To develop hybridomas secreting monoclonal antibodies to be used as unlimited sources of reagents indispensable for the diagnosis and treatement of leukemic malignancy, a monoclonal antibody was generated to human pre-T leukemia cells (Jurkat). Hybridomas were produced against Jurkat cell line by fusing spleen cells from hyperimmunized mice with murine plasmacytoma cells (P3$\times$63Ag8. V653). One monoclonal antibody derived from this fusion, designated DMJ-2 was reactive with T-cell lines (Jurkat, Molt-4 and RPMI-8402) and normal peripheral E-rosette forming T cells, but unreactive with B-cell lines (Daudi, Nalm-6) and non-T, non-B cell line (K562). Conclusively DMJ-2 reactive with mature and immature T-lineage lymphoid cells.

It is reasonably well known that there is a relationship between myelin formation and methylation of myelin basic protein in nerve cells. One of the suggestions is that arginine methylation of myelin basic protein could be of aid in the conjugation of myelin protein with the nonpolar lipid to form myelin. Abnormality in methylation of myclin basic protein might induce the neurological diseases in experimental animals as well as in human being. In the biological system, the methylation reaction is catalyzed by protein methaylse I using S-adenosyl-L-methionine as methyl donor. In this study, we examined the changes of S-adenosyl-L-methionine concentration and protein methylase I activity in developing rat brain tissues. The results are sumraerized as followings: (1) In brain tissues of fetus rat, the concentration of S-adenosyl-L-methionine was gradually decreased until to birth. However, the concentration in brain tissues of infant rat was suddenly increased at 7th day(just before myelination occur) birth. (2) Protein methylase I activity was decreased until to birth in brain of fetus rat and increased temporally just after birth, However, the enzyme activity showed no changes around 7th day after birth.

The effects of organic acidic drugs on the absorption, excretion and diuretic action of furosemide were studied. Cefalexin, p-aminohippuric acid (PAH), ibuprofen and p-amino salicylic acid (PAS) were selected as organic acidic drugs. The in situ absorption rate and absorption rate constant of furosemide (30$\mu{M}$) were significantly (p<0.05) decreased by 30$\mu{M}$ of cefalexin, PAH, ibuprofen and PAS in rat small intestine. The plasma concentration of furosemide was significantly (p<0.01) increased by cefalexin, PAH and ibuprofen in rabbits. But the urinary excretion rate, renal clearance and diuretic action of furosemide were significantly (p<0.05) decreased by cefalexin, PAH, ibuprofen and PAS in rabbits.

There are three plausible bioconversion pathways in biodegradation mechanism of capsaicinoids; first, side chain degradation through $\omega$-hydroxylation and $\beta$-oxidation, secondly, aromatic ring hydroxylation, and lastly, hydrolysis on the acidaraide linkage. In microbes, it was reported that capsaicin and its synthetic, analog, nonoylvanillylamide(NVA), could be metabolized to N-vanillylcarbamoylbutyric acid via $\omega$-hydroxylation and consecutive $\beta$-oxidations by Aspergillus niger. In order to broaden the scope of microbial degradation of capsaicinoids, over thirty strains of various fungi including Aspergillus, Penicillum, Mycotypha, Gliocladium, Paecilomyces, Byssoclamys, Conidiobolus, Thamnidium, and Entomophthora. It was observed that almost all the strains examined oxidized, the side chain of capsaicids as A. niger did. These observations strongly support the notion that side chain degradation is the most dominant pathway in the microbial degradation of capsaicinoids.

The relationship between in vitro release and in vivo bioavailability of acetaminophen from suppositories was investigated. Effect of glycyrrhizin on the drug release and rectal absorption in rats was also examined. Suppositories containing 25mg of acetaminophen were prepared with Wecobee FS (fatty base) or PEG (water-soluble base) bases. The release from the suppositories were determined with USP rotating basket dissolution apparatus and with the suppository release tester. The temperature of the dissolution medium was very critical for the dissolution of acetaminophen from Wecobee FS suppositories. The bioavailability of acetaminophen was calculated from the plasma concentration-time curve after rectal administration of the suppositories to the rats. There were no significant differences in AUC following rectal administration of Wecobee FS and PEG suppositories, but the release and absorption from the Wecobee FS suppositories were faster than those from PEG suppositories. The dissolution rate obtained by the suppository release tester was better correlated with in vivo absorption rate constant than that by the USP dissolution apparatus. It suggests that the partitioning between rectal fluid and suppository base is the rate-limiting step in the rectal absorption of acetaminophen from suppositories. Glycyrrhizin was found not to affect in vitro dissolution and rectal absorption of acetaminophen.

Polyamines of Korean red ginseng were extracted with 5% trichloroacetic acid and purified by ion exchange chromatography using Dowex-50Wx8 resin. Four spots having R$_f$ values of 0.19, 0.28, 0.35, and 0.45 were detected. It was observed under microscopy that those polyamines stimulated the growth and differentiation of chicken embryonic muscle cell. The development of muscle cells from the stage of myoblast to that of myotube was found to be enhanced by those polyamines. It was also observed that those polyamines most likely lengthened, the life-span of the cultured chicken embryonic skeletal muscle cells.

The increase in the serum cholesterol level of the rabbit fed a high cholesterol diet was prevented by the administration of a Lactobacillus sporogenes preparation, proportionally to the amount of the preparation. About 90% of the increase in the serum cholesterol level was inhibited in the rabbits fed the preparation (2%) as feed additives. In vitro, L. sporogenes cultured in MRS broth containing PPLO serum fraction as a cholesterol source decreased the cholesterol concentration in the spent broth and increased the intracellular cholesterol concentration. The ability of L. sporogenes to accumulate cholesterol was enhanced by the presence of oxgall in the medium. These results suggest that L. sporogenes may directly assimilate the cholesterol in the gastrointestinal tracts and may prevent the increase of the serum cholesterol level.

In cardiovascular disease the flow adaptation of erythrocytes can be affected by reduced shear stresses and metabolic influences on red cell fluidity as a consequence of tissue hypoxia. In addition there are indications that risk factors of cardiovascular diseases are able to decrease the intrinsic red cell deformability. Erythrocyte deformability was studied by the filtration technique of Reid et al. to investigate the relationship between blood pressure chances and erythrocyte deformability. In this experiment normotensive rats, spontaneously and DOCA-salt treated hypertensive rats were used. Erythrocyte deformability was significantly reduced by blood pressure elevation in hypertensive rats but was not fully recovered by normalization of blood pressure after antihypertensive drug treatment. Therefore other factors than blood pressure may be involved in erythrocyte deformability reduction during blood pressure elevation.

A high performance liquid chromatographic method was developed for the determination of glycyrrhetinic acid contained in licorice powder. Glycyrrhetinic acid which is hydrolysate of glycyrrhizin extracted from licorice powder, was determined with good result by HPLC using 2-bromoacetyltriphenylene labeling reagent. The glycyrrhetinic acids were labeled with 2-bromoacetyltriphenylene in acetonitrile using 18-crown-6-ether and KOH as a catalyst. Derivatized glycyrrhetinic acids were separated from the extracted licorice powder on a reversed-phase column (chemopak $C_{18}$) using 100% acetonitrile as a mobile phase and monitored by an UV-detector at 268nm. Linearity of calibration curve was obtained between 5 ng and 20 ng, and the lower limit of detection was 2 ng. The recovery of glycyrrhetinic acid to licorice powder was about 99.3%. This method was sensitive, reliable and useful for, determination of glycyrrhetinic acid.

A rapid and sensitive spectrophotometric method has been developed for quantitation of some phenothiazine derivatives (PTZDS). The method depends on the formation of insoluble yellow complexes between PTZDS and picric acid (PCA) in aqueous layer. These complexes are quantitatively extracted from aqueous phase into chloroform. The binding ratio of PTZDS-PCA complexes were presumed as PTZDS-1 to PCA-1 by means of mole ratio and continuous variation methods. The complexes are stable for more than 24hours in chloroform layer at room temperature. Most of compounds associated with pharmaceutical preparation of PTZDS do not interfere with this method. However, chlorpheniramine and diphenhydramine interfere with the riaethod. The binding state of PTZDS-PCA complexes were presumed by IR and $^1$H-NMR spectra as intermolecular hydrogen bonding.

The weak UV absorbing ephedrine alkaloids such as ephedrine, pseudoephedrine, methylephedrine and norephedrine could be analyzed by charge-transfer spectrophotometric method. The results obtained are summarized as follows: (1) It was possible to determine a weak UV absorbing ephedrine alkaloids using the intense charge-transfer UV bands in chloroform. (2) This method was suitable for the spectrophotometric determination of ephedrine alkaloids in mixed pharmaceutical preparation. (3) Linear relationship was found between absorbance and concentration in the range of 1.0$\times$$10^{-5}M$~5$\times$$10^{-5}M$ of ephedrine ($\varepsilon$= 2.72$\times$$10^{4}LM^{-1}cm^{-1}$ and pseudoephedrine ($\varepsilon$=2.84$\times$$10^{4]LM^{-1}cm^{-1}$), 1.0$\times$$10^{-5}M$~5$\times$$10^{-5}$M of methylephdrine ($\varepsilon$=1.68$\times$$10^{4}LM^{-1}cm^{-1}$) and 1/3$\times$$10^{-4}M$~4/3$\times$$10^{-4}M$ of norephedrine ($\varepsilon$=0.74$\times$$10^{4}LM^{-1}cm^{-1}$. (4) CT- complex of ephedrine, pseudoephedrine and methylephedrine has absorption maxima at 293nm and norephedrine have absorption maximum at 253nm. (5) CT-complexes were formed in a 1:1 ratio between ephedrine alkaloids and iodine in chloroform. (6) By UV, IR, and $^1H$-NMR spectra, it could be inferred that CT-complexes were formed by interaction between the basic nitrogen of ephedrine alkaloids as electron (n) donor and iodine as electron ($\sigma$) acceptor.

8-tert-Butyl-6,7-dihydro-5-methyl-8H-pyrrolo [3,2-e]-s-triazolo [1,5-a] pyrimidine (Bumepidil), one of the s-triazolo [1,5-a] pyrimidine derivatives, has been recently found to be the most promising potential coronary vasodilator and antihypertensive agent. In this report, a new synthetic approach for Bumepidil, via direct N-amination of amino pyrimidine intermediate, was studied and found to be useful method. The novel synthetic method comprise the following steps, acylation of $\gamma$-butyrolactone, condensation with guanidine, direct N-amination, cyclization, chlorination, and finally cyclization using tert-butyl amine.

The effect of mixing ratios of active ingredient on the content uniformity of tablets was studied using caffeine as active ingredient and hydroxypropyl starch as diluent. In the case that caffeine content was not more than 1%, the standard deviation of individual tablet assays was inversely proportional to caffeine content. In the case of more than 1%, the standard deviation was constant independently of caffeine content. In the case that the designed tablet weight was not more than 200mg, the standard deviation of individual tablet assays was inversley proportional to tablet weight designed. In the case of more than 200mg, it was constant and the results for weight variation against the designed weight showed same tendency.
