLC. 1) Among 84 beef samples positive by EEC-4 plate, 20 samples were positive by Charm II and identified as 7 sulfamethazine, 9 sulfadimethoxine, 1 sulfamonomethoxine and 3 unknown status. 2) Among 205 pork samples positive by EEC-4 plate, 42 samples were positive by Charm II and identified as 19 sulfamethazine, 1 sulfadimethoxine, 4 sulfamonomethoxine and 5 unknown status.

Detection of residual antibiotics by TLC and EEC-4 plate method in slaughtered pigs (도축돈에서 TLC와 EEC-4 plate법을 이용한 항생물질 잔류조사)

  • The antibiotic residues of the urine, the liver, the lung, the kidney and the spleen in slaughtered pigs at Kyongbuk province were detected by TLC(505 kit) and EEC-4 plate method. 1. The positive rate of residual sulfamethazine which was detected by 505 kit in the urine (n=200) was 0.0%. 2. The positive rate of residual sulfamethazine which was detected by EEC-4 plate in the urine (n=126), the liver(n=98), the kidney(n=72), the spleen (n=68) and the lung(n=48) were 63%, 49%, 36%, 34% and 24%, respectively. 3. By EEC-4 plate method, the positive detection rates of the urine were 53.0% in BS(pH 6.0), 29.0% in BS(pH 7.2), 11.5% in BS(pH 8.0) and 13.0% in ML(pH 8.0) medium, that of the liver 41.5% in BS(pH 6.0), 22.0% in BS(pH 7.2), 6.5% in BS(pH 8.0) and 5.0%, in ML (pH 8.0) medium, that of the lung 21.0% in BS(pH 6.0), 9.5%, in BS(pH 7.2) and 8.5% in BS(pH 8.0) medium, and that of the kidney 31.5% in BS(pH 6.0), 14.5% in BS(pH 7.2), 20.0% in BS(pH 8.0) and 3.0% in ML(pH 8.0) medium. In the spleen, only in BS(pH 6.0) medium the positive rate was detected as 33.5 %. 4. The positive rates of samples which shown TLC-positive detected by EEC-4 plate method were 53.9% in no band, 77.8% in one band, 80.9% in two bands, 66.7% in three bands, respectively. In conclusion, the EEC-4 plate method could be applied for the detection of residual antibiotics in samples which shown as out of standard Rf values by TLC-method (SOS kit).

Characteristic Occurrence and Distributions of Pharmaceuticals in the Nakdong River (낙동강 수계 내 의약물질 발생 및 분포 특성)

  • In this study, the occurrence and temporal variation of eight pharmaceuticals comprising antibiotics (clarithromycin, sulfathiazole, sulfamethazine, sulfamethoxazole, trimethoprim), scabicide (carbamazepine) and nonsteroidal anti-inflammatory drugs (acetylsalicylic acid, naproxen) in main stream and its tributary of the Nakdong River basin, were investigated. Concentrations of the target compounds ranged from $1.076{\mu}g/L$. The highest average concentration was observed for clarithromycin ($0.0316{\mu}g/L$), followed by sulfamethazine ($0.0170{\mu}g/L$), sulfamethoxazole ($0.0161{\mu}g/L$), naproxen ($0.0129{\mu}g/L$), carbamazepine ($0.0093{\mu}g/L$), acetylsalicylic acid ($0.0047{\mu}g/L$), sulfathiazole ($0.0024{\mu}g/L$) and trimethoprim ($0.0022{\mu}g/L$). The decreasing pattern of pharmaceutical concentrations was observed along with Nakdong River and the higher concentrations in downstream were observed than those in upstream. There was no temporal variation of the target compounds although the highest level was found in February. The calculated hazard quotients (HQs) for eight pharmaceuticals were below 1, indicating no environmental hazard in Nakdong River. However, further monitoring is still needed due to the other pharmaceuticals widely used in Korea.

A study on simultaneous determination of residual sulfonamides in livestock productions by high performance liquid chromatography (HPLC를 이용한 축산식품중 잔류 설폰아미드제의 동시분석법 연구)

  • This study was carried out to explore the most sensitive and useful method for simultaneous determination of five sulfa drugs(sulfamethazine, sulfamerazine, sulfamonomethoxine, sulfadimethoxine, sulfaquinoxaline) in livestock productions(pork muscle, bovine muscle, chicken muscle, milk ) by HPLC with UV detector and reverse phase column. The results obtained were as follows:1. For mobile phase acetonitrile-0.01M ammonium acetate (23:77) showed more applicable sensitivity and retention times than acetonitrile-1% acetic acid(23:77). Thus acetonitrile-0.01M ammonium acetate(23:77) selected and applied to the modification test, from which it was found pH 6.75 was the most adequate. 2. Optimal wavelength of UV for SMT(sulfamethazine), SMR(sulfamerazine), SMM(sulfamonomethoxine), SD(sulfadimethoxine), and SQ(sulfaquinoxaline) were 266, 266, 265, 269 and 250nm, respectively, and that for simultaneous application it was 263nm. 3. The average recovery rate by extractant(chloroform, dichloromethane, chlorform+dich-loromethane) in the classic method was not significantly different(p>0.05) but that by chloroform higher than the others. Thus chloroform was found to be adequate as extractant in this classic method. 4. The average recovery rate was 86.5% by the MSPD(matrix solid phase disperse) method, which was significantly higher than that by the classic method(p<0.05). Also the recovery rates by method were significantly different(p<0.05) in accordance with sample and type of drug. The MSPD method was much superior to classic method on clean-up.

Studies on the detection of sulfonamide residues in swine edible tissues (돈육내 sulfonamides의 잔류물질 검출에 관한 연구)

  • The study was carried out to determine the residues of sulfonamides in swine edible tissues with high performance thin layer chromatography. For this purpose, the Rf values of sulfonamides in various solvent systems and the recovery rate of sulfameathazine from sampike saples were obtained. Thirty-four samples collected from meat market in Seoul were analyzed. The results obtained from the present study were followings: 1. The average recovery rate of sulfamathazine residues from spiked tissues 0.05, 0.1, 0.5 and 1mg/kg sample weight was 85%. 2. Two of 34 samples of pork for domestic consumption were reported to have been exceeded 0.05 ppm in sulfamethazine residues degree. 3. On the basis of the results, the degree of residues of sulfamethazine in swine meat for domestic consumption is seemed not to be dangerous for public health.

Survey of residual antibiotics in muscle of slaughtered cattle and pig in Gyeongnam Province (경남지역에서 도축우 및 돼지의 근육내 잔류항균물질 검색)

  • The present studies were carried out to determine antibiotics residues in pork and beef muscles by EEC-4-plate and HPLC. A total of 2,534 samples of pork muscles and 1,070 samples of beef muscles from slaughter houses were collected in Gyeongnam area from January to December, 2001. The results were summarized as follows; 1. Recovery rates of TCs, Sulfa drug, Penicillin G from fortified pork and beef muscles ranged as 68.79~98.24%, 78.21~94.58% and penicillin G 63.35~67.24% respectively, by HPLC. 2. Antibiotics residues were detected in 36 sample(1.42%) of pork muscles, 29 sample (2.71%) of beef muscles by EEC-4-plate. 3. Detection rate of antibiotic residues 14 samples(0.55%) and 26 samples(2.43%), in pork and beef muscles, respectively by HPLC. Concentration of residues in 22 sample(2.06%) of beef muscle were higher than tolerance level in korea. 4. Antibiotics detected were sulfamethazine(47.37%), tetracycline(15.79%), oxytetracycline (15.79%), penicillin G(15.79%), sulfamerazine(5.26%) in pork muscle samples and oxyteracycline (37.21%), penicillin G(30.23%), sulfamethazine(20.93%), tetracycline(4.65%), sulfamerazine (2.33%), sulfadimethoxine(2.33%), sulfaquinoxine(2.33%) in beef muscle samples.

Antibiotic Uptake by Plants from Soil Applied with Antibiotic-Treated Animal Manure (가축분뇨 시용 토양에서 식물체의 축산용 항생물질 흡수양상)

  • Food contamination through antibiotic uptake by plants has been one of the major concerns regarding animal manure amendment to agricultural land. Antibiotic uptake by plants was tested with three veterinary antibiotics such as chlortetracycline (CTC), tylosin (TYL), and sulfamethazine (SMT) and three plants such as lettuce, tomato, and hairy vetch. Antibiotic-treated swine slurry was applied to a greenhouse soil before transplanting or sowing the plants. The treated antibiotic concentrations to the swine slurry were 22.9 mg CTC $L^{-1}$, 27.8 mg TYL $L^{-1}$, and 32.4 mg SMT $L^{-1}$. Lettuce was harvested on 64 days after transplanting and 94 days for tomato and hairy vetch. Concentrations of CTC were 3.4 ng $g^{-1}$ for lettuce and 0.7 ng $g^{-1}$ for tomato on a fresh weight basis. For TYL, 20.1 ng $g^{-1}$ from lettuce leaves and 3.0 ng $g^{-1}$ from whole hairy vetch were detected. Sulfamethazine showed greatest concentration among the three antibiotics, 63.3 ng $g^{-1}$ in lettuce leaves, 30.2 ng $g^{-1}$ in tomato fruits, and 20.9 ng $g^{-1}$ in hairy vetch. The results imply that antibiotic uptake by plants may be dependent on antibiotic type and plant type.

  • Sensitive and specific monoclonal antibody (MAb) was produced from hybridoma (1H11-5) obtained by fusion of myeloma cell (V653) and spleen cell isolated from mouse immunized sulfamthazine (SMZ)-HG-KLH. Direct competitive ELISA was developed for rapid detection of SMZ in milk samples using MAb against SMZ with optimized conditions between MAb and SMZ-HG-HRP conjugate, and applicable conditions for analysis of milk samples were established. Detection range of immunoassay was 0.1 to 100 ppb. Recoveries from spiked raw milk and processed milk samples averaged 82.1-120.7 and 82.1-97.1%, respectively.
