Antioxidant Activities of Hot Water and Ethanol Extracts from the stem of Sorbus commixta Heal (마가목 줄기 열수 및 에탄올 추출물의 항산화 활성)

  • Objectives : In order to investigate the possibility of Sorbus commixta Heal. stem (SC) as a natural material, antioxidant activities of the hot water and ethanol extracts were examined. Methods : The samples of SC were pulverized, and fractions were extracted repeatedly three times from hot water and 70% ethanol at room temperature for 2 hours. The antioxidant activities were analyzed from total polyphenol, flavonoid contents, DPPH, ABTS, hydroxyl radical, $Fe2^+$ chelating, and nitrite scavenging activity. Results : Total polyphenol contents were significantly higher in ethanol extract group ($504.39{\mu}g/m{\ell}$) than in hot water extract group ($364.64{\mu}g/m{\ell}$). Total flavonoid contents were also significantly higher in ethanol extract group ($160.09{\mu}g/m{\ell}$) than in hot water extract group ($124.59{\mu}g/m{\ell}$). DPPH, ABTS, $Fe2^+$ chelating were slightly higher in ethanol extract gorups than in hot water extract groups, and increased in a dose-dependent manner. The hydroxyl radical scavenging activity (18.42~23.61%) of ethanol extract groups were shown to be approximately twice higher than that (7.63~10.37%) of hot water extract groups at $12.5{\sim}50{\mu}g/m{\ell}$ concentration. Nitrite scavenging activities of both ethanol and hot-water extract groups were shown to be higher in a dose dependent manner at the concentration of $12.5{\sim}50{\mu}g/m{\ell}$ at pH 3.0 than at pH 1.2, and ethanol extract groups (86.55~96.64%) had higher activity than the hot water extract groups (42.59~92.63%), which was higher than that of the control group antioxidant BHT (72.96~80.11%). Conclusions : The extracts of SC displayed antioxidant activities which suggested a natural material can be developed to functional material.

  • This study investigated the antioxidative and antimicrobial activities of nutmeg (water, ethanol extract and essential oil). The total phenol contents of water, ethanol extract and essential oil were 3.4%, 16.9%, and 3.2%, respectively. Hydrogen donating abilities of water, ethanol extract and essential oil at 1,000 ppm were 4.9%, 41.8% and 6.8%, respectively. The antioxidative activities in linoleic acid substrates were in the order of BHT > ethanol > extract > ${\alpha}$-tocopherol > essential oil > water extract. The antioxidative activities in linoleic acid emulsion substrates were in the order of BHT > water extract > essential oil > ethanol extract > ${\alpha}$-tocopherol. In antimicrobial activity, ethanol extract showed growth inhibition effect against Micrococcus luteus, Bacillus cereus and Salmonella enteritidis, and the essential oil showed growth inhibition effect against Micrococcus luteus. However, no antimicrobial activity of water extract was observed. The nitrite-scavenging abilities of water, ethanol extract and essential oil at 1,000 ppm were 4.5%, 28.8% and 98.8%, respectively, and the ACE inhibitory activities were 0.2%, 11.0% and 10.0%, respectively.

Preparation and Evaluation of Dry Alcohol Containing Red Ginseng Extract (홍삼 엑기스를 함유한 분말주의 제조 및 평가)

  • To develop a dry alcohol containing red ginseng extract, dry alcohols composed of ethanol, water, dextrin and sodium lauryl sulfate were prepared using spray dryer, and their ethanol contents and encapsulation efficiencies were determined. An optimal dry alcohol containing red ginseng extract was chosen and the feeling for its oral administration was evaluated. Dextrin at dextrin/water weight ratios below 1.6/l and ethanol at ethanol/water weight ratios below 1/1 remarkably Increased both the ethanol contents and encapsulation efficiencies of dry alcohols. However dextrin at dextrin/water weight ratios above 1.6/1 and ethanol at ethanol/water weight ratios above 1/1 slightly decreased the both parameters. It might be due to the low solubility of dextrin in ethanol and limited diffusion coefficient of ethanol to the dextrin shell. furthermore, 0.5% (w/w) sodium lauryl sulfate gave the maximum ethanol content of dry alcohol. The more increased amounts of red ginseng extract were added, the more increased amounts of ginsenoside Rb1 but the more decreased amounts of ethanol were encapsulated in dry alcohols. A dry alcohol containing red ginseng extract was prepared with dextrin/ethanol/water (1/1/1, w/w/w) mixed solution, in which 0.5% (w/w) sodium lauryl sulfate and 20% (w/w) red ginseng extract were dissolved. It contained the ethanol contents of31.17$\pm$ 1.33% (w/w) and ginsenoside Rbl of 243.0$\pm$7.0 $\mu$g/g. It gave the moderate taste of red ginseng extract at Its oral administration with or without water Thus, the dry alcohol containing red ginseng extract can be further developed as a more convenient dosage form for red ginseng extract.

Oenanthe javanica extract accelerates ethanol metabolism in ethanol-treated animals

  • The effect of water dropwort (Oenanthe javanica DC) extract in eliminating ethanol was evaluated in New Zealand white rabbit and ICR mice. When a hot-water extract of water dropwort extract and ethanol was injected into New Zealand white rabbit, the plasma ethanol level was rapidly reduced, similar to metadoxine treatment. Specifically, the n-butanol fraction of hot-water extract was the strongest in eliminating plasma alcohol in ICR mice. When ethanol was orally ingested, administration of the hot-water extract eliminated up to 44% of the plasma ethanol in mice while the n-butanol fraction eliminated around 70%. Alcohol removal behaved in a dose-dependent manner in response to 50-200 mg/kg of n-butanol fraction. These data show O. javanica extract is effective in overcoming alcohol intoxication by the accelerating ethanol metabolism.

Antioxidant and Anti-Inflammatory Activities of Extracts from Eugenia caryophyllata Thunb. (정향 열수 및 에탄올 추출물의 항산화 및 항염 효과)

  • The objective of this study was to investigate the antioxidant and anti-inflammatory activities of Eugenia caryophyllata Thunb. water and 70% ethanol extracts. The content of total polyphenol was significantly higher in water extract than in 70% ethanol extract. The DPPH radical scavenging activity of water extract was similar to that of Vit. C at a concentration of $1,000{\mu}g/mL$. The ABTS radical scavenging activities of water and 70% ethanol extract were similar to that of Vit. C at a concentration of $1,000{\mu}g/mL$. SOD-like activity of water extract was higher than that of 70% ethanol extract at a concentration of $1,000{\mu}g/mL$ but lower than that of Vit. C. The DPPH radical scavenging activity, ABTS radical scavenging activity, and SOD-like activity increased as concentrations of water and 70% ethanol extracts increased. Cell cytotoxicity was not observed at all concentrations except at $100{\mu}g/mL$ concentration of water extract. Inhibitory activity on NO production effect of water extract was significantly higher than that of 70% ethanol extract. These results show that E. caryophyllata Thunb. has potent biological activities, and their activities were different depending on extraction solvent.

Effect of Extraction Solvent on the Antioxidant Activity of Lentinula edodes GNA01 Extract (추출용매에 따른 이슬송이버섯(Lentinula edodes GNA01) 추출물의 항산화 활성)

  • Antioxidant activities of 80% methanol, water, and 70% ethanol extracts of Lentinula edodes GNA01 were compared and estimated. The yield of Lentinula edodes GNA01 was identified to be in the following order: water>70% ethanol>80% methanol, but there was no significant difference between 80% methanol and 70% ethanol extracts. The highest total phenolic content (TPC) and total flavonoid content (TFC) were found in water extract, and TPC of 80% methanol extract was higher than that of 70% ethanol extract and TFC of 70% ethanol extract was higher than that of 80% methanol extract. Water extract exhibited the strongest DPPH, ABTS radicals, and nitrite scavenging activities, $Fe^{2+}$ chelating ability, and FRAP among the three extracts. In addition, antioxidant activity of 80% methanol extract was higher than that of 70% ethanol extract in most of the experiments. As a result, antioxidant activity of Lentinula edodes GNA01 showed a difference according to extraction solvent and concentration; nevertheless, water extract exhibiting high polarity had the strongest antioxidant effect. Consequently, water extract from Lentinula edodes GNA01 is anticipated to be useful for the development of a high value-added functional product.

Biological Activities of Extracts from Leaf of Angelica gigas Nakai (참당귀(Angelica gigas Nakai)잎 용매추출에 따른 생리활성)

  • Antioxidant properties and antioxidant activities were analyzed for water extracts and 50% and 70% ethanol extracts of the leaf of Angelica gigas Nakai. The polyphenol and flavonoid contents in water, 50%, and 70% ethanol extract of the leaf of Angelica gigas Nakai, it was found that the polyphenol contents were 18.75 mg GAE/g, 28.95 mg GAE/g, and 34.73 mg GAE/g, respectively, and flavonoid contents were respectively. The DPPH IC50 scavenging activity was 45.84 mg/mL, 36.44 mg/mL, 19.11 mg/mL, respectively, and theABTS+ radical scavenging ability (1 mg/mL) was 28.73%, 22.79%, and 12.70%, respectively. Tyrosinase inhibitory activity, 70% ethanol extract, 50% ethanol extract, and water extract 33.14%, and 4.53%, respectively. Nitrite scavenging activity, 70% ethanol extract, 50% ethanol extract, and water extract were in the order of 1 mg/mL scavenging activity, 36.43%, 34.80%, and 18.85%, respectively.

Antioxidant Activities of Ulmi cortex Extracts According to Ethanol Contents (에탄올 함량변화에 따른 유백피 추출물의 항산화 활성)

  • Objectives : This study was performed to find best extraction solvent for application of Ulmi cortex to food or herbal medicine as an antioxidant only using water, ethanol and their mixtures. Methods : The Ulmi cortex extracts were prepared using water and 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90% and 100% (v/v) ethanol, and were evaluated yields, total polyphenol contents, DPPH and ABTS radical scavenging activities, lipid peroxidation activities, and catechin and epicatechin contents. Results : Among the Ulmi cortex extracts, the yield was highest in water extract (8.9%) and lowest in ethanol extract (3.8%). The yield of 30% ethanol extract (8.5%) also was very high to similar with water extract. The total polyphenol content was highest in the 30% ethanol extract ($253.6{\mu}g/mg$ extract) and lowest in water extract ($109.0{\mu}g/mg$ extract). The DPPH radical scavenging activity was highest in ethanol extract (IC50, $8.53{\mu}g/ml$), ABTS radical scavenging activity was highest in 60% ethanol extract (IC50, $3.08{\mu}g/ml$), and the inhibition of lipid peroxidation was highest in 70% ethanol extract (IC50, $7.96{\mu}g/ml$). As ethanol content of extraction solvent increased from 0% to 30%, the antioxidant activities were remarkably increased whereas from 30% to 100%, the antioxidant activities were increased or decreased a little. Conclusions : The findings of the present study suggest that 30% ethanol is best solvent for extraction of Ulmi cortex, considering yield, polyphenol content, and antioxidant activities with extraction cost.

The Effects of Houttuyniae Herba extract on the Activity of Anti-bacteria, Anti-inflammation and Anti-oxidation (어성초(魚腥草) 추출물의 항여드름 효과에 관한 연구)

  • Objective : This experimental study was performed to investigate the effects of Houttuyniae Herba extract on anti-inflammation and anti-oxidation. Methods : The cytotoxicity of Houttuyniae Herba water extract and ethanol extract about viability of Raw 264.7 cell were tested using a colormetric tetrazolium assay(MTT assay). We investigated the inhibitory effects of Houttuyniae Herba water extract and ethanol extract on Propionibactrium acnes using paper disk diffusion method. To investigate the anti-inflammation effects of Houttuyniae Herba water extract and ethanol extract on LPS-induced macrophage Raw 264.7 cell, we used ELISA kit. We evaluated anti-oxidation effects of Houttuyniae Herba water extract and ethanol extract on HaCaT cell by Enzyme recycling method. Results : 1. In Houttuyniae Herba water extract and ethanol extract, cell toxicity depended on the density and wasn't difference between two extracts. 2. Houttuyniae Herba water extract and ethanol extract has not the significant inhibition effect of Propionibactrium acnes. 3. Concentration of 50, $100{\mu}g/m{\ell}$ Houttuyniae Herba water extract inhibited the production of NO in the Raw 264.7 cell stimulated with LPS. 4. All extracts except for $20{\mu}g/m{\ell}$ Houttuyniae Herba water extract showed anti-oxidation effect by decreasing the DPPH radicals. Conclusion : These results indicate that Houttuyniae Herba extract has anti-inflammation and anti-oxidation effects. If further study is performed, the use of Houttuyniae Herba extract will be valuable and benificial in the therapy of Propionibactrium acnes.

Comparison on the Antioxidative Activity of Ethanol and Hot Water Extracts of Euphorbia supina Rafinesque (애기땅빈대의 열수 추출 및 에탄올 추출물의 항산화 활성 비교)

  • Objectives : In this study, the antioxidant activities of the 80% ethanol and hot water extracts of Euphorbia supina Rafinesque were investigated. Methods : We measured total phenol contents, flavonoid contents, 2,2-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging assay. The production of reactive oxygen species was measured in LPS-induced RAW 264.7 cells using flow cytometry system. Results : The content of phenol in the hot water extract was $65.529{\pm}0.462mg/g$ and $126.932{\pm}1.894mg/g$ in the 80% ethanol extract, and that of flavonoid in the hot water extract was $16.063{\pm}0.471mg/g$ and $29.159{\pm}1.963mg/g$ in the ethanol extract. The 80% ethanol extract also showed higher DPPH and ABTS radical scavenging activities ($90.8{\pm}1.0%$ and $92.5{\pm}0.7%$) than the hot water extract ($81.5{\pm}0.5%$ and $91.5{\pm}0.2%$). The production of reactive oxygen species(ROS) was reduced dose-dependently by 80% ethanol and hot water extract at concentration of 1, 10 and $100{\mu}g/m{\ell}$ of RAW 264.7 cells. Conclusion : According to these results, the 80% ethanol extract of Euphorbia supina Rafinesque has a good anti-oxidative effects than the hot water extract. Thus, the 80% ethanol extract of Euphorbia supina Rafinesque may serve as useful natural antioxidants.
