A Study for Natural Dyeing and Functional Property of Paper and Fabrics with Green Laver Extracts (파래 추출액을 이용한 지사용 원지와 직물의 천연 염색성 및 기능성에 관한 연구)

  • This study explored applicability of natural dyeing and functional property of base paper using paper yarn and fabrics with green laver extracts. As a result of measuring dyeability and functional property of cotton, silk, and a blended fabric of cotton/mulberry, $60^{\circ}C$ and 40 minutes was the optimal dyeing condition for the cotton fabric under an alkali condition; $50^{\circ}C$ and 60 minutes for the silk fabric; $60^{\circ}C$ and 50 minutes for the blended fabric of cotton/mulberry. As a result of measuring the color fastness after dyeing cotton, silk and the blended fabric of cotton/mulberry, fastness to washing, water, rubbing, dry cleaning and light was superior. In terms of functional property, the silk and the blended fabric of cotton/mulberry were superior concerning antimicrobial properties of the fabric dyed with green laver extracts while the cotton fabric showed 90% or over. Regarding deodorization, the blended fabric of cotton/mulberry was the most superior. All the fabrics were harmless to the human body as they had heavy metal content below the standard. Considering the research results comprehensively, the green laver extracts possibly has enough applicability and functional property as a natural dyes. Moreover, it has potential to be developed new eco-friendly fashion materials.

Desmutagenic Effect of Extracts Obtained from Seaweeds (해조류의 항돌연변이 효과)

  • Extracts of six seaweeds obtained from solvents such as water, methanol, hexane and ethyl ether have been tested for desmutagenic activities in the Salmonella typhimurium/microsome systems. Water extracts obtained from tangle, sea-tangle, and gompi were moderately effective on the mutagenicity of Trp-P-2 and aflatoxin $B_1$. Methanol extracts of sea-tangle and gompi were effective against Trp-p-2, and all samples of methanol extracts were very effective from 1.0mg to 2.0mg per plate on the mutagenicity of MeIQ and aflatoxin $B_1$. Hexane extracts of tangle, ses-tangle, gompi and sea-straghorn were very effective on the mutagenicity of Trp-p-2, MeIQ and aflatoxin $B_1$ except laver and green laver. Ethyl ether extracts of gompi and sea-straghorn were most effective from 1.0mg to 2.0mg per plate on the mutagenicity of Trp-P-2. Especially, ethyl ether extracts of all samples were great effective on the mutagenicity of MeIQ and aflatoxin $B_1$.

Angiotensin-I Converting Enzyme Inhibitory Activity of Algae (해조류의 Angiotensin-I 전환효소 저해작용)

  • This study was conducted to investigate the inhibitory activity of water extracts and its enzymatic hydrolysates from algae against angiotensin-I converting enzyme (ACE). The 7 kinds of algae were extracted with water at $50^{\circ}C,\;70^{\circ}C$ and $98^{\circ}C$. ACE inhibitory activities of water extracts were the highest at $70^{\circ}C$, and those of ceylon moss, layer, green layer, sea mustard, seaweed fusiforme sea tangle and sea staghorn were $10.9\%,\;9.3\%,\;8.9\%,\;8.2\%,\;7.5\%,\;7.1\%$ and $7.0\%$, respectively. Layer, green laver sea mustard and ceylon moss of high ACE inhibitory activities among the 7 kinds of water extracts were hydrolyzed by maxazyme and papain during 24hrs. ACE inhibitory activity of enzymatic hydrolysates was higher than that of water extracts, and was the highest in enzymatic hydrolysates of laver among the tested samples. In laver hydrolysates by proteases, the highest ACE inhibitory activity and peptide-nitrogen contents were observed at 8 hours hydrolysis and the hydrolysates by maxazyme showed relatively higher activity than those by papain(31.3 and $27.9\%$, respectively). But peptide-nitrogen contents were greater in papain hydrolysates than in maxazyme.

Desmutagenic Effects of Seaweed and Vegetable Extracts against Mutagenicity of Maillard Reaction Products (Maillard 반응생성물의 돌연변이원성에 대한 해조 및 야채추출물의 억제효과)

  • The desmutagenic effects of seaweed and vegetable extracts were investigated on the mutagenicity of Maillard reaction products (MRP) obtained from equimolar amounts of glucose and amino acid (arginine and lysine${\cdot}$HCl) for Salmonella typhimurium TA 100 without S9 mix. The mutagenicities were inhibited by water-soluble extracts of seaweeds(laver, sea-straghorn, sea-mustard and tangle) and vegetables(ginger, garlic, onion, chinese-pepper, green-onion and cabbage). Cabbage, chinese-pepper, green-onion and sea-straghorn exhibited especially high desmutagenic effects. The desmutagenicities of these extracts(cabbage, green-onion and sea-straghorn) except for sea-straghorn were decreased by heat treatment at $100^{\circ}C$ for 10 min. It is assumed that the desmutagenic effect of seaweed and vegetable extract is due to the reducing power and action of enzyme such as peroxidase and catalase.

Characteristics of marine algae extracts using subcritical water extract method (아임계 추출법을 이용한 해조류 추출물의 특성)

  • This study was performed in order to investigate the functional components of 5 kinds of marine algae. We have collected 5 samples of marine algae, such as the sea mustard (Undaria pinnatifida), sea tangle (Laminaria iaponice), sea weed fusiforme (Hizikia fusiforme), green laver (Entetomotpha), laver (Phophyratenera), which have been harvested in Jeollanam-do. In order to examine the functional effects, 5 kinds of marine algae were extracted with hot water ($80^{\circ}C$, 4 hr), ethanol and methanol (R.T., 4 hr), and subcritical water extract (SWE, 3 MPa, $90^{\circ}C$, $150^{\circ}C$, $210^{\circ}C$). A higher yield of extract was obtained through SWE method (3 MPa, $210^{\circ}C$) in all of the samples obtained. The highest total sugar content was 427.4 mg/g in green laver extracted with SWE (3 MPa, $210^{\circ}C$). The content of the SWE total phenolic compounds was higher than that of the water and solvent (methanol, ethanol) extracts. The anti-oxidative activities of the extracts from 5 kinds of marine algae were examined through the DPPH radical scavenging activity test. The SWE (3 MPa, $150^{\circ}C$ and $210^{\circ}C$) of the marine algae was the highest among all of the extracts. As per the results, the SWE of the marine algae contained more functional components and it had a higher antioxidant activity than those of the other extracts. The $IC_{50}$ value of tyrosinase in seaweed fusiforme and laver were higher than those of the other samples. These results strongly support the possible use of marine algae as functional materials.

Porphyra tenera induces apoptosis of oral cancer cells (구강암 세포주에서 김 추출물에 의한 세포자멸사 유도)

  • Objectives : Laver (Porphyra tenera), a red algae species, is one of the most widely consumed edible seaweed in Korea. Laver contains various substances such as essential amino acid, fiber, minerals and polyphenols that benefit human health. In the present study, we prepared ethanol extracts from commercially processed product of Porphyra tenera, and evaluated the growth inhibitory effect against human oral squamous carcinoma YD-10B cells. Methods : Cell viability was measured by MTT assay. Apoptosis was confirmed by TUNEL assay and flow cytometry with the green fluorescent dye FITC annexin V entering apoptotic cells and the red fluorescent dye PI not entering. The expression of the relevant proteins was detected using Western blot. Results : Ethanol extracts of Porphyra tenera (PTE, $50-200{\mu}g/m{\ell}$) caused a significant decrease of cell viability in a dose dependant manner. The cell death occurred as a result of apoptotic process as determined by TUNEL assay and flow cytometric analysis. In line with this observation, decrease in procaspase proteins and increase in cytosolic cytochrome c were observed in cells treated with PTE. In addition, exposure to PTE decreased the expression levels of Bcl-2, and induced PARP cleavage and AIF translocation from mitochondria to nucleus. Conclusions : In conclusion, PTE exerts anti-cancer effects by inducing apoptosis via caspase activation and AIF nuclear translocation in YD-10B cells. These results provide evidence for the possible therapeutic effect of Porphyra tenera in oral cancer cells.

The Correlation of Antioxidative Effects of 5 Korean Common Edible Seaweeds and Total Polyphenol Content (한국산 5종 해조류의 항산화효과와 총 폴리페놀 함량과의 관련성)

  • Antioxidative activities of 5 common edible seaweeds in Korea, three brown algae (seaweed fusiforme, sea mustard, sea tangle), one green algae (sea lettuce) and one red algae (laver), were examined. The antioxidative activities of ethanol extracts from these seaweeds were examined by measuring of inhibition rates against iron-induced linoleate peroxidation, DPPH (1,1-diphenyl -2-picrylhydrazyl) radical generation and MDA-BSA (malondialdehyde-bovine serum albumin) conjugation. Sea lettuce ethanol extract showed the strongest anti-oxidative activity among them, especially in inhibition against conjugation of lipid peroxide and protein. Second to sea lettuce, laver and sea tangle ethanol extracts showed high DPPH radical scavenging activity and inhibition against MDA-BSA conjugation. However, seaweed fusiforme and sea mustard ethanol extracts did not show antioxidative activities. Sea mustard contained the highest total flavonoids (11.33 mg/g dry wt) and sea lettuce contained the highest total polyphenol (8.97 mg/g dry wt) among these seaweeds. In addition, there was strong positive correlation between the antioxidative activity and total polyphenol content in these seaweeds, suggesting polyphenol compounds may contribute to antioxidative effect of seaweeds. From these data, it is suggested to consume much of seaweeds such as sea lettuce, laver and sea tangle to prevent age-related chronic diseases, and also develope neutraceutical products using polyphenol rich fraction from sea lettuce.

  • The purpose of this study is screening of microfloras involved in hydrolysis of seaweed tenella, seaweed fusiforme and green laver. This is a part of studies on the hydrolysis of seaweed using microorganisms. First, about two hundred microflora samples were obtained from mountain, rice field, dry field, sea, seaside and fish market in the vicinity of Yeosu. Thirty-three microflora samples were screened from the destruction of tissue in sea tangle and sea mustard. It was sufficient that results of the naked eye observation were obtained at eight microflora samples as a feces of bull, a decayed pine tree, a soil of dry field, the mud of the banks in a rice field, the water of a ditch in a rice field, the weed of the banks in a rice field, the water in a rice field and leaved in the air. Above all, extraction rate and contents of reducing sugar in extracts of seaweeds added a decayed pine tree(sample No. 8) and the water of a ditch in a rice field(sample No. 27) were showed high value. And the value of chemical analysis of the sample is much better in comparison with control. Accordingly the hydrolysis of seaweed using microorganisms in the inside of these microflora samples can be possible.

Effect of Natural Plant Components on the Nitrite-scavenging (천연식물성분이 아질산염 소거에 미치는 영향)

  • The purpose of this presents is to investigate, using natural food extracts(Green tea; Camellia sinesis, Du'chung; Eucommia ulmoides Oliver, Eu sung cho; Houttuynia cordate Thunb, Sam back cho; Saurus Chinensis, Baek hwa sa seal oho; Oldenladia diffusa Roxb., Laver; Porphyra tenera, Sea mustard; Undaria pinnatifda and Sea staghorn; Condium fragile) and juices(Sweet pepper; Capsicum annuum var. angulosum, Kale; Brassia oleracea var. acephala, Cucumber; Cucumis sativus, Onion; Allium cepa, Tomato; Lycopericon esculentum, Maesil; Prunus mume, Plum; Prunus salicina and Grape; Vitis spp.), the effect of natural plant components on the nitrite-scavenging under the different levels of pH. From the above mentioned extracts and juices, the content of vitamin C was detected, containing 65.1∼77.1 mg/100g, at the highest level in the Green tea, and followed by vegetables and fruits in order. The nitrite scavenging effect of teas, medicinal plants, and seaweed extracts, in the reaction system under the condition of pH 1.2, were 57.0∼100%, 50.0∼100%, and 18.0∼99.0%, respectively. Antimutagenic and Cytotoxic Effects of Ethanol Extracts from Five Kinds of Seaweeds (다섯 가지 해조류 에탄올 추출물의 항돌연변이 활성 및 암세포 성장억제 효과)

  • The protective effects of ethanol extracts from 5 seaweeds on the mutagenic and cytotoxic damage were evaluated. They were separately extracted using ethanol from dried samples at room temperature, and freeze-dried. The inhibition effects on the mutagenicity in Salmonella assay by Ames test and cancer cell inhibitory effect in HeLa cell, MCF-7 cell and SNU -638 cell by MTT assay were assayed. Seaweed fusiforme, sea tangle and green laver showed strong inhibitory effect against 2-nitrofluorene, sodium azide- or 2-anthramine-induced mutagenicities in Salmonella Typhimurium TA 98 and TA 100 at the level of 2.5 mg ethanol extract per plate. Cancer cell inhibitory effect was shown with all of the seaweed extracts. Green laver, sea mustard, sea tangle and seaweed fusiforme showed strong cytotoxicity against HeLa and MCF-7 cells, with inhibiting by $92\~93\%$ and $89\~92\%$, respectively. These data show that 5 seaweeds tested in this study might be potent functional foods for cancer prevention, and consumption of these seaweeds in adequate amount is recommended.
