Construction of ELISA System for the Detection of Indian citrus ringspot virus (Indian citrus ringspot virus의 ELISA 진단 시스템 구축)

  • Indian citrus ring spot virus (ICRSV) is known to cause a serious disease to citrus, especially to Kinnow mandarin, the popular cultivated citrus species in India. In this study, we developed diagnostic systems based on enzyme-linked immunosorbent assay (ELISA). In order to generate antibodies against ICRSV coat protein, we overexpressed the coat protein in Escherichia coli using the pET15b expression vector containing an optimized ICRSV coat protein gene. The recombinant ICRSV coat protein was overexpressed as soluble form at $37^{\circ}C$ upon IPTG induction. The protein was purified to 95% in purity by Ni-NTA column chromatography. The purified protein was immunized to rabbit for the generation of polyclonal antibody (PAb). The PAb showed a specific immunoreaction to recombinant ICRSV coat protein in western blot analysis and ELISA. Diluted rabbit antisera (10,000 fold) could detect less than 10 ng and 5 ng of the target protein in western blot and ELISA analysis, respectively.

  • A mutant strain which hyperproduced thermostable ${\alpha}-amylase$ was obtained by chemical mutagenesis of Bacillus licheniformis. The mutant strain, SK-5, produced the enzyme about 50 times higher than the original strain. The mutant was longer and slimmer in shape, slower in growth compared to the original strain. Nucleotide sequence analysis of the SK-5 ${\alpha}-amylase$ gene revealed no changes in the the structural gene. The changes found in the promoter region might be responsible for the hyperproduction of the enzyme by the mutant. No structural changes in the enzyme structure could be observed when the secreted enzymes at various culture times were analyzed by Western blot.

EA-D p45-IgG as a Potential Biomarker for Nasopharyngeal Carcinoma Diagnosis

  • Aim: To identify new biomarkers for NPC diagnosis with an anti-EBV Western blot test kit. Methods: Serum samples from 64 NPC patients and healthy subjects with four specific VCA-IgA/EA-IgA profiles were tested with an anti-EBV Western blot test kit from EUROIMMUN AG. Proteins were quantified with scores of intensity visually assigned to the protein bands. The markers which showed statistical differences between the NPC and non-NPC subjects were further evaluated in another 32 NPC patients and 32 controls in comparison with established biomarkers including VCA-IgA, EA-IgA, EBV-related protein IgG, and EBV DNA. Results: Among the markers screened, EA-D p45-IgG showed a statistically significant difference (p < 0.05) between NPC and non-NPC subjects with VCA-IgA positivy. In 32 VCA-IgA positive NPC patients and 32 control subjects, the diagnostic accuracy of EA-D p45-IgG was 78.1% with a positive predictive value of 77.8% and a negative predictive value of 78.6%. In the verification experiment, the specificity and sensitivity of EA-D p45-IgG were 75.0% and 90.6 %, respectively. Conclusions: EA-D p45-IgG might be a potential biomarker for NPC diagnosis, especially among VCA-IgA positive subjects.

Analysis of outer mombrane proteins of Brucella abortus using two dimensional polyacrylamide gel electrophoresis (2차원 전기영동법을 이용한 Brucella abortus 세포외막 특이단백질의 분석)

  • Outer membrane proteins(OMPs) of Brucella abortus 1119-3 strain were extracted by Triton X-100 treatment, and fractionated by DEAE-cellulose column chromatography and Sephacryl S-300 column chromatography. The antigenic proteins in these fractions were identified by Western blot analysis. In Western blot analysis, a single band(38kDa) was observed in the DEAE fractions from 36th fraction to 38th fraction against sera of cattle infected with B abortus. And other fractions have several bands. However, the Sephacryl S-300 fractions exhibited a total of 3 peaks of proteins with a broad range from about 30 to 116kDa. In order to characterize further, the extracted OMPs and the DEAE fractions were analyzed by two dimensional polyacrylamide gel electrophoresis(2-DE) and Western blot using serum from naturally infected cattle with Brucella spp. The 2-DE immunoblots of DEAE fraction showed immunoreactive spots more than twenty two. The major protein spots have ranging from about 32 to 47kDa. The pI values of the spots were detected from pH 4.7 to 5.4. Among the major protein spots, the 38kDa protein which is a specific antigen, located at the point of approximately a pI 4.8.

Expression of Survivin and HIAP-1 in Korean Gastric Cancers (한국인 위암에서 Survivin과 HIAP-1 유전자 발현)

  • Purpose: Dysregulation of apoptosis may attribute to development of cancer by abnormally prolonging cell viability with accumulation of transforming mutations. Survivin and HIAP (Human Inhibitors of Apoptosis)-1 were recently described as apoptosis inhibitors. Their pathogenic roles in gastric cancer are largely unknown. In the present study, we examined the expression of survivin and HIAP-1 in gastric cancer tissues and cell lines in order to elucidate the roles of survivin and HIAP-1 in the process of gastric carcinogenesis. Materials and Methods: Eight gastric cancer cell lines and five gastric cancer tissues were studied. The expression of survivin and HIAP-1 were evaluated by reverse transcription -polymerase chain reaction (RT-PCR), immunohistochemistry, and Western blot. Results: Western blot and RT-PCR analysis revealed survivin and HIAP-1 expression in all gastric cancer cell lines. Increased expression of survivin and HIAP-1 were found in all cases of gastric cancer tissues compared to normal tissues by Western blot analysis. In immunohistochemical analysis tumor cells were stained with anti-survivin and anti-HIAP-1 antibodies. Cell cycle dependence of survivin expression was preserved in gastric cancer cell lines. Conclusion: The results indicate that increased expression of survivin and HIAP-1 genes may play an important role in gastric cancer.

Expression of Antibody Genes Specific for Human Hepatitis-B Virus in Transgenic Tabacco Plants (형질전환된 담배에서 사람 B형 간염바이러스 항체 유전자의 발현)

  • Chimeric kappa chain and gamma chain cDNA clones (pCKS2 and pCHS2) of a monoclonal antibody specific for pre-S2 surface antigen of human hepatitis-B virus were ligated into Xbal site of plant expression vector pBKS-1. Plasmid DNA containing each of the chimeric gene were then mobilized from E, coli to Agrobacterium tumefaciens strain LBA4404. The chimeric antibody genes were then introduced into tobacco by Ti plasmid-mediated transformation. The putative Transformants were selected on medium containing kamaycin sulfate. Shoots that formed on shoot induction medium were analyzed by Western blot analysis for the expression of kappa-chain or gamma-chain genes. The Western blot analyses clearly showed that the introduced genes were stably expressed in transgenic plants.

Serological cross-reaction with Brucella abortus antigen extracted by sodium dodecyl sulfate and Yersinia enterocolitica 0:9 (SDS 처리한 브루셀라 항원과 Yersinia enterocolitica 0:9주의 혈청학적 교차반응 연구)

  • Brucella abortus cell wall antigen was extracted from Brucella abortus 1119-3 by ultrasonication and followed by sodium dodecyl sulfate(SDS) treatment. In order to confirm whether this preparation is serologically cross reactive with Yersinia enterocolitica 0:9, Western blot analysis with mouse anit-Brucella abortus1119-3 and with mouse anti-Yersinia enterocolitica 0:9 was performed. ELISA results from using those Brucella antigen and Yersinia antigen were assessed whether they had correlation. According to the results of western blot analysis and ELISA, there was no evidence of cross reactivity between the Brucella abortus 1119-3 antigen preparation and Yersinia enterocolitica 0:9. Therefore the SDS treated antigen prepared in this study could be suitably used as specific ELISA antigen without confusion in the interpretation of serological tests for brucellosis in cattle.

Cloning and Expression of Lactadherin Gene from Korean Women (한국 여성의 Lactadherin 유전자 Cloning과 발현 연구)

  • Lactadherin is a glycoprotein of human milk fat globule membrane that binds to mucin and butyrophilin forming the protein complex. Especially, mucin and lactadherin in human milk efficiently protect infants with poor immune functions right after birth from infections by microorganisms and play important roles for their early survival, growth and development. Lactadherin inhibits the propagation and growth of rotavirus that is a global pathogen causing infants' diarrhea. Recently this protein was known to promote neovascularization and its deficiency related to develop Alzheimer's disease. In this study, the basic biochemical and physiological aspects of lactadherin were investigated. Messenger RNAs were isolated from mammary tissues from Korean women patients to clone a 1.2 kb cDNA and sequenced its DNA to determine its amino acid sequences. The cDNA was cloned to express its 43 kD protein in E. coli, which was confirmed by Western blot. The recombinant protein was purified and injected to 2 rabbits to raise antibodies against it. The semi-purified milk fat globule membrane proteins from Korean women was analyzed by Western blot using the rabbit antibody to give 70, 55, 46, 30 kD bands. Also several polymorphism and SNPs of lactadherin gene from Korean women were observed compared with those of Caucasian women.

Production and Characterization of Monoclonal Antibodies against the 90-kDa Heat Shock Protein (HSP90에 대한 단일클론항체의 생성 및 특성 조사)

  • The 90 kDa-heat shock protein (HSP90) is one of the major ubiquitous heat shock proteins induced by a vadety of ceilular stresses. HSP90 Is constitutively synthesized even under nonstressed condidons and found In association with several regulatory and structural proteins such as protein kinases and steroid hormone receptors. In the present study, to facilitate its biochemical characterization, HSP90 was pudfied from chick muscle by sequential column chromatography steps including DEAE- cellulose, hydroxyapatite, and Sephacryl S-300 gel filtration and monoclonal antillodies specific to HSP90 were produced by the inurine hybridomal technique. We report the production of 4 posItive hybridoma clones, named as A204, C112, C302 and A204, C112, C302. Among these MoAbs, Cl 12 strongly reconnized chick HSP90 in Western blot and native immunoprocipitation. In addition, C112 showed the crossreactivitles against HSP90 from human, rabbit, mouse, fish and chick but not from Drosophila and E. coil.

  • Cyclosproin A(CsA)는 세포 이식거부방지를 위한 면역 억제제 및 자가 면역질환 치료제로 널리 사용되어 왔다. CsA는 매양된 사람 치은섬유아세포를 증식시킴이 알려져 있지만 CsA에 의한 세포증식기전에 대한 세포사멸기전 및 Bcl-2의 역할은 연구되어 있지 않다. 이번 연구는 사람 섬유아세포에서 CsA에 의한 세포증삭기전에 세포고사기전 및 Bcl-2 family가 관여하는지 밝히는 데에 목적이 있다. 세포 생장력은 MTT 방법으로 측정하였다. Bcl-2 family와 Fas 발현 정도는 RT-PCR 방법이나 western blot으로 확인하였다. Caspase-3 및 -9의 활성은 ELISER reader로, reactive oxygen species(ROS)는 fluorescence spectrometer에 의해 측정되었다. 미토콘드리아에서 세포질로 분비된 cytochrome c는 Western blot으로 조사하였다. CsA는 $0.1{\sim}10\;{\mu}M$에서 사람 섬유아세포의 생존률을 시간과 농도 의존적으로 증가시켰으며, 50 ${\mu}M$ CsA에서는 오히려 세포가 죽였다. 또한, CsA 처리로 미토콘드리아에서 세포질로 유리되는 cytochrome c 양과 VDAC 1 및 3 발현량이 감소되었고, caspase-9과 caspase-3의 활성도도 감소되었다. 한편, CsA 처리한 섬유아세포에서 death receptor 구성요소인 Fas 발현이 감소되었다. Bcl-2 family에 대한 RT-PCR, western blot 분석결과, 세포고사를 억제하는 Bel-2 발현은 증가되었으나 세포고사를 자극하는 Bax와 Bid의 발현은 감소되었다. 이러한 결과들은 사람 섬유아세포에서 CsA유도 세포증식에 Bcl-2 family와 ROS가 매개하는 미토콘드리아 의존 및 death receptor 의존 세포고사기전이 관여함을 시사하였다.
