A Study on the Effects of Bee Venom Aqua-Acupuncture on Writhing Reflex (봉독약침(蜂毒藥鍼)이 Writhing reflex 및 척수내(脊髓內) 통증관련(痛症關聯) 신경세포(神經細胞)의 활성(活性)에 미치는 영향(影響))

  • Introduction : In spite of the use of Bee Venom aqua-acupuncture in the clinics, the scientific evaluation on effects is not enough. Bee Venom aqua-acupuncture is used according to the stimulation of acupuncture point and the chemical effects of Bee Venom. The aims of this study is to investigate the analgegic effects of the Bee Venom aqua-acupuncture, through the change of writhing reflex Materials and Methods : Pain animal model was used acetic acid method. The changes of writhing reflex of the mice which were derived pain by injecting acetic acid into the abdomen, after stimulating Bee Venom aqua-acupuncture on Chungwan($CV_{12}$) and non acupuncture point on the backside were measured. Results : 1. It showed that the writhing reflex were appeared on the groups which injected aceticacid only, and saline-acetic acid group(sample I), but not on the group bee venom-saline group(sample II). 2. The change of writhing reflex by Chungwan($CV_{12}$) Bee Venom aqua-acupuncture showed significant decrease in the order of Chungwan($CV_{12}$) Bee Venom aqua-acupuncture group III($2.5{\times}10^{-3}$g/kg), II($2.5{\times}10^{-4}$g/kg), and I($2.5{\times}10^{-5}$g/kg), compared with control group. There were significant decrease of number of writhing reflex in $5{\sim}10,\;10{\sim}15\;and\;15{\sim}20$ minutes intervals of Chung wan($CV_{12}$) Bee Venom aqua-acupuncture group I, and in $0{\sim}5,\;5{\sim}10,\;10{\sim}15\;and\;15{\sim}20$ minutes intervals of II and III, compared with control group. 3. The change of writhing reflex by non acupuncture point Bee Venom aqua-acupuncture showed significant decrease in the $0{\sim}5\;and\;5{\sim}10$ minutes intervals and the total number of writhing reflex in $2.5{\times}10^{-4}$g/kg group, compared with control group 4. The effects of writhing reflex of Chungwan($CV_{12}$) Bee Venom aqua-acupuncture group showed significant decrease, compared with non acupuncture point Bee Venom aqua-acupuncture group. Conclusion : This study shows that the Bee Venom aqua-acupuncture on Chungwan($CV_{12}$) decreases the numbers of writhing reflex. As the analgegic effects of Bee Venom aqua-acupuncture is recognized. Bee Venom aqua-acupuncture treatment is expected for pain modulation. In order to use it in many ways, more researches are needed for the dose and stability of Bee Venom aqua-acupuncture.

  • Ethanolic extract of Aphanamixis polystachya bark was used to investigate its analgesic activity by acetic acid induced writhing in mice. The bark extract exhibited statistically significant and dose dependent analgesic activity in mice. The bark extract at the doses of 250 and 500 mg/kg body weight showed 40.69% and 62.07% writhing inhibition respectively in mice whereas diclofenac-Na produced 75.17% writhing inhibition as a positive control.

  • The intentional test of the clinical test on the mouse, which are orally administered salicylamide combined with chlorpheniramine maleate were made by writhing syndrome method of 0.7% acetic acid-saline solution and following effects were found. 1) The cross-over test of writhing syndrome method have intention in case of three days rest after the first examination. 2) The most active rate of salicylamide by administration combined with chlorpheniramine maleate is salicylamide 20mg/kg: chlorpheniramine maleate 20mg/kg (1:1) 3) The most active range of chlorpheniramine maleate used for assistant action of salicylamide is $15mg/kg{\sim}20mg/kg$

  • The current experiment was carried out to investigate the analgesic effects of several herbal drugs in acetic acid - induced pain model. In a single drug group : after administration of herbal drugs(1g/kg or 3g/kg) orally for 30 minutes, 1% acetic acid $(250{\mu}l)$ was administered into abdominal cavity of mouse. And then the number of times of writhing response was measured for 30 minutes. In a combination drug group : after administration of herbal drugs (1g/kg and it's compound 2g/kg) orally for 30 minutes, 1% acetic acid $(250{\mu}l)$ was administered into abdominal cavity of mouse. And then the number of times of writhing response was measured for 30 minutes. The results were summarized as follows; 1. Water extracts of Akebiae caulis(木通) and Stephaniae tetrandrae radix(防己) decreased significantly the number of writhing response. 2. Methanol extracts of Achyranthis bidentatae radix(牛膝), Carthami flos(紅花), Akebiae caaulis(木通), Stephaniae tetrandrae radix(防己), Myrrha(沒藥), Corydalidis tuber(玄胡索) and Persicae semen(桃仁) decreased significantly the number of writhing response. 3. Water extracts of Achyranthis bidentatae radix(牛膝) plus Akebiae caulis(木通), Achyranthis bidentatae radix(牛膝) plus Stephaniae tetrandrae radix(防己) and Achyranthis bidentatae radix(牛膝) plus Ledebouriellae radix(防風) decreased the number of writhing response significantly. 4. Methanol extracts of Achyranthis bidentatae radix(牛膝) plus Myrrha(沒藥), Achyranthis bidentatae radix(牛膝) plus Stephaniae tetrandrae radlx(防己) and Achyranthis bidentatae radix(牛膝) plus Ledebouriellae radix(防風) decreased the number of writhing response significantly.

Acupuncture on ST36 Increases c-Fos Expression in vlPAG of Visceral Pain-induced Mice

  • Background : Acupuncture has been used as a clinical treatment in Oriental medicine for various diseases including pain relief. The descending pain control system of periaqueductal gray (PAG) is a powerful pain control system in mammalians. Expression of c-Fos is used as a marker for stimuli-induced metabolic changes of neurons. Objective : In the present study, the effects of acupuncture on analgesic effect in visceral pain were investigated through the writhing reflex and c-Fos expression in ventrolateral PAG (vlPAG) area using immunohistochemistry in mice. Method : For the writhing test, mice were divided into five groups. Immediately after finishing the behavioral test, the animals were weighed and overdosed with Zoletil. After a complete lack of response was observed, the brains of the mice were dissected into serial coronal sections, and c-Fos immunohistochemistry was performed. Statistical analysis of all data was performed using one-way ANOVA. Result : The present results showed that acupuncture affected the writhing reflex and that Choksamni (zusnali) acupoint and aspirin significantlysuppressed acetic acid treatment-induced increased writhing reflex, and the expression of c-Fos in vlPAG was significantly increased in the acupunctured group. Conclusion : The present study suggests that acupuncture has an antinociceptive effect on acetic acid-induced visceral pain by increase of c-Fos expression in mice. Aspirin also showed analgesic effect, however the mechanism is different from the acupuncture.

The Analgesic Effect and Mechanisms of Dianthus chinensis L Extract in the mice.

  • In the present study, the antinociceptive profiles of Dianthus chinensis L extract were examined in ICR mice. Dianthus chinensis L extract administered orally (200 mg/kg) showed an antinociceptive effect as measured by the tail-flick and hot-plate tests. In addition, Dianthus chinensis L extract attenuated the writhing numbers in the acetic acid-induced writhing test. Furthermore, the cumulative nociceptive response time for intrathecal (i.t.) injection of substance P ($0.7\;{\mu}g$) was diminished by Dianthus chinensis L extract. Intraperitoneal (i.p.) pretreatment with yohimbine ($\alpha_2$-adrenergic receptor antagonist) attenuated antinociceptive effect induced by Dianthus chinensis L extract in the writhing test. However, naloxone (opioid receptor antagonist) or methysergide (5-HT serotonergic receptor antagonist) did not affect antinociception induced by Dianthus chinensis L extract in the writhing test. Our results suggest that Dianthus chinensis L extract shows an antinociceptive property in various pain models. Furthermore, this antinociceptive effect of Dianthus chinensis L extract may be mediated by $\alpha_2$-adrenergic receptor, but not opioidergic and serotonergic receptors.

Effects of Jakyakgamchotang extract on the Analgesic effect in Mice (작약감초탕(芍藥甘草湯) 및 구성약물(構成藥物)이 진통(鎭痛)에 미치는 영향(影響))

  • This study was designed to elucidate the analgesic effects of Jakyakgamchotang and Radix paeinia, Radix Glycyrrhiziae which are the component of Jakyakgamchotang in mice. The analgesic effects were measured by the Whittle's method and hot plate method. The results were as follows ; After the administration of Jakyakgamchotang extrace, the frequency of writhing syndrome was significantly inhibited. After the administration of Radix paeinia extract, the frequency of writhing syndrome was significantly inhibited. After the administration of Radix Glycyrrhiziae extract, the frequency of writhing syndrome was significantly inhibited. After the administration of Jakyakgamchotang extrace, paw licking time and escape time were significantly prolonged. After the administration of Radix paeinia extract, paw licking time was significantly prolonged. After the administration of Radix Glycyrrhiziae extract, paw licking time was significantly prolonged. It is concluded that the analgesic effects of Jakyakgamchotang were found to be more effective than Radix paeinia and Radix Glycyrrhiziae.

  • 소합향원 (蘇合香元) 가감방(加減方) 정유향기의 진통효과가 투여방법에 따라 어떤 차이가 있는지를 비교, 검토함으로서 본 약재의 새로운 적용방법을 탐색해 보고자 한다. 방법 : 가감소합향원에서 추출한 정유향기를 실험동물을 대상으로 7일간 경구 (50mg/kg, 100mg/kg), 흡입 (매일 3시간씩 1일 2회), 또는 피부도찰 (10mg) 방법으로 투여하여 페닐퀴논-유도 writhing test, 아세트산-유도 writhing test 및 hot-plate test를 실시하였다. 결과 : 가감소합향원 정유향기는 페닐퀴논-유도 writhing test에서는 피부도찰이 가장 효과적 이었으며 그 효과는 양성 대조약물인 아세트아미노펜 보다 약간 더 우수하였다 (p<0.05). 아세트산-유도 writhing test에서도 피부도찰법이 가장 좋은 효과를 나타내었으며 양성대조 약물인 아미노피린보다는 좀 더 좋은 효과를 보여주었다 (p<0.05). 그러나 hot-plate test에서는 경구투여가 가장 좋은 진통효과를 나타내었는데 고용량 (100mg/kg)에서는 양성 대조약물인 아세트아미노펜보다 유의적으로 더 효과적이었으나 저용량 (50mg/kg )에서는 이보다 다소 약하였다. 이상의 연구결과, 가감소합향원의 정유향기는 피부도찰시에는 비특이적 화학자극제인 페닐퀴논이나 아세트산에 의한 통증의 완화에 보다 효과적이며, 경구투여시에는 중추신경계에 영향을 미치는 hot plate에 의한 열자극성 통증에 더 유효함을 의미한다. 결론 : 가감소합향원의 정유향기액은 향기흡입보다는 피부흡수나 경구투여방법으로 진통의 목적으로 응용될 수 있을 것으로 사료된다.

Improvement of Analgesic and Anti-inflammatory Effects by Mixing with Bucillamine and Lornoxicam (Bucillamine과 Lornoxicam의 혼합에 의한 진통소염 효과 향상)

  • The aim of this study was to reduce the side effects and to develop effective drugs using bucillamine (B), lornoxicam (L), and its mixtures on the analgesic and anti-inflammatory effects. With this goal, we compared their effects on the four different mixtures with a sole treatment (B 40 mg/kg and L 1.60 mg/kg). The mixture 1, 2, 3, and 4 ratios of B to L (mg/kg) were 20 to 0.80, 40 to 1.60, 80 to 3.20, and 40 to 1.14, respectively. In terms of acetic acid-induced vascular permeability, B and L inhibited the amount of dye leakage approximately 37.8 and 66.5%, respectively. And mixture 1, 2 and 3 showed inhibition of 47.4%, 81.5%, and 84.3%. The mixture 4 inhibited approximately 49.4%. In carrageenan- induced paw edema model, mixtures of B and L effectively inhibited paw edema measured 1/2~3 hours after carrageenan injection. Especially, mixture 2 inhibited 50.7%, 52.7%, 50.9% of paw edema after 1, 2, and 3 hr, significantly. We also examined an analgesic effect using the writhing test. In terms of the acetic acid-induced writhing syndrome, the control group showed writhing syndrome 18.5 times. B and L showed 9 and 6.3 times, inhibiting 51.6% and 65.9% respectively. And aspirin, as a positive control drug, showed the 7.1 times writhing syndrome. The mixture 1, 2, 3, and 4 also significantly inhibited the writhing syndrome to 62.2%, 93.0%, 51.4%, and 77.8%, respectively. From these results, we could suggest that the range of B and L ratio of 25 : 1 to 35 : 1 may be applicable to developing analgesic and anti-inflammatory drugs.

Antiinflammatory and Analgesic Effects of Higenamine, a Component of Aconiti Tuber

  • The antiinflammatory and analgesic activities of higenamine were evaluated by measuring edema volume and pain threshold in adjuvant arthritic rats and acetic acid-induced writhing test in mice. Higenamine, with consecutive oral administrations at doses of 10 and 50 mg/kg/day, showed significant antiedemic effect and elevation of pain threshold during the secondary lesion of adjuvant arthritis. Higenamine also showed a significant inhibition of acetic acid-induced writhing syndrome with a single oral administration (200 mg/kg). From these results, it is postulated that higenamine might possess both of centrally and peripherally mediated analgesic properties.
