The Effect of Notch on Bending Fatigue Strength of Structural Steel (구조용 강의 굽힘 피로강도에 미치는 Notch의 영향)

  • This experimental work was carried out to investigate the fatigue bending strength on various shapes and sizes of notches of the domestically manufactured steel plate. The notch types tested were a circular hole, U-and V-notches. The S-N diagram for different notch shapes were discussed in relation to plan bending strength and stress concentration factor of notches .The results of the experiments are summarized as follows : (1) The difference between stress concentration factor and notch factor was greater as the radium of notch root became smaller, and these values approached to an identical value as the radium of notch root increased. (2) It was shown that the plane bending fatigue limit of bar without notch for the hotrolled steel having the tensile strength of 33.1kg/$mm^2$was 17.0kg/$mm^2$. (3) U-and V-notch had a greater effect of stress concentration factor on the endurance limit, but O-hole showed the same effect only for $\o\pm2mm$. (4) For the same radius of notch root, U-notch showed a lower value of fatigue limit compared to V-notch and O-hole.

Nerve Growth Factor Stimulates Glioblastoma Proliferation through Notch1 Receptor Signaling

  • Objective : Notch receptors are heterodimeric transmembrane proteins that regulate cell fate, such as differentiation, proliferation, and apoptosis. Dysregulated Notch pathway signaling has been observed in glioblastomas, as well as in other human malignancies. Nerve growth factor (NGF) is essential for cell growth and differentiation in the nervous system. Recent reports suggest that NGF stimulates glioblastoma proliferation. However, the relationship between NGF and Notch1 in glioblastomas remains unknown. Therefore, we investigated expression of Notch1 in a glioblastoma cell line (U87-MG), and examined the relationship between NGF and Notch1 signaling. Methods : We evaluated expression of Notch1 in human glioblastomas and normal brain tissues by immunohistochemical staining. The effect of NGF on glioblastoma cell line (U87-MG) was evaluated by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. To evaluate the relationship between NGF and Notch1 signaling, Notch1 and Hes1 expression were evaluated by reverse transcription polymerase chain reaction (RT-PCR) and Western blot analysis, respectively. To confirm the effects of NGF on Notch1 signaling, Notch1 and Hes1 small interfering RNAs (siRNAs) were used. Results : In immunohistochemistry, Notch1 expression was higher in glioblastoma than in normal brain tissue. MTT assay showed that NGF stimulates U87-MG cells in a dose-dependent manner. RT-PCR and Western blot analysis demonstrated that Notch1 and Hes1 expression were increased by NGF in a dose-dependent manner. After transfection with Notch1 and Hes1 siRNAs, there was no significant difference between controls and 100 nM $NGF-{\beta}$, which means that U87-MG cell proliferation was suppressed by Notch1 and Hes1 siRNAs. Conclusion : These results indicate that NGF stimulates glioblastoma cell proliferation via Notch1 signaling through Hes 1.

Averaged strain energy density to assess mixed mode I/III fracture of U-notched GPPS samples

  • In the present contribution, fracture resistance of U-notched GPPS members under mixed mode I/III loading conditions is assessed by using the Averaged Strain Energy Density (ASED) criterion. This criterion has been founded based on the ASED parameter averaged over a well-defined control volume embracing the notch edge. The validation of the theoretical criterion predictions is evaluated through comparing with the results of a series of mixed mode I/III fracture tests conducted on rectangular-shaped GPPS specimens weakened by a single edge U-notch. A recently developed apparatus for mixed mode I/III fracture experiments is employed for measuring the fracture loads of the specimens. The test samples are fabricated with different notch tip radii with the aim of evaluating the influence of this major feature of the U-notched components on the mixed mode I/III fracture behavior. It is shown that the onset of brittle fracture in U-notched GPPS specimens under various combinations of tension and out-of-plane shear can well be predicted by means of the ASED criterion.

A Study on the Corrosion Fatigue Fracture of U-notch Radius (U-노치반경에 따른 부식피로 파괴거동)

  • This study has performed rotary bending fatigue testing that smooth specimen using SM45C materials and notched specimen whose radii were R6, R4 and R2, were processed in 3% NaCl aqueous solutions or in the air. The results are as followed; 1. In the air fatigue limit at 10$^{7}$ cycles remarkably reduced as notch radius goes small. 2. In 3% NaCl aqueous solution fatigue strength at 10$^{6}$ cycles also had large range of reduction as notch radius goes small. 3. Comparing fatigue strength in the air, fatigue strength at 10$^{6}$ cycles in 3% NaCl aqueous solution reduced by 46.2% at smooth specimen, 55.3% at R6 notch radius, 45.8% at R4 and 39.7% at R2 respectively. 4. The reason that fatigue strength reduced in the reduction of notch radius sire was because the surface of notch exposed in corrosion was small and consequently it was less corroded.

THE EFFECT OF CAVITY CONFIGURATION ON MARGINAL LEAKAGE OF CLASS 5 RESTORATION (와동형태가 5급 와동 수복물의 변연누출에 미치는 영향)

  • The purpose of this study was to investigate the effect of cavity configurations on the marginal leakage of class 5 glass ionomer cement and composite resin restorations. Four types of cavities such as saucer shape. notch shape. combined shape(notch shape occlusally and saucer shape gingivally). and U shape were prepared on the buccal and lingual surfaces of 80 extracted premolars(40 cavities for each shape). Occlusal cavity margins were placed at enamel and cervical margins were placed at dentin. 10 cavities of each shape were restored with Ketac Fil as a conventional glass ionomer cement. Fuji II LC improved as a resin modified glass ionomer cement, Z 100 as a hybrid composite resin. and Tetric Flow as a flowable composite resin (40 cavities for each material). After thermocycling, teeth were immersed in 5% basic fuchsin solution for 6 hours and sectioned longitudinally in a buccolingual direction through the center of the restoration. The dye penetrations at the tooth restoration interface were examined by stereomicroscope. The Result were as follows 1. In saucer shape, notch shape and combined shape, composite resin restorations showed lesser leakage than glass ionomer restorations(p<0.05) and in U shape. Tetric Flow showed the least marginal leakage and others were decreased as Z 100. Fuji II LC improved, Ketac Fil in that order. There were statistically significant difference between Tetric Flow and Fuji II LC improved. Ketac Fil and between Z 100 and Ketac Fil(p<0.05). 2. In Ketac Fil restoration group, saucer shape showed the highest marginal leakage and U shape showed the least marginal leakage and others were decreased as notch shape, combined shape in that order. There were statistically significant difference between saucer shape and combined shape, U shape and between notch shape and U shape(p<0.05). 3. In Fuji II LC improved restoration group, U shape showed the least marginal leakage. There were statistically significant difference between U shape and other three shapes(p<0.05). 4. The cavity configuration had no significant effect on marginal leakage of composite resin restorations(p>0.05).

Mixed mode fracture assessment of U-notched graphite Brazilian disk specimens by means of the local energy

  • A fracture criterion based on the strain energy density (SED) over a control volume, which embraces the notch edge, is employed in the present paper to assess the fracture loads of some U-notched Brazilian disk (UNBD) specimens. The specimens are made of commercial graphite and have been tested under pure mode I, pure mode II and mixed mode I/II loading. The results show that the SED criterion allows to successfully assess the fracture loads of graphite specimens for different notch tip radii and various mode mixity conditions with discrepancies that fall inside the scatter band of ${\pm}20%$.

Propranolol Inhibits the Proliferation of Human Glioblastoma Cell Lines through Notch1 and Hes1 Signaling System

  • Objective : The anti-tumor effect of the beta-adrenergic receptor antagonist propranolol in breast cancer is well known; however, its activity in glioblastoma is not well-evaluated. The Notch-Hes pathway is known to regulate cell differentiation, proliferation, and apoptosis. We investigated the effect of propranolol to human glioblastoma cell lines, and the role of Notch and Hes signaling in this process. Methods : We performed immunohistochemical staining on 31 surgically resected primary human glioblastoma tissues. We also used glioblastoma cell lines of U87-MG, LN229, and neuroblastoma cell line of SH-SY5Y in this study. The effect of propranolol and isoproterenol on cell proliferation was evaluated using the MTT assay (absorbance 570 nm). The impact of propranolol on gene expression (Notch and Hes) was evaluated using real-time polymerase chain reaction (RT-PCR, whereas protein levels of Notch1 and Hes1 were measured using Western blotting (WB), simultaneously. Small interfering RNA (siRNA) was used to suppress the Notch gene to investigate its role in the proliferation of glioblastoma. Results : Propranolol and isoproterenol caused a dose-dependent decrease in cell proliferation (MTT assay). RT-PCR showed an increase in Notch1 and Hes1 expression by propranolol, whereas WB demonstrated increase in Notch1 protein, but a decrease in Hes1 by propranolol. The proliferation of U87-MG and LN229 was not significantly suppressed after transfection with Notch siRNA. Conclusion : These results demonstrated that propranolol suppressed the proliferation of glioblastoma cell lines and neuroblastoma cell line, and Hes1 was more closely involved than Notch1 was in glioblastoma proliferation.

Fatigue Behavior of the U-Notched Specimens of Structural Carbin Steel under the two Level Stressing(I) (2段階 應力下에서 U - 노치 를 갖는 構造용 炭素鋼材 의 피勞擧動 I)

  • In order to analyze and investigate in the fatigue behavior of the notched specimens under the two level stressing, the U-notched specimens of structural carbon steel which is generally used is prepared. The obtained results are summarized as follows; (1) The fatigue limit of the U-notched specimens is lower than that of the unnotched. The fatifue notch factor (.betha.) of the U-notched specimens is 1.44 for mild steel and 1.52 for harden steel. The notch sensitivity (q) is 0.68 for mild steel and o.8 for harden steel. That is, these facts show that harden steel is more sensitive to the notch than mild steel. (2) The fatigue life time of the U-notched specimens under the overstressing is shorter than under the constant stressing, and the degree of fatigue life time decrease is different to each stress level; the degree for the high stress level is more than for the low stress level. (3) The fatigue life time of the U-notched specimes under the understressing is longer than under the constant stressing, and the degree of fatigue life time increase for the low stress level is more than for the high stress level.

Stress Concentration Factor and Stress Intensity Factor with U-notch and Crack in the Beam (U-노치 및 균열을 갖는 보의 응력집중계수 및 응력확대계수)

  • The stress concentration factors and stress intensity factors for a simple beam and a cantilever are analyzed by using finite element method and phtoelasticity. Using the analyzed results, the estimated graphs on stress concentration factors and stress intensity factors are obtained. To analyze stress concentration factors of notch, the dimensionless notch length H(height of specimen)/h=1.1~2 and dimensionless gap space r(radius at the notch tip)/h=0.1~0.5 are used. where h=H-c and c is the notch length. As the notch gap length increases and the gap decreases, the stress concentration factors increase. Stress concentration factors of a simple beam are greater than those of a cantilever beam. However, actually, the maximum stress values under a load, a notch length and a gap occur more greatly in the cantilever beam than in the simple beam. To analyze stress intensity factors, the normalized crack length a(crack length)/H=0.2~0.5 is used. As the length of the crack increases, the normalized stress intensity factors increase. The stress intensity factors under a constant load and a crack length occur more greatly in the cantilever beam than in the simple beam.
